본 발명은 식물성장 조절제로 유용한 신규의 다음 일반식(I)의 히다토인의 제조방법에 관한 것이다.

Q는 CH또는 N이고 R1, R2및 R3는 같거나 다를수 있으며, 수소, 직쇄 포는 측쇄 C1-4알킬, 또는 직쇄 또는 측쇄 C3-4알케닐이며 R4및 R5는 같거나 다를수 있으며, C1-4-O, C1-4-S 또는 임의로 치환된 페닐-0 또는 페닐-S 그룹으로 치환될 수 있는 C1-4알킬, C2-4알케닐, 저급알킬 그룹으로 치환될 수 있는 C3-6사이클로알킬, 또는 임의로 치환된 페닐 또는 벤질을 나타대고, W는 산소이거나, Q가 CH일때 황일수도 있으며, X 및 Y는 같거나 다를수 있으며, 할로겐, C1-4알킬, C1-4알콕시 또는 트리클루오로메틸이고, X는 또한 수소도 나타낸다.

c) Q가 CH인 화합물을 제조하기 위해서는, 일반식(V)의 아미노 화합물을 탈아미노화 시켜 E 및/또는 E' 그룹을 제거하거나,

d) Q가 N인 일반식(I)의 화합물을 제조하기 위해서는, 일반식(VI)의 스피로히단토인을 일반식(Ⅶ)의 피리딘과 염기성 화합물(예 : 탄산칼륨, 탄산나트륨, 수산화칼륨, 수산화나트륨 또는 수산화칼륨) 첨가하에 약 0℃ 내지 80℃에서 반응시켜 제조한다.

발명에 따른 화합물은 수많은 잡초 특히 단자엽성 잡초에 대해 그들의 강력한 활성을 나타낸다. 이 화합물은 바람직하게는 발아전에 적용한다. 본 발명의 활성물질은 내성이 있으므로 수많은 작물(예 : 콩, 옥수수, 쌀, 목화, 보리, 사탕무우, 감자, 토마토, 또는 양파)에 사용할 수 있다.
사용량은 사용된 물질, 잡초종류 및 외부조건에 따라 0.1 내지 10kg/ha, 더욱 특히 0.3 내지 3kg/ha으로 다양하다.
일반식(I)화합물을 공지의 제초제와 혼합하여 사용하는 것이 바람직하다. 그 실례로는 우레아 유도체(예 : 클로로톨루론), 트리아진 유도체(예 : 아트라진, 시메진), 디니트로아닐린 유도체(예 : 트리폴루라린), 클로로 아세트아니리드 유도체(예 : 알라크를), 티오 카바에이트(예:티오벤카브) 및 디페닐에테르(예 : 아시플루오펜)와의 혼합물이 있다.
상용으로 일반식(I)의 활성물질은 통상방법으로 과립제, 살포제, 현탁분말 또는 농축제 또는 수분산성과립제 같은 통상적인 제제로 만들 수 있다.
이들 제제는 활성물질을 희석제 또는 증량제(예 : 용매 및/또는 고체 담체), 가능하게는 표면 활성제(유화제 또는 분산제) 및/또는 안정제 및/또는 소포제의 첨가와 가능하게는 다른 첨가물과 혼합 또는 분쇄하여 제조한다.
바람직한 용매는 물이며 ; 적당한 고체 담체는, 예를들어, 미네랄 가루(예, 카올린, 점토, 탈크, 분필, 석영, 고도로 분산된 실릭산, 산와 알루미늄 및 실리케이트)가 있다. 과립제에 적당한 담체는 한편 깨어지고 파편으로된 암석(예, 방해석, 대리석 또는 경석)이 있고 또한 유기물질(예 : 톱밥, 야자껍질 및 옥수수속)의 과립이 있다.
적당한 유화제에는 폴리옥시에틸렌 지방산 에스테르, 폴리옥시에틸렌 지방알콜에테르(예 : 알킬아린-폴리글리콜 에테르), 알킬설포네이트, 알킬설페이트, 아릴설포네이트 및 단백질 가수분해물 같은 비이온성 및 음이온성 화합물이 포함된다.
분산제로는 목재 가공처리시 얻어지는 페설파이트액 또는 메틸 셀룰로즈가 있고 소포제로는 측쇄 고급알콜이 있다.
농축된 제제는 일반적으로 활성성분을 0.1 내지 95%, 바람직하게는 0.5 내지 90중량% 함유하며 이를 물로 희석하여 소정의 농도로 만들어 분산액으로 임의로 제조할 수 있다.
조성물은 제제에 따라 붓거나, 분무하거나 뿌리거나 살포한다.

