Abstract
La presente se refiere a moléculas de polinucleótidos que comprenden secuencias de nucleótidos que codifican un producto génico AveC, moléculas de polinucleótidos que pueden utilizarse para alterar la relación o cantidad de avermectina de la clase 2:1 producidas en cultivos de fermentación de S. avermitilis. La presente se refiere, además, a vectores, células huésped y cepas mutantes de S. avermitilis en las que el gen aveC ha sido inactivado o mutado con el fin de cambiar la relación o cantidad de avermectinas de la clase 2:1 producidas. Reivindicación 1: Molécula de polinucleótido caracterizada porque comprende una secuencia de nucleótidos que, por lo demás, es la misma que el alelo aveC Streptomyces avermitilis, la secuencia del plásmido pSE186 (ATCC 209604) codificadora del producto génico aveC de S. avermitilis o la secuencia de nucleótidos del marco de lectura abierta (ORF) AveC de S. avermitilis según se presenta en la Figura 1 (SEC ID N°: 1) o una variante degenerada de la misma, pero secuencia de nucleótidos que comprende, adicionalmente, mutaciones que codifican una combinación de sustituciones de aminoácidos en los residuos aminoácidos correspondientes a las posiciones de aminoácidos de SEC ID N°: 2, de modo que células de la cepa ATCC 53692 de S. avermitilis, en las que el alelo aveC de tipo salvaje ha sido inactivado y que expresan la molécula de polinucleótidos que comprende la secuencia de nucleótidos mutada son capaces de producir una relación de clase 2:1 de avermectinas que está reducida en comparación con la relación producida por células de la cepa ATCC 53692 de S. avermitilis que, en su lugar, expresan únicamente el alilo aveC de tipo salvaje, en donde, cuando las avermectinas de clase 2:1 son avermectinas ciclohexilo B2: ciclohexilo B1, la relación de avermectinas de clase 2:1 es 0,35:1 o menor.
