Abstract
1. Мышь, содержащая в своей зародышевой линии:(a) гуманизированный вариабельный локус тяжелой цепи иммуноглобулина, содержащий по меньшей мере один нереаранжированный сегмент V человека, по меньшей мере один нереаранжированный сегмент D человека и по меньшей мере один нереаранжированный сегмент J человека, функционально связанный с геном константной области тяжелой цепи;(b) гуманизированный вариабельный локус легкой цепи иммуноглобулина, содержащий не более чем одну или не более чем две реаранжированные последовательности V/J легкой цепи человека, функционально связанные с геном константной области легкой цепи; и,(c) эктопическую последовательность нуклеиновой кислоты, кодирующую белок ADAM6 мыши, или его ортолог, или гомолог, или функциональный фрагмент.2. Мышь по п. 1, у которой последовательность нуклеиновой кислоты, кодирующая белок ADAM6 мыши, или его ортолог, или гомолог, или функциональный фрагмент, находится на локусе, отличном от вариабельного локуса тяжелой цепи иммуноглобулина.3. Мышь по п. 1, у которой ген константной области тяжелой цепи представляет собой ген мыши.4. Мышь по п. 1, у которой ген константной области легкой цепи представляет собой ген мыши.5. Мышь, содержащая гуманизированный вариабельный локус тяжелой цепи иммуноглобулина и гуманизированный вариабельный локус легкой цепи иммуноглобулина, причем мышь экспрессирует единственную легкую цепь и при этом мышь содержит эктопическую последовательность нуклеиновой кислоты, кодирующую ген ADAM6, являющийся функциональным у самца мыши.6. Claims (20)
1. Мышь, содержащая в своей зародышевой линии:
(a) гуманизированный вариабельный локус тяжелой цепи иммуноглобулина, содержащий по меньшей мере один нереаранжированный сегмент V человека, по меньшей мере один нереаранжированный сегмент D человека и по меньшей мере один нереаранжированный сегмент J человека, функционально связанный с геном константной области тяжелой цепи;
(b) гуманизированный вариабельный локус легкой цепи иммуноглобулина, содержащий не более чем одну или не более чем две реаранжированные последовательности V/J легкой цепи человека, функционально связанные с геном константной области легкой цепи; и,
(c) эктопическую последовательность нуклеиновой кислоты, кодирующую белок ADAM6 мыши, или его ортолог, или гомолог, или функциональный фрагмент.
2. Мышь по п. 1, у которой последовательность нуклеиновой кислоты, кодирующая белок ADAM6 мыши, или его ортолог, или гомолог, или функциональный фрагмент, находится на локусе, отличном от вариабельного локуса тяжелой цепи иммуноглобулина.
3. Мышь по п. 1, у которой ген константной области тяжелой цепи представляет собой ген мыши.
4. Мышь по п. 1, у которой ген константной области легкой цепи представляет собой ген мыши.
5. Мышь, содержащая гуманизированный вариабельный локус тяжелой цепи иммуноглобулина и гуманизированный вариабельный локус легкой цепи иммуноглобулина, причем мышь экспрессирует единственную легкую цепь и при этом мышь содержит эктопическую последовательность нуклеиновой кислоты, кодирующую ген ADAM6, являющийся функциональным у самца мыши.
6. Мышь по п. 5, у которой гуманизированный локус тяжелой цепи иммуноглобулина функционально связан с эндогенным геном константной области тяжелой цепи мыши.
7. Мышь по п. 5, у которой единственная легкая цепь кодируется вариабельным генным локусом легкой цепи иммуноглобулина, который кодирует не более чем один генный сегмент V легкой цепи и генный сегмент J легкой цепи, функционально связанные с геном константной области легкой цепи.
8. Мышь по п. 5, у которой единственная легкая цепь происходит из вариабельного генного локуса легкой цепи иммуноглобулина в зародышевой линии мыши, который кодирует не более чем один реаранжированный генный сегмент V/J легкой цепи в зародышевой линии, функционально связанный с геном константной области легкой цепи.
9. Мышь по п. 5, у которой эктопическая последовательность нуклеиновой кислоты кодирует белок ADAM6 мыши или кодирует фрагмент белка ADAM6 мыши, который является функциональным у самца мыши.
10. Мышь по п. 5, у которой эктопическая последовательность нуклеиновой кислоты находится в положении, которое не находится в пределах эндогенного локуса тяжелой цепи иммуноглобулина.
11. Генетически модифицированная мышь, которая экспрессирует множество различных тяжелых цепей IgG, причем каждая содержит вариабельный домен человека, причем каждая из множества различных тяжелых цепей IgG ассоциирована с последовательностью легкой цепи, кодирующей вариабельный домен легкой цепи иммуноглобулина человека, который происходит из одного генного сегмента V иммуноглобулина человека, причем мышь содержит эктопическую последовательность нуклеиновой кислоты, кодирующую белок ADAM6, который является функциональным у самца мыши.
12. Мышь по п. 11, причем мышь экспрессирует белок ADAM6 или его фрагмент, который является функциональным у самца мыши, и причем белок ADAM6 или его фрагмент экспрессируется из эктопической последовательности нуклеиновой кислоты.
13. Клетка мыши, содержащая:
гуманизированный вариабельный генный локус тяжелой цепи иммуноглобулина, функционально связанный с геном константной области тяжелой цепи;
гуманизированный локус легкой цепи иммуноглобулина, который содержит не более чем один или не более чем два генных сегмента V легкой цепи, функционально связанных с геном константной области легкой цепи; и,
эктопическую последовательность нуклеиновой кислоты, кодирующую белок ADAM6 или его фрагмент, причем белок ADAM6 или его фрагмент является функциональным у самца мыши.
14. Клетка мыши по п. 13, в которой эктопическая последовательность нуклеиновой кислоты кодирует белок ADAM6 мыши.
15. Клетка мыши по п. 13, у которой ген константной области тяжелой цепи представляет собой не относящийся к человеку ген константной области тяжелой цепи.
16. Клетка мыши по п. 13, у которой ген константной области легкой цепи представляет собой не относящийся к человеку ген константной области легкой цепи.
17. Клетка мыши по п. 13, у которой не более чем один или не более чем два генных сегмента V легкой цепи присутствуют в реаранжированном генном сегменте V/J.
18. В-клетка мыши, которая экспрессирует химерную тяжелую цепь иммуноглобулина, содержащую вариабельный домен тяжелой цепи иммуноглобулина, происходящий из генного сегмента V тяжелой цепи человека; и легкую цепь, происходящую из
(a) реаранжированной последовательности Vκ1-39/J человека,
(b) реаранжированной последовательности Vκ3-20/J человека или
(c) их комбинации;
причем вариабельный домен тяжелой цепи слит с константной областью и вариабельный домен легкой цепи слит с константной областью и при этом В-клетка содержит эктопическую последовательность нуклеиновой кислоты ADAM6.
19. В-клетка мыши по п. 18, в которой константный домен, слитый с вариабельным доменом тяжелой цепи, содержит константную область мыши.
20. В-клетка мыши по п. 18, в которой константная область, слитая с вариабельным доменом легкой цепи, выбрана из константной области мыши и константной области человека.