[실시예 1]
사이클로헥산-5'-스피로-3'-(3,5-디클로로페닐)-히단토인
1-아미노-사이클로헥산 카복실산을 HC1/EtOH으로 에스테르화하여 제조된 에틸 1-아미노-사이클로헥산 카복실레이트 171g(1몰)을 에테르 500ml에 용해시킨다. 에테르 500ml에 3,5-디클로로페닐 이소시아네이트 188.5g(1몰)을 녹인 용액을 얼음으로 냉각하면서 15분내에 가한다. 다 가한후, 생성혼합물을 주위온도에서 30분간 교반시키고 그리고 나서 흡인 여과한다. 여과기 상의 잔류물을 500ml의 에탄올에 현탁시키고 트리에틸아민 10.1g(0.1몰)을 가한후 3시간 동안 환류시킨다. 그리고 나서 빙수 2리터에 붓고 생성된 침전물은 흡인 여과시킨다. 물로 충분히 세척하고 순환공기 건조기에서 건조하여 표제 화합물 약 280g(이론치의 89%)을 수득한다.
융점 : 210 내지 211℃(에탄올로부터).
[실시예 2]
시스-2-메틸 사이클로헥산-5'-스피로-3'-(3,5-디클로로페닐)-2'-티오-히단토인
나트륨 11.5g(0.5몰)을 에탄올 500ml에 용해시키고 시스-1-아미노-2-메틸-사이클로헥산 카르복실산 79g(0.5몰)을 용액에 가한다. 이어서 미세하게 분말화된 3,5-디클로로페닐 이소타오-시아네이트를 30분내에 생성된 현탁액에 가한다. 맑은 용액을 주위온도에서 3시간동안 교반시키고 3시간 환류시킨다. 빙수 2ℓ에 붓고, 형성된 침전물을 흡인 여과시킨후 물로 충분히 세척하고, 건조시켜 표제화합물 140g(이론치의 82%)을 수득한다.
융점 : 237 내지 239℃(에탄올로부터).
입체화학적구조(시스 또는 트랜스)측정은 칸-인골드-프레로그계(Cahn-Ingold-Rrelog System)에 따라 수행한다.

