Description
ただし、好ましいペプチドは、少なくとも下線を施したトリペプチド配列を有し、且つ0から4又は5個までの周囲のアミノ酸をさらに有し、より好ましくは、プロリン残基(P)が、N末端から第2番目の残基であるぺプチドである。これらのペプチドを、そのまま、又はいずれかの混合物、特に、加水分解物の一部として使用することができる。
Gly−Pro−p−ニトロアニリド(Sigma G−0513)をDPP−IVの基質として使用して、385nmにおける吸収の増加を測定することによって、DPP−IV活性を決定することができる。DPP−IV活性の減少が、阻害の測定値である。
加水分解物1:
1000グラムのカゼイン(DMV International、オランダ)を、7.2リットルの水に溶解した。pH及び温度を、それぞれ8.8及び55℃に調節した後、増量して正確に8リットルとした。次いで、40グラムのAlkalase(Novo Nordisk、デンマーク)、4グラムのProtease N(Amano、日本)、及び4グラムのFlavourzyme(Novo Nordisk)を、混合液に添加した。18時間インキュベートした後、90℃、10分間の熱による不活性化によって反応を停止させた。50℃まで冷却した後、混合液を3kDaのカットオフを有するろ過膜でろ過し、透過水を乾燥させた。
1400グラムのカゼイン(DMV International、オランダ)を、7.2リットルの水に溶解した。pH及び温度を、それぞれ8.3及び50℃に調節した後、増量して正確に8リットルとした。1.5グラムのTrypsin PTN 6.0(Novo Nordisk、デンマーク)及び200mlのエタノールを添加した。次いで、混合液を、一様に撹拌しながら、2時間インキュベートした。9グラムのCorolase LAP F(Rohm、デンマーク)及び100mlのエタノールを添加した。さらに21時間インキュベートした後、90℃、10分間の熱による不活性化によって反応を停止させた。次いで、混合液を50℃まで冷却した後、乾燥させた。
1000グラムの乳清タンパク質の濃縮物(WPC80;Arla、デンマーク)を、7.2リットルの水に溶解した。pH及び温度を、それぞれ8.3及び60℃に調節した後、増量して正確に8リットルとした。12グラムのProtease N(Amano、日本)及び250mlのエタノールを添加した。次いで、混合液を、一様に撹拌しながら、18時間インキュベートした。90℃において、10分間熱を加えることによって反応を停止させた。得られた混合液を50℃まで冷却した後、3kDaのフィルターでろ過し、乾燥させた。
乾燥させた加水分解物2を、水中に再溶解させ、3kDaのろ過膜でナノろ過を実施した。濃縮水及び透過水を用いて、DPP−IV阻害効力及び質量バランス(濃縮水及び透過水対全加水分解物)を試験した。DPP−IVの阻害は、上記の実施例1で記載した方法に従って実施され、質量バランスには、オルトフタルジアルデヒド法を使用した。簡単に説明すると、この方法は、タンパク質及びペプチド中に存在する第一級アミン基のような、第一級アミン基のオルトフタルジアルデヒドによる修飾に基づいている。この方法のために、試料を、0.1Mホウ砂、1%(w/v)ドデシル硫酸ナトリウム、pH9.3、に溶解した。混合した後、オルトフタルジアルデヒド試薬を添加し、次いで、室温において、10分間インキュベートした。吸光度を340nmで測定した。質量バランスは、濃縮水及び透過水の吸光度を、加水分解物中のタンパク質の全量と関係付けることによって決定した。表1は、出発加水分解物と比較して、透過水のDPP−IV阻害活性が、2倍に増加することを示している。
実施例2にしたがって調製した種々のタンパク質の加水分解物を、DPP−IV阻害活性に関して、さらに検討した。Van Elswijkら(上記)で議論されているように、高分解能スクリーニング(HRS)装置を使用して、加水分解されたタンパク質の試料をDPP−IV阻害活性に関して分析した。
加水分解物4、5及び6:
100グラムのカゼインナトリウムを、1800gの水に撹拌しながら溶解した。pHを7に調節した。次いで、水を加え、最終量を2000mlとした。表3に特定するプロテアーゼ(ブロメライン、Scintillase CS150L(Genencor)、パパイン)1グラムを水に溶解し、添加した後、混合液を、内部温度50℃において、24時間インキュベートした。次いで、95℃において5分間加熱することによって、酵素反応を停止し、次いで、混合液を凍結乾燥した。凍結乾燥生成物を水に溶解し、3000rpmで遠心分離した。上澄を、1kDaのカットオフ値を有するUFフィルターで透析ろ過した。出発材料、乾燥させた遠心分離ペレット、乾燥させたUF濃縮水及び乾燥させたUF透過水のDPP−IV阻害効果を決定した(表3、図1、2、3)。
Claims (13)
- MPLW(配列番号16)、LPQYL(配列番号47)、PLLQ(配列番号44)、VPYPQ(配列番号49)、VPLGTQ(配列番号54)及びLPVPQK(配列番号55)から選択される単離ペプチドを含む、DPP−IVが仲介する状態の予防及び/又は治療のための医薬品又は食品。
- 有効量の請求項1に記載のペプチドの1種以上を含有するタンパク質加水分解物を含む、DPP−IVが仲介する状態の予防及び/又は治療のための医薬品又は食品であって、前記加水分解物は、最大で600μg/mlのDPP−IV阻害活性についてのIC 50 を有する、上記医薬品又は食品。
- 前記DPP−IVが仲介する状態が、肥満、2型糖尿病及び免疫学的障害からなる群から選択される、請求項1又は2に記載の医薬品又は食品。
- 肥満の予防及び/又は治療のための、請求項3に記載の医薬品又は食品。
- 2型糖尿病の予防及び/又は治療のための、請求項3に記載の医薬品又は食品。
- 免疫学的障害の予防及び/又は治療のための、請求項3に記載の医薬品又は食品。
- 関節リウマチ、多発性硬化症及びグレーブス病からなる群から選択される自己免疫疾患の予防及び/又は治療のための、請求項6に記載の医薬品又は食品。
- KVLP(配列番号45)及びLPL(配列番号2)から選択されるさらなるペプチドの1種以上を含む、請求項1〜7のいずれか一項に記載の医薬品又は食品。
- MPLW(配列番号16)、LPQYL(配列番号47)、PLLQ(配列番号44)、VPYPQ(配列番号49)、VPLGTQ(配列番号54)及びLPVPQK(配列番号55)から選択される、DPP−IV阻害活性を有するペプチド。
- 請求項9に記載のペプチドの1種以上を含有するタンパク質加水分解物であって、前記加水分解物は、最大で600μg/mlのDPP−IV阻害活性についてのIC 50 を有する、上記タンパク質加水分解物。
- 前記タンパク質加水分解物が、乳タンパク質加水分解物である、請求項10に記載のタンパク質加水分解物。
- 前記タンパク質加水分解物が、カゼイン加水分解物である、請求項11に記載のタンパク質加水分解物。
- 3〜7個のアミノ酸からなる長さを有するペプチドを、20重量%から80重量%の間で含有する、請求項10〜12のいずれか一項に記載のタンパク質加水分解物。
