九、發明說明: 【發明所屬之技術領域】 本發明係關於一種即使於培養開始時的培養基中存在乳 酸的條件下亦可生產γ-胺基丁酸(以下稱為「GABA」)之屬 於乳酸桿菌屬(lactobacillus)之乳酸菌,進而係關於使用該 乳酸桿菌之含γ-胺基丁酸培養物之製造方法。 【先前技術】 GABΑ係因具有降低血壓作用、利尿作用、精神安定作 用等生理效果,而目前受到關注之胺基酸,但含GABA食 品僅限於一部分蔬菜及茶、米等,而且GABA之含量亦較 少,故現提出有若干利用GABA醱酵性乳酸菌之GABA製 造方法(例如,參照曰本專利文獻1〜3)。至於該等生產 GABA之乳酸菌,已知有希氏乳酸桿菌(Lactobacillus hilgardii)、乳酸乳球菌乳脂亞種(Lactococcus lactis subsp. Cremoris)、乳酸乳球菌乳酸亞種(Lactococcus lactis subsp. Lactis)、路黃腸球菌(Enterococcus casseliflavus)、短乳桿 菌(Lactobacillus brevis)、胚芽乳酸桿菌(Lactobacillus plantarum)、嗜熱鏈球菌(Streptococcus thermophilus)等, 其具有藉由脫碳酸反應而由L-麩胺酸或L-麩胺酸鹽生產 GABA之能力。 但是,培養該等乳酸菌,因必須將乳清粉(whey powder)、脱脂乳、酪蛋白、蛋白脒等高價成分作為培養 基,故自成本方面考慮生產性欠佳(例如,參照日本專利 文獻1或4)。又,以酸或者以酶、麴分解作為廉價材料之 118872.doc 1379003 醬油麴及麵質(gluten)麴,且利用鲢醯胺酸酶將所溶出的 麩醯胺轉化成麩胺酸,並以上述麩胺酸作為培養基之情形 肖’於中和時所產生的食鹽及酶分解時作為環保二案:添 加的食鹽,會抑制乳酸菌生產GABA。關於耐鹽性生產 GABA之乳酸g,亦有若干報告,但⑽靖酵之最佳食 鹽濃度為12.5〜18%,屬於濃度極高之鹽(例如,參照曰本 專利文獻5 )。若考慮推行意圖減鹽的飲食生活以避免患上 # 生活習慣病,則含高鹽之食品或調味料並不理想。又, GABA具有所期待之降低血壓作用,但若將其與高濃度食 鹽組合,則會失去其本來的意義。 根據以上所述,於利用乳酸菌生產GABA時,較理想的 是以低鹽狀態下分解使用如麴及麩質之價廉材料,但於低 鹽下會出現野生菌造成污染之問題。污染係於培養初始階 段產生,其結果導致產生乙醇或者使{)11降低,使可生產 GABA之乳酸菌的生長受到抑制,而無法獲得充分量之目 • 標GABA。防止該等於低鹽下由野生菌所導致之污染的方 法,有利用通常用作食品防腐劑之乙醇、醋酸、乳酸的方 法,然而乙醇、醋酸雖然可抑制污染菌,但亦會抑制生產 GABA之乳酸菌。另一方面,乳酸菌因自身會產生乳酸, 故對乳酸的耐受性較其他細菌更高。 因此’人們考慮將乳酸用作防腐劑,但若於食鹽存在下 開始培養時添加乳酸,則生產Gaba之乳酸菌無法生長、 或者所生長者無法進行GAB A生產,導致出現新問題。 【專利文獻1】曰本專利特開2000-210075號公報 118872.doc 1379003 【專利文獻2】曰本專利特開2004-215529號公報 【專利文獻3】曰本專利特開2〇〇5_1〇2559號公報 【專利文獻4】曰本專利第3426157號公報 【專利文獻5】曰本專利第27〇4493號公報 【發明内容】 [發明所欲解決之問題] 本發明之課題在於提供,於培養開始時的培養基中共存 φ 有乳酸與食鹽的條件下可生產GABA之乳酸菌、以及含 GAB A培養物之製造方法。 [解決澤題之技術手段] 本發明者們為解決上述課題而反覆進行努力研究,結果 發現:自醬曝中分離出即使於自培養開始時共存有乳酸與 食鹽的狀態下亦可生產〇舰之乳酸菌,並可藉由利用該 乳酸菌解決上述問題,從而完成本發明。 即’本發明係關於以下(丨)〜(4)。 • ⑴一種屬於乳酸桿菌屬之乳酸菌,其可於培養開始時的 培養基中共存有乳酸及食鹽的條件下,生產γ_胺基丁酸。 ⑺-種屬於乳酸桿㈣之乳酸g,其可於培養開始時的 培養基中共存有1〜4%之乳酸、及8〜12%之食鹽的條件下, 生產γ-胺基丁酸。 (3) 如上述⑴或⑺項之乳酸菌’其中乳酸菌係凝乳酸桿 菌。 (4) 一種含γ_胺基丁酸培養物之製造方法,其特徵在於: 將上述⑴至(3)巾任-項之乳酸菌接種於含有l•麵胺酸及/ I18872.doc 或其鹽類之培養基中,進行培養。 [發明之效果] 根據本發明,可提供一種使用廉價材料,不抑制野性菌 之醱酵而穩定生產GABA之方法。 【實施方式】 以下,詳細說明本發明。 本發明之乳酸菌,係屬於乳酸桿菌屬之菌株,例如可使 用自醬醪中分離出之乳酸菌。本發明之乳酸菌,於培養開 始時的培養基中共存有乳酸與食鹽的條件下,具有生產 GABA之能力,可於共存有1〜4%之乳酸及8〜12%之食鹽的 條件下生產GABA ^該乳酸菌,較好的是凝乳酸桿菌,例 如凝乳酸桿菌KG34株及凝乳酸桿菌KG43株等。 凝乳酸桿菌KG34株,係於2006年2月7日移交獨立行政 法人製品評價技術基盤機構(NITE)專利微生物寄存中心 (NPMD)(郵政編碼292-0818日本國千葉縣木更津市上總鐮 足2-5-8)進行國内寄存(受託碼號NITE P-177)(BCRC 9103 49),且於2006年9月1曰移交至基於布達佩斯條約進 行國際寄存管理(受託號碼NITE BP-177)(BCRC 910349)。 又,凝乳酸桿菌KG43株,係於2007年1月31日移交至獨 立行政法人製品評價技術基盤機構(NITE)曰本專利微生物 寄存中心(NPMD)(郵政編碼292-0818曰本國千葉県木更津 市上總鐮足2-5-8),基於布達佩斯條約進行國際寄存(收件 號碼 NITE ABP-309)(BCRC 910350)。 將乳酸菌自醬醪中分離,係採用如下之方法。即,將25 g 118872.doc 醬醪進行鐵胃處理,將所獲得液汁適量加入到置於杜罕氏 醱酵管(Durham tube)内之 MRS-SOYTONE 培養基(5.5% MRS肉湯、0.5% bactosoyt0ne、〇_5%麩胺酸鈉、8%食鹽、 5%食品添加用乳酸pH 5.1)中,於3〇°C下進行靜置培養。 以MRS-SO YTONE培養基將確認於杜罕氏醱酵管中有氣體 產生的培養液加以適當稀釋,進而將其塗佈於MRS-SOYTONE複月旨培養基上’置於AnaeroPack-kenki(三菱氣 體化學股份有限公司)中以30°C培養7天。將所獲得單株菌 再次接種到内置杜罕氏醱酵管的MRS-SOYTONE培養基 中,選取確認有氣體產生之菌。 以上述方法分離〖034、〖043、81、〖031、^:064、尺0-18-4、KG4-14-1株之7種株菌。於該等7株菌·中,調查 KG34、KG43兩株之細菌學性質,將其結果示於表1。作為 比較對照,亦記載有作為凝乳酸桿菌標準菌之凝乳酸桿菌 DSMZ20253的資料。已知,標準菌凝乳酸桿菌DSMZ20253 不具有麩胺酸脫碳酸能力,因而無法生產GABA。 118872.doc 10· 1379003 [表i]