시스-4-메틸사이클로헥산-5'-스피로-3'-(3,5-디브로모페닐)히단토인
a)시스-4-메틸사이클로헥산-5'-스피로-3'-(2-니트로게닐)히단토인
시스-4-메틸사이클로헥산-5'-스피로-히단토인 18.2g(0.1몰), 2-니트로-클로로벤젠 18g(0.1몰) 및 칼륨 카보네이트 41.4g(0.3몰)을 디에틸포름아미드 100 ml에 용해시키거나 현탁시키고 이것을 약 120℃에서 5시간 교반시킨다. 혼합물을 얼음에 붓고, 형성된 침전물은 흡인 여과시킨 후 물로 충분히 세척하고 아세토니트릴로 결정화한다.
수득량 : 15.5g(이론치의 51%)
융점 : 227℃ 내지 229℃.
b) 시스-4-메틸사이클로헥산-5'-스피로-3'-(2-아미노페닐)-히단토인
시스-4-메틸사이클로헥산-5'-스피로-3'-(2-니트로페닐)-히단토인 15.2g (0.05몰)을 메틸 알콜 200ml 및 진한 염산 5ml에 용해시키고 촉매(Pb/c,10%) 4g을 부가한 후에 혼합물을 40℃ 최대 온도에서 5바 압력하에 수소화시킨다.
H2가 모두 흡수된 후 촉매를 흡인 여과에 의해 제거시키고 모액을 증발에 의해 농축시킨다. 잔사를 물에 흡수시키고 중탄산염으로 중화시킨다. 얻어진 고체 생성물을 흡인 여과시키고 물로 세척한 후 이소프로판물로 결정화한다.
수득량 : 11.5g(이론치의 84%)
융점 : 255℃ 내지 256℃
c)시스-4-메틸사이클로헥산-5'-스피로-3'-(2-아미노-3,5-디브로모페닐)-히단토인
시스-4-베틸-사이클로헥산-5'-스피로-3'-(2-아미노-페닐)히단토인 27.3 3g(0.1몰)을 빙초산 200ml에 용해시킨다. 빙초산 50ml에 녹인 브롬 32g(0.2몰) 용액을 주위온도에서 30분내에 적가한다. 생성된 혼합물을 약 30분간 교반시키고, 물 11로 희석한 후 침전물을 흡인 여과시키고 물로 세척하고, 아세토니트릴로부터 결정화한다.
수득량 : 35.4g(이론치의 82%)
융점 : 264 내지 266℃.
d)시스-4-메틸사이클로헥산-5'-스피로-3'-(3,5-디브로모페닐)-히단토인
시스-4-메틸-5'-스피로-3'-(3,5-디브로모페닐)-히단토인 21.6g(0.05몰)을 에탄올 300ml 및 진한 황산 1ml에 용해시키고, 혼합물은 끓는점까지 가열한다. 그리고 아질산 나트륨 3.8g(0.055몰)을 약 20분내로 배치내에서 적가하고 생성된 혼합물은 교반하면서 1시간동안 환류시킨다. 냉각한 후에 빙수11로 희석시키고 생성된 침전물은 흡인 여과시키고 물로 세척하고 아세토니트릴로 결정화한다.
수득량 : 12.3g(이론치의 59%)
융점 : 247 내지 249℃

시스-2-에틸사이클로헥산-5'-스피로-3'-(3,5-디브로모페닐)-히단토인
a) 시스-2-에틸사이클로헥산-5'-스피로-3'-(2,4-디니트로페널)-히단토인
시스-2-에틸사이클로헥산-5'-스피로-히단토인 39g(0.2몰), 1-클로로-2,4-디니트로벤젠 40.4g(0.2몰), 탄산칼륨 83g(0.6몰) 및 테트라부틸 암모니움 하이드로겐 설페이트 1g을 10℃에서 6시간 동안 디메틸설폭사이드 200ml내에서 교반시킨다. 그리고 혼합물을 물 500ml로 희석시키고 흡인 여과하고, 생성물을 물 및 냉메탄올로 세척시키고 건조한다.
수득량 : 62.4g(이론치의 86%)
융점 : 231 내지 232℃
b)시스-2-에틸사이클로헥산-5'-스피로-3'-(2,4-디아미노페닐)-히단토인
시스-2-에틸사이클로헥산-5'-스피로-3'-(2,4-디아미노페닐)-히단토인 36.2g(0.1몰)을 메탄올 500ml 및 진한 염산 20ml에 용해시킨다. 촉매(10% Pb/C) 10g을 가하고 혼합물을 5바 기압하에 65℃ 이하 온도에서 수소화시킨다. 이어서 촉매를 흡인 여과에 의해 제거시키고 혼합물을 증발, 농축시키고 물에 녹인다. 중탄산염으로 중화시키고 형성된 침전물은 흡인 여과시키고 물로 세척하고 이소프로판물로 절정화한다.
수득량 : 25.4g(이론치의 84%)
융점 : 183 내지 185℃
c) 시스-2-에틸사이클로헥산-5'-스피로-3'-(2,4-디아미노-3,5-디브로모페닐)-히단토인
시스-2-에틸사이클로헥산-5'-스피로-3'-(2,4-디아미노페닐)-히단토인 15g (0.05몰)을 빙초산 100ml에 용해시킨다. 주위온도에서 빙초산 20ml에 녹인 브롬 5.06g(0.1몰) 용액을 10분내에 적가한다. 생정된 혼합물을 20분간 더 교반시킨 후 빙수 300ml를 가하고 고체 물질을 흡인 여과에 의해 분리시키고 물로 충분히 세척하고 진공에서 건조한다.
수득량 : 18g(이론치의 78.3%)
융점 : 263 내지 265℃
d) 시스-2-에틸사이클로헥산-5'-스피로-3'-(3,5-디브로모페닐)-히단토인
디메틸포름아미드 50ml에 녹인 이소아밀니트라이트 3.5g(0.03몰) 용액을 55℃로 가열하고 이 온도에서 디메틸포름아미드 20ml에 녹인 시스-2-에틸-사이클로헥산-5'-스피로-3'-(2,4-디아미노-3,5-디브로모페닐)-히단토인 6.9g(0.015몰) 용액을 1시간내에 적가한다. 온도를, N2의 형성이 끝날때까지 70 내지 80℃로 올린다. 혼합물을 진공에서 증발, 농축시키고 암색 잔사를 컬럼크로마토그라피(실리카겔/에틸 아세테이트)로 정제한다.
수득량 : 3.9g(이론치의 59%)
융점 : 213 내지 215℃

