Abstract
N-arilamidas de ácidos sulfonilamino-carboxílicos sustituidas con azufre, de la fórmula (1), en la cual A1 representa un radical bivalente de la serie fenileno, naftileno y heteroarileno, todos los cuales pueden estar sustituidos por uno o más sustituyentes, iguales o diferentes, seleccionados de la serie consistente en: halógeno, alquilo C1-5, fenilo, tolilo, CF3, NO2, OH, -O-C1-5-alquilo, -O-O-C2-4-alquil-O-C1-3-alquilo, C1-2-alquilendioxi, NH2, -NH-C1-3-alquilo, -N(C1-3-alquilo)2, -NH-CHO, -NH-CO-C1-5-alquilo, -CN, -CO-NH2, -CO-C1-3-alquilo, -CO-N(C1-3-alquilo)2, -CO-OH, -CO-O-C1-5-alquilo, heterociclilo, CHO, -CO-(C1-5-alquilo)2, -S(O)n-C1-4-alquilo, -S(O)n-fenilo y -S(O)n-tolilo; el anillo A2, que abarca los átomos de carbono que llevan los grupos C(=X)-NH- y NH-SO2R2, representa un anillo benzol, un anillo naftalina, un carbociclo de 3 a 7 miembros, saturado o parcialmente no saturado, un heterociclo monocíclico de 5 a 7 miembros, saturado, parcialmente no saturado, o aromático, que contiene uno o más heteroátomos de anillo seleccionados de la serie consistente en N, O y S, o un heterociclo bicíclico de 8 a 10 miembros, saturado, parcialmente no saturado, o aromático, que lleva uno o varios heteroátomos de anillo seleccionados de la serie consistente en N, O y S; R1 representa arilo, heterociclilo o C1-18-alquilo, que puede estar sustituido por uno o más radicales R4, iguales o diferentes, o cuando el número n en el grupo R1-S(O)n- vale 2, también representa NR5R6, o cuando el número n en el grupo R1-S(O)n- vale 0, también representa -CN; R2 representa arilo, heterociclilo, NR5R6 o alquilo C1-10 que puede estar sustituido por uno o más radicales R4, iguales o diferentes; R3 representa uno o más radicales, iguales o diferentes, seleccionados de entre la siguiente serie: hidrógeno, halógeno, CF3, OH, -O-C1-7-alquilo, -O-C2-4-alquil-O-C1-7-alquilo, -O-arilo, C1-2-alquilendioxi, NO2, -CN, NR7R8, -CO-NR7R8, -CO-OH, -CO-C1-5-alquilo, heterociclilo, S(O)n-C1-5-alquilo y C1-5-alquilo, que puede estar sustituido por uno o más radicales R4, iguales o diferentes; R4 representa flúor, OH, -O-C1-10-alquilo, -O-C1-4-alquil-O-C1-7-alquilo, -O-arilo, -CN, NR7R8, -CO-NH2, -CO-NH-C1-3-alquilo, -CO-N(C1-3-alquilo)2, -CO-OH, -CO-O-C1-5-alquilo, heterociclilo u oxo; R5 representa hidrógeno, C1-10-alquilo, que puede estar sustituido por uno o más radicales R4, iguales o diferentes y/o por arilo, heterociclilo, -CO-NR7R8, -CO-arilo ó -CO-C1-10-alquilo, pudiendo el radical alquilo estar sustituido por uno o más radicales R4, iguales o diferentes; R6, independientemente de R5, tiene uno de los significados indicados para R5, o R5 y R6 conjuntamente con el átomo de nitrógeno al cual están vinculados, forman un anillo de 5 a 8 miembros, saturado o parcialmente no saturado, que adicionalmente del átomo de nitrógeno que lleva los grupos R5 y R6 también puede contener uno o más heteroátomos de anillo de la serie N, O y S, y que puede estar sustituido por uno o varios sustituyentes, iguales o diferentes, tomados de la serie consistente en: flúor, C1-5-alquilo, hidroxi-C1-3-alquilo, -C1-3-alquil-O-C1-4-alquilo, arilo, CF3, OH, -O-C1-7-alquilo, -O-arilo, -O-C2-4-alquil-C1-7-alquilo, C2-3-alquilendioxi, NR7R8, -CN, -CO-NH2, -CO-NH-C1-3-alquilo, -CO-N(C1-3-alquilo)2, -CO-OH, -CO-O-C1-5-alquilo, CHO, CO-C1-5-alquilo, S(O)n-C1-4-alquilo, S(O)n-NH2, S(O)n-NH-C1-3-alquilo, S(O)n-N(C1-3-alquilo)2, oxo, (CH2)m-NH2, -(CH2)m-NH-C1-4-alquilo y -(CH2)m-N(C1-4-alquilo)2, pudiendo en el sustituyente (CH2)m-N(C1-4-alquilo)2 ambos grupos alquilo estar vinculados mediante un enlace simple y entonces formar conjuntamente con