Claims (16)
1. Биспецифический связывающий белок, связывающий антиген А и антиген В, содержащий три полипептидных цепи, при этом первая полипептидная цепь содержит в направлении от амино конца к карбоксильному концу или (1) VLa-CL-VHB-CH1-Fc; или (2) VHB-CH1-VLA-CL-Fc, где VLA представляет собой вариабельный домен легкой цепи антитела первого исходного антитела, которое связывает антиген A, CL представляет собой константный домен легкой цепи антитела, VHB представляет собой вариабельный домен тяжелой цепи антитела из второго исходного антитела, которое связывает антиген В, и СН1 представляет собой первый константный домен тяжелой цепи антитела;
указанная вторая полипептидная цепь содержит, в направлении от амино конца к карбоксильному концу. VHA-СН1, где VHA представляет собой вариабельный домен тяжелой цепи первого исходного антитела, которое связывает антиген А, и СН1 представляет собой первый константный домен тяжелой цепи антитела; и указанная третья полипептидная цепь содержит, в направлении от амино конца к карбоксильному концу, VLB-CL, где VLB представляет собой вариабельный домен легкой цепи второго исходного антитела, которое связывает антиген В и CL представляет собой константный домен легкой цепи антитела;
причем указанный биспецифический связывающий белок связывает антигены А и В и содержит вариабельные домены VLA, VHA, VLB, и VHB, содержащие последовательности аминокислот для каждой из трех последовательностей CDR (CDR-1, CDR-2, CDR-3), выбранных из следующих последовательностей:
2. Биспецифический связывающий белок по п. 1, отличающийся тем, что указанный биспецифический связывающий белок связывает антигены А и В и содержит домены VLA, VHA, VLB и VHB, содержащие последовательность аминокислот для VLA, VHA, VLB и VHB, выбранные из группы:
3. Биспецифический связывающий белок по п. 2, отличающийся тем, что указанный связывающий белок связывает антигены А и В и содержит первую, вторую и третью полипептидную цепи, имеющие последовательности аминокислот, выбранные из группы:
4. Связывающий белок с одной или более консервативными заменами аминокислот в связывающем белке по любому из пп. 1-3, обладающий активностью, эквивалентной соответствующему связывающему белку по пп. 1-3 без консервативных замен аминокислот.
5. Фармацевтическая композиция, содержащая связывающий белок по любому из пп. 1-3 и один или более фармацевтически приемлемый носитель.
6. Способ лечения или предотвращения состояния у нуждающегося в этом субъекта, включающий введение указанному субъекту эффективного количества фармацевтической композиции по п. 5.
7. Способ по п. 6, отличающийся тем, что указанное состояние представляет собой воспалительное заболевание, аутоиммунное заболевание, нейродегенеративное заболевание, рак или повреждение спинного мозга.
