Abstract

La invención se refiere a una combinación de agentes de reprogramación de moléculas pequeñas para inducir la generación de células madre totipotentes, un método para preparar las células madre totipotentes inducidas, y las células madre totipotentes inducidas generadas a partir de los mismos. Reivindicación 1: Una composición que comprende: (a) un activador de la vía de señalización de RA; y (b) uno o más de un inhibidor de GSK-3, un inhibidor de la vía de señalización de IKK, un inhibidor de HDAC, un inhibidor de histona metiltransferasa, un inhibidor de quinasa Src, un activador de AMPc, y un inhibidor del metabolismo celular. Reivindicación 2: Un equipo que comprende: (a) un activador de la vía de señalización de RA; y (b) uno o más de un inhibidor de GSK-3, un inhibidor de la vía de señalización de IKK, un inhibidor de HDAC, un inhibidor de histona metiltransferasa, un inhibidor de quinasa Src, un activador de AMPc, y un inhibidor del metabolismo celular. Reivindicación 12: Un medio de cultivo que comprende la composición de acuerdo con cualquiera de las reivindicaciones anteriores. Reivindicación 15: Un método para fabricar células madre totipotentes inducidas, que comprende cultivar células madre pluripotentes en el medio de cultivo de acuerdo con cualquiera de las reivindicaciones anteriores, para fabricar así las células madre totipotentes inducidas. Reivindicación 21: Un cultivo que comprende el medio de cultivo de acuerdo con cualquiera de las reivindicaciones anteriores y células madre pluripotentes. Reivindicación 23: Un cultivo que comprende el medio de cultivo de acuerdo con cualquiera de las reivindicaciones anteriores y células madre totipotentes. Reivindicación 25: Una célula madre totipotente inducida, caracterizada por uno o más de los siguientes: (a) aumento de la transcripción de uno o más marcadores de transcripción totipotentes seleccionados del grupo que consiste en MERVL, Zscan4c, Zscan4d, Zscan4f, Zfp352, Tcstv1, Tcstv3, Teme92, Gm6763; (b) transcripción reducida de uno o más marcadores de transcripción pluripotentes seleccionados del grupo que consiste en POU5f1, ZFP42, NANOG, KLF4, ESRRB; y (c) la capacidad de diferenciarse en tipos celulares extraembrionarios; preferentemente, las células madre totipotentes inducidas pueden fabricarse mediante el método de acuerdo con cualquiera de las reivindicaciones anteriores. Reivindicación 27: Un organismo derivado de la célula madre totipotente inducida de acuerdo con cualquiera de las reivindicaciones anteriores, preferentemente donde el organismo es un animal roedor o un animal mamífero. Reivindicación 28: Un organoide producido a partir de la célula madre totipotente inducida de acuerdo con cualquiera de las reivindicaciones anteriores. Reivindicación 29: Un tejido producido a partir de la célula madre totipotente inducida de acuerdo con cualquiera de las reivindicaciones anteriores, donde preferentemente el tejido es sangre. Reivindicación 30: Una célula diferenciada, diferenciada de la célula madre totipotente inducida de acuerdo con cualquiera de las reivindicaciones anteriores, preferentemente donde la célula diferenciada es una célula sanguínea o una célula inmunitaria, tal como una célula T o una célula NK.