el átomo de nitrógeno que los lleva un anillo de 5 a 7 miembros, que además del átomo de nitrógeno y de los átomos de carbono puede contener también un átomo de oxígeno, un átomo de azufre o un grupo NR5 a título de miembro del anillo; R7 representa hidrógeno o C1-7-alquilo, puede estar sustituido por uno o varios sustituyentes, iguales o diferentes, de la serie: OH, -O-C1-5-alquilo, NH2, -NH-(C1-4-alquilo)2 y N(C1-4-alquilo)2, pudiendo en el sustituyente N(C1-4-alquilo)2 ambos grupos alquilo estar vinculados mediante un enlace simple y entonces formar conjuntamente con el átomo de nitrógeno que los lleva un anillo de 5 a 7 miembros, que además del átomo de nitrógeno y de los átomos de carbono puede contener también un átomo de oxígeno, un átomo de azufre o un grupo NR5 a título de miembro del anillo; R8, independientemente de R7, tiene uno de los significados de R7 o representa -CO-C1-4-alquilo; arilo representa fenilo, naftilo o heteroarilo todos los cuales pueden estar sustituidos por uno o varios sustituyentes, iguales o diferentes, seleccionados de entre el grupo consistente en: halógeno, C1-5-alquilo, fenilo, tolilo, CF3, -O-CF3, NO2, OH, -O-C1-5-alquilo, -O-C2-4-alquil-C1-3-alquilo, C1-2-alquilendioxi, NH2, -NH-C1-3-alquilo, N-(C1-3-alquilo)2, NH-CHO, -NH-CO-C1-5-alquilo, -CN, CO-NH2, -CO-NH-C1-3-alquilo, CO-N(C1-3-alquilo)2, -CO-OH, -CO-O-C1-5-alquilo, heterociclilo, CHO, CO-C1-5-alquilo, S(O)n-C1-4-alquilo, S(O)n-fenilo y S(O)n-tolilo; heterociclilo representa el radical de un heterociclo monocíclico o policíclico, de 5 a 11 miembros, saturado o parcialmente no saturado, que contiene uno o más heteroátomos de anillo de la serie, N, O y S, y que puede estar sustituido por uno o más sustituyentes, iguales o diferentes, de la serie consistente en: flúor, C1-5-alquilo, OH, O-C1-5-alquilo, -O-C2-4-alquil-O-C1-3-alquilo, NH2, -NH-C1-3-alquilo, -N(C1-3-alquilo)2, -CN, CO-NH2, CO-NH-C1-3-alquilo, CO-N(C1-3-alquilo)2, -CO-OH y CO-O-C1-5-alquilo; n representa 0, 1 ó 2; m representa 2, 3 ó 4; X representa O ó NH, o X representa un átomo de nitrógeno, que por intermedio de un enlace simple está vinculado a un átomo de carbono de anillo en el grupo A1, que está directamente próximo al átomo de carbono portador del grupo -NH-C(=X)- en A1, de manera tal que los grupos -NH-C(=X)- juntamente con los átomos de carbono que los lleva en el grupo A1 forma un anillo imidazol anelado; en todas sus formas estereoisoméricas y mezclas de las mismas en todas las proporciones, como también sus sales fisiológicamente aceptables, excluyéndose el compuesto de la fórmula (1) en la cual simultáneamente el anillo A2, que abarca los átomos de carbono que llevan los grupos C(=X)-NH- y NH-SO2R2, representa un anillo benzol, que en las posiciones 3 y 5 está sustituido con cloro, R2 representa metilo, X representa oxígeno y R1-S(O)n-A1 representa un radical 5-cloro-1-(4-clorofenilmercapto)-fenilo, procedimiento para la preparación de los compuestos de la fórmula (1), y preparado farmacéutico que contiene los compuestos de la fórmula (1). Estos compuestos son valiosas sustancias activas como medicamentos para la terapia y profilaxis de enfermedades, por ejemplo de enfermedades cardio-circulatorias tales como la elevada tensión arterial, angina pectoral, insuficiencia cardíaca, trombosis o aterosclerosis. Los compuestos de la fórmula (1) tienen la capacidad de modular la producción, propia del cuerpo, del monofosfato de guanosina cíclico (cGMP, cyclic guanosinmonophosphat) y en términos generales son adecuados para la terapia y profilaxis de los estados patológicos relacionados con una alteración del metabolismo del cGMP.
