Abstract

A method of preparing 9-acyl-11,12-secoprostaglandins of the formula: **(See formula)** where A is ethylene, trimethylene, α-methylethylene, β-methylethylene, α, α-dimethylethylene, β, β-dimethylethylene or oxymethylene; Z is methylene, ethylene, trimethylene, tetramethylene, vinyl, or ethylene; R2 and R4 are independently selected from the group consisting of hydrogen and methyl; R5 is selected from the group consisting of hydrogen, linear or branched lower alkyl of 1 to 4 carbon atoms (eg methyl, ethyl, propyl, isopropyl, butyl, tert-butyl), vinyl and 2,2,2-trifluoroethyl; in addition, when R5 is lower alkyl and R2 is methyl, they can be bonded together (with hydrogen abstraction) to form a 6- to 9-membered carbocyclic ring; likewise, when R5 is lower alkyl and R2 is hydrogen, R5 may be attached to the carbon atom bearing the R2 and OR3 groups to form a 5- to 8-membered carbocyclic ring; whose procedure is to react a compound of formula: **(See formula)** where R10 is lower alkyl of 1 to 5 carbon atoms, with one equivalent of a strong base and then with one of compounds A or B, then react the resulting product with one equivalent of a strong base followed by reaction with the another of compounds A or B, where compound A responds to the formula **(See formula)** where X is halogen; A is ethylene, trimethylene, α-methylene, β-methylene, α, α-dimethylethylene, β, β-dimethylethylene or oxymethylene; and R10 is lower alkyl of 1 to 5 carbon atoms; and where compound B responds to the formula: **(See formula)** where X is halogen, preferably chlorine or bromine; Z'is methylene, ethylene, trimethylene or tetramethylene and R4 and R5 are as defined above; recovering the intermediate product thus produced and then subjecting the product to basic hydrolysis, decarboxylation and elimination when R10 is tert-butyl or subjecting the product to basic hydrolysis and decarboxylation when R10 is primary or secondary alkyl to form the product **(See formula)** where A, R4, R5 and Z'are as defined above.
