Claims (1)
- 물과 아세톤의 혼합물에 용해된 공지된 3'-아미노-3'-에피-다우노루비신을 살리실알데히드와 반응시켜 상응하는 3'-에피-N-살리실리덴 유도체를 수득하고, 이를 무수 피리딘의 존재하에 무수 디클로로메탄 속에서 트리플루오로메탄설폰산 무수물과 반응시켜 3'-에피-N-살리실리덴-4'-O-트리플루오로메탄 설포네이트를 수득하고, 메탄올에 용해된 상기 화합물을 실온에서 p-톨루엔설폰산을 이용하여 살리실리덴 보호 그룹을 산 가수분해하고 이어서 트리플루오로메탄 설포네이트 이탈 그룹을 제거하여 중간체 3'-데아미노-4'-데옥시-3'-하이드록시-4'-에피-3',4'-에피미노-다우노루비신을 수득하고, 이를 트리플루오로 아세트산 무수물로 처리하여 상응하는 N-트리플루오로아세틸 유도체를 수득하고, 생성된 N-트리플루오로아세틸 유도체를 촉매량의 황산과 반응시켜 4'-데옥시-4'-에피-N-트리플루오로아세틸-3'-데아미노-3'-하이드록시-다우노루비신을 수득하고, 이를 0℃의 온도에서 바람직하게는 0.1N 수성 수산화나트륨을 사용하여 온순한 알칼리 가수분해하여 목적하는 화합물 3'-데아미노-4'-데옥시-3'-하이드록시-4'-에피-4'-아미노-다우노루비신(IIa : R1=H)을 유리 염기로서 수득하고, 이를 메탄올성 염화수소로 처리하여 의의 염산염으로서 분리시키고, 경우에 따라, 브롬 용액으로 처리하여 이의 14-브로모-유도체로 치환시킨 다음, 나트륨 포르메이트의 수용액으로 가수분해하여 3'-데아미노-4'-데옥시-3'-하이드록시-4'-에피-4'-아미노 독소루비신(IIb : R1=OH)을 유리 염기로서 수득하고, 최종적으로 상응하는 염산염으로서 분리시킴을 특징으로 하여, R1이 수소원자 또는 하이드록실 그룹을 나타내는 일반식(II)(a : R1=H, b : R1=OH)의 안트라사이클린 글리코사이드를 제조하는 방법.
