Claims (31)
1. Соединение, включающее модифицированный олигонуклеотид, состоящий из 12-30 связанных нуклеозидов и содержащий нуклеотидную последовательность, включающую часть по меньшей мере из 8 смежных нуклеотидных оснований, комплементарных части последовательности нуклеотидных оснований 1275-1318 SEQ ID NO:1, имеющей такую же длину, где указанная нуклеотидная последовательность модифицированного олигонуклеотида по меньшей мере на 90% комплементарна последовательности SEQ ID NO:1.
2. Соединение по п.1, где указанная нуклеотидная последовательность модифицированного олигонуклеотида по меньшей мере на 95%, по меньшей мере на 96%, по меньшей мере на 97%, по меньшей мере на 98%, по меньшей мере на 99% или по меньшей мере на 100% комплементарна последовательности SEQ ID NO:1.
3. Соединение по п.1 или 2, имеющее:
(i) нуклеотидную последовательность, содержащую по меньшей мере 8, по меньшей мере 10, по меньшей мере 12, по меньшей мере 14, по меньшей мере 16, по меньшей мере 18 или 20 смежных нуклеотидных оснований последовательности SEQ ID NO:223, 217, 51, 52, 53, 114, 115, 213, 214, 215, 216, 218, 219, 220, 221, 222, 224, 225, 226, 227, 228, 229, 230, 231, 232, 242, 243, 245, 246, 247, 248, 250, 251, 252, 253, 254, 255, 256, 257, 258, 259, 260, 261, 262, 263, 264, 265, 266, 267, 268 или 269; или
(ii) нуклеотидную последовательность, содержащую по меньшей мере 8, по меньшей мере 10, по меньшей мере 12, по меньшей мере 14, по меньшей мере 16, по меньшей мере 18 или 20 смежных нуклеотидных оснований последовательности SEQ ID NO:223, 217, 51, 52, 53, 114, 115, 213, 214, 215, 216, 218, 219, 220, 221, 222, 224, 225, 226, 227, 228, 229, 230, 231 или 232.
4. Соединение по п.1, имеющее нуклеотидную последовательность, содержащую по меньшей мере 8, по меньшей мере 10, по меньшей мере 12, по меньшей мере 14, по меньшей мере 16, по меньшей мере 18 или 20 смежных нуклеотидных оснований последовательности SEQ ID NO:223 или 217.
5. Соединение по п.1, где указанный модифицированный олигонуклеотид состоит из 15-30, 18-24, 19-22 или 20 смежных нуклеозидов.
6. Соединение по п.1, состоящее из одноцепочечного модифицированного олигонуклеотида.
7. Соединение по п.1, где по меньшей мере одной межнуклеозидной связью является модифицированная межнуклеозидная связь.
8. Соединение по п.7, где каждая межнуклеозидная связь представляет собой фосфортиоатную межнуклеозидную связь.
9. Соединение по п.1, где по меньшей мере один нуклеозид содержит модифицированный сахар.
10. Соединение по п.9, где по меньшей мере одним модифицированным сахаром является бициклический сахар.
11. Соединение по п.10, где каждый из по меньшей мере одного бициклического сахара содержит 4'-(CH2)n-O-2'-мостик, где n равно 1 или 2.
12. Соединение по п.10, где каждый из по меньшей мере одного бициклического сахара содержит 4'-CH(CH3)-O-2'-мостик.
13. Соединение по п.9, где по меньшей мере один модифицированный сахар содержит 2'-O-метоксиэтильную группу.
14. Соединение по п.1, содержащее по меньшей мере один модифицированный тетрагидропираном нуклеозид, в котором фуранозное кольцо заменено тетрагидропирановым кольцом.
16. Соединение по п.1, где по меньшей мере один нуклеозид включает модифицированное нуклеотидное основание.
17. Соединение по п.16, где указанное модифицированное нуклеотидное основание представляет собой 5-метилцитозин.
