Claims (1)
- 무수 피리딘을 함유하는 메틸렌 디클로라이드에 용해된 구조식(Ⅲ)의 4'-에피 -N-트리플루오로아세틸-다우노루비신을 0℃에서 트리플루오로메틸설폰산 무수물과 반응시켜 구조식(Ⅳ)의 4'-에피-4'-트리플루오로메틸 설포닐옥시-N-트리플루오로아세틸-다오노루비신을 생성시키고; 생성된 구조식(Ⅳ)의 화합물을 테트라부틸암모늄 요오다이드와 반응시켜 구조식(Ⅴ)의 4'-데옥시-4'-요오도-N-트리플루오로아세틸 -다우노루비신을 생성시킨 후, 계속해서 무수 톨루엔중, 환류온도에서 α,α'-아조비스-이소부티로니트릴 존재하에 트리부틸틴 하이드라이드를 사용하여 환원적 탈할로겐화시켜 구조식(Ⅵ)의 4'-데옥시-N-트리플루오로 아세틸-다우노루비신을 생성시키고; 생성된 중간체(Ⅵ)를 10℃에서 0.1N 수산화나트륨 수용액으로 온화하게 알카리 가수분해하여 유리 글리코사이드 염기 4'-데옥시-다우노루비신을 수득하고, 계속해서 이를 메탄올성 염산으로 처리하여 구조식(Ⅶ)의 상응하는 염산염으로 분리한 후, 생성물을 메탄올-디옥산 혼합물에 용해시켜 에틸 오르토포름에이트 존재하에서, 클로로포름에 용해된 브롬으로 처리하여 상응하는 14-브로모 유도체를 수득하고, 최종적으로 실온에서 나트륨 포름에이트 수용액으로 가수분해하여 4'-데옥시-독소루비신(Ⅰ)을 상응하는 염산염으로 수득하고 분리함을 특징으로 하여, 구조식(Ⅰ)의 4'-데옥시-독소루비신 염산염을 제조하는 방법.
