Abstract

On a identifié SULTn, gène codant pour une nouvelle sulfotransférase du système nerveux, en recherchant dans une base de données de marqueurs de séquence exprimée destinés à la sulfotransférase spécifiquement exprimée dans le cerveau. SULTn est un polypeptide de 33.085 Mr qui structurellement ressemble à des sulfotransférases cystosoliques (identique à 34-39 %, par exemple), avec des motifs structurels de signature de site de liaison de cosubstrat de 3'-phosphoadénosine 5'-sulfotransférase dans le domaine catalytique du polypeptide. Sultn se lie à l'adénosine 3',5'-diphosphate-agarose, analogue du cosubstrat, et des expériences de la concurrence ont démontré la spécificité de cette liaison. A la différence de toutes les sulfotransférases cystosoliques connues, qui sont hautement exprimées dans le foie humain, des transcrits de SULTn de 2,4 kb ont été détectés par analyse de transfert Northern uniquement dans le cerveau humain, mais pas dans le foie, ni dans le coeur, ni dans le placenta, ni dans les poumons, ni dans le muscle squelettique, ni dans le rein, ni dans le pancréas. L'hybridation in situ d'embryons de rat a révélé une expression de développement précoce du gène SULTn dans le système nerveux central, périphérique et entérique. Sultn est largement exprimé dans le système nerveux de l'adulte, y compris dans la couche de cellules de Purkinje du cortex cérébelleux. Le schéma d'hybridation in situ dans le système nerveux était cohérent avec la coloration des neurones, et était différent de l'hybridation de protéine acide fibrillaire gliale, marqueur d'astrocyte. Le gène SULTn humain possède un total de 7 exons codants sur le chromosome 22 (22q13) qui couvre plus de 47kb d'ADN génomique. L'identification d'une nouvelle sulfotransférase cystosolique exprimée exclusivement dans les tissus nerveux révèle ainsi l'existence d'une sulfonation spécifique du système nerveux.