18. Соединение по п.1, где указанный модифицированный олигонуклеотид включает:
гэп-сегмент, состоящий из связанных дезоксинуклеозидов;
5'-сегмент-крыло, состоящий из связанных нуклеозидов;
3'-сегмент-крыло, состоящий из связанных нуклеозидов,
где указанный гэп-сегмент расположен непосредственно между 5'-сегментом-крылом и 3'-сегментом-крылом и является смежным с этими сегментами, и где каждый нуклеозид каждого сегмента-крыла содержит модифицированный сахар.
19. Соединение по п.18, где указанный модифицированный олигонуклеотид включает:
гэп-сегмент, состоящий из десяти связанных дезоксинуклеозидов;
5'-сегмент-крыло, состоящий из пяти связанных нуклеозидов;
3'-сегмент-крыло, состоящий из пяти связанных нуклеозидов, где указанный гэп-сегмент расположен непосредственно между 5'-сегментом-крылом и 3'-сегментом-крылом и является смежным с этими сегментами, и где каждый нуклеозид каждого сегмента-крыла содержит 2'-O-метоксиэтилированный сахар; и каждая межнуклеозидная связь представляет собой фосфортиоатную связь.
20. Соединение по п.18 или 19, где каждый цитозин представляет собой 5-метилцитозин.
21. Соединение по п.1, где указанный модифицированный олигонуклеотид состоит из 20 связанных нуклеозидов и имеет нуклеотидную последовательность, содержащую 20 смежных нуклеотидных оснований SEQ ID NO:223, и где указанный модифицированный олигонуклеотид включает:
гэп-сегмент, состоящий из десяти связанных дезоксинуклеозидов;
5'-сегмент-крыло, состоящий из пяти связанных нуклеозидов;
3'-сегмент-крыло, состоящий из пяти связанных нуклеозидов, где указанный гэп-сегмент расположен непосредственно между 5'-сегментом-крылом и 3'-сегментом-крылом и является смежным с этими сегментами, и где каждый нуклеозид каждого сегмента-крыла содержит 2'-O-метоксиэтилированный сахар; каждой межнуклеозидной связью является фосфортиоатная связь, и каждый цитозин представляет собой 5-метилцитозин.
22. Соединение по п.1, где указанный модифицированный олигонуклеотид состоит из 20 связанных нуклеозидов и имеет нуклеотидную последовательность, содержащую 20 смежных нуклеотидных оснований SEQ ID NO:217, и где указанный модифицированный олигонуклеотид включает:
гэп-сегмент, состоящий из десяти связанных дезоксинуклеозидов;
5'-сегмент-крыло, состоящий из пяти связанных нуклеозидов;
3'-сегмент-крыло, состоящий из пяти связанных нуклеозидов, где указанный гэп-сегмент расположен непосредственно между 5'-сегментом-крылом и 3'-сегментом-крылом и является смежным с этими сегментами, и где каждый нуклеозид каждого сегмента-крыла содержит 2'-O-метоксиэтилированный сахар; каждой межнуклеозидной связью является фосфортиоатная связь, и каждый цитозин представляет собой 5-метилцитозин.
23. Соединение по любому из пп.1-22 для применения в терапии.
24. Соединение по п.23 для применения в лечении или предупреждении тромбоэмболического осложнения у животного.
25. Соединение по п.24, где указанное тромбоэмболическое осложнение выбрано из тромбоза глубокой вены, эмболии легких, инфаркта миокарда и инсульта.
26. Соединение по п.23 для применения в лечении нарушения свертывания крови у животного.
27. Композиция, включающая соединение по любому из пп.1-22 или его соль и фармацевтически приемлемый носитель или разбавитель.
28. Способ лечения или предупреждения тромбоэмболического осложнения у животного, включающий введение животному, нуждающемуся в этом, соединения по любому из пп.1-22 или композиции по п.27.
29. Способ по п.28, где указанное тромбоэмболическое осложнение выбрано из тромбоза глубокой вены, эмболии легких, инфаркта миокарда и инсульта.
30. Способ лечения нарушения свертывания крови у животного, включающий введение животному, нуждающемуся в этом, соединения по любому из пп.1-22 или композиции по п.27.
31. Способ ингибирования экспрессии фактора 11 у животного, включающий введение указанному животному терапевтически эффективного количества соединения по любому из пп.1-22 или композиции по п.27.