시스-2-메틸사이클로헥산-5'-스피로-히단토인 192g(1몰), 2,6-디클로로-4-니트로피리딘 194g(1몰), 탄산칼륨 207g(1.5몰) 및 테트라부틸 암모니움 하이드로겐 설페이트 3.4g(0.01몰)을 주위온도에서 디메틸설폭사이드 1000ml에 용해시키거나 현탁시킨다. 혼합물을 16시간 더 15 내지 20℃에서 교반시킨다. 그리고 빙수 약 5000ml를 가하고 형성된 침전물을 흡인 여과시키고 물로 충분히 세척하고 아세토니트릴로 재결정시킨다.
수득량 : 250g(이론치의 79%)
융점 : 227 내지 229℃

5-메틸-5-사이클로프로필-3-[4-(2,6-디클로로피리딜) ]-히단토인
디메틸포름아미드 10ml 내에 함유되어 있는 5-메틸-5-사이클로프로필-히단토인 3.1g, 2,6-디클로로-4-니트로피리딘 3.86g 및 탄산칼륨 5.6g으로 이루어진 현탁액을 20℃에서 18시간 교반시키고 물 100ml를 가한다. 30분 교반한 후에, 생성된 침전물을 흡인 여과시키고 염화메틸렌에 녹인다. 건조시키고 용매를 제거시켜 5-메틸-5-사이클로프로필-3-[4-(2,6-디클로로피리딜)]-히단토인 3.2g을 수득한다.
융점 : 160℃(톨루엔/석유에테르 재결정시킴).
수율 : 이론치의 64%.
[실시예 7]
5-메틸-5-사이클로프로필-3-(3,5-디메틸페닐)-히단토인
트리에틸아민 0.3g을, 에탄올 30ml에 함유되어 있는 메틸 1-(3,5-디메틸페닐) -3-[2-(2-사이클로프로필)-프로파노에이트]-요소 0.8g 용액에 가하고 혼합물을 12시간 동안 환류시킨다. 용매를 제거한 후에 잔사를 석유에테르로 처리하고 얻어진 결정성 침전물을 흡인 여과시킨다.
융점 : 121℃
수득량 : 0.6g(이론치의 85%)
[실시예 8]
5-n-프로필-5-(1-메틸에틸)-3-(3,5-디클로로페닐)-2-티오-히단토인
무수 테트라히드로푸란 10ml에 녹인 3,5-디클로로페닐 이소티오시아네이트 2.0g 용액을 20℃에서 무수테트라하이드로푸란 10ml에 녹인 메틸-2-아미노-2-(1-메틸에틸)-펜타노에이트 1.7g 용액에 적가하고 혼합물을 20℃에서 4시간 더 교반시키고 용매를 제거한다. 잔사를 디이소프로필에테르로 재결정시킨다.
융점 : 160℃
수득량 : 1.6g(이론치의 46%).
