Abstract
Reivindicación 1: Un diacuerpo optimizado en secuencia capaz del enlace específico a un epítopo de CD123 y a un epítopo de CD3, caracterizado porque el diacuerpo comprende una primera cadena de polipéptido y una segunda cadena de polipéptido, covalentemente enlazadas entre sí, en donde: A) la primera cadena de polipéptido comprende, en la dirección N-terminal a C-terminal: i) un Dominio 1, que comprende (1) un sub-Dominio (1A), que comprende un Dominio VL de un anticuerpo monoclonal capaz de enlazar a CD3 (VLCD₃) (SEQ ID Nº 21); y (2) un sub-Dominio (1B), que comprende un Dominio VH de un anticuerpo monoclonal capaz de enlazar a CD123 (VHCD₁₂₃) (SEQ ID Nº 26), en donde los sub-Dominios 1A y 1B se separan entre si por un enlazador de péptido (SEQ ID Nº 29); ii) un Dominio 2, en donde el Dominio 2 es un Dominio con Espiral E (SEQ ID Nº 34) o un Dominio con espiral K (SEQ ID Nº 35), en donde el Dominio 2 se separa del Dominio 1 por un enlazador de péptido (SEQ ID Nº 30); y B) la segunda cadena de polipéptido comprende, en la dirección N-terminal a C-terminal: i) un Dominio 1, que comprende (1) un sub-Dominio (1A), que comprende un Dominio VL de un anticuerpo monoclonal capaz de enlazar a CD123 (VLCD₁₂₃) (SEQ ID Nº 25); y (2) un sub-Dominio (1B), que comprende un Dominio VH de un anticuerpo monoclonal capaz de enlazar a CD3 (VHCD₃) (SEQ ID Nº 22), en donde los sub-Dominios 1A y 1B se separan entre sí por un enlazador de péptido (SEQ ID Nº 29); ii) un Dominio 2, en donde el Dominio 2 es un Dominio con espiral K (SEQ ID Nº 35) o un Dominio con Espiral E (SEQ ID Nº 34), en donde el Dominio 2 se separa del Dominio 1 por un enlazador de péptido (SEQ ID Nº 30); y en donde el Dominio 2 de la primera y la segunda cadenas de polipéptido no son ambos Dominios con Espiral E o ambos Dominios con espiral K y en donde: (a) el Dominio VL de la primera cadena de polipéptido y el Dominio VH de la segunda cadena de polipéptido forman un Dominio de Enlace de Antígeno capaz de enlazar específicamente a un epítopo de CD3; y (b) el Dominio VL de la segunda cadena de polipéptido y el Dominio VH de la primera cadena de polipéptido forman un Dominio de enlace a antígeno capaz de enlazar específicamente a un epítopo de CD123. Reivindicación 15: Una composición farmacéutica, caracterizada porque comprende el diacuerpo de cualquiera de las reivindicaciones 1 a 8 y un portador fisiológicamente aceptable. Reivindicación 16: Uso de la composición farmacéutica de conformidad con la reivindicación 15, caracterizado porque es para el tratamiento en una enfermedad o condición asociada con o representada por la expresión de CD123. Reivindicación 20: El uso de conformidad con la reivindicación 19, caracterizado porque la condición inflamatoria se selecciona del grupo que consiste de: lupus autoinmune (SLE), alergia y asma.
