Claims (10)
- 제1항에 있어서, 할로겐이 2,2-위치에 있는 화합물 및 약제학적으로 허용되는 그의 염.
- 제1항에 있어서, 할로겐이 6,6-위치에 있는 화합물 및 약제학적으로 허용되는 그의 염.
- 제1항에 있어서, 할로겐이 7,7-위치에 있는 화합물 및 약제학적으로 허용되는 그의 염.
- 제1항에 있어서, 7,7-디플루오로-1,8-디아미노-4-아자-옥탄인 화합물 및 약제학적으로 허용되는 그의 염.
- 제1항에 있어서, 6,6-디플루오로-1,8-디아미노-4-아자-옥탄인 화합물 및 약제학적으로 허용되는 그의 염.
- (a) 일반식(2)의 2,2-디할로-1,4-부탄디올을 벤질 브로마이드와 반응시켜 일반식(3)의 상응하는 1-벤질옥시-2,2-디할로-4-하이드록시부탄 및 1-벤질옥시-3,3-디할로-4-하이드록시부탄의 혼합물을 생성시키고; (b) 상기 이성체를 분리하고, 유리 하이드록실 그룹을 메탄설포닐 클로라이드와 반응시킨 다음, 수득된 1-벤질옥시-젬-디할로-4-메탄설포닐옥시부탄을 칼륨 프탈이마이드와 반응시켜 일반식(4)의 상응하는 N-(4-벤질옥시-젬-디할로부틸)프탈이마이드 유도체를 수득하고; (c) 상기 프탈이마이드를 하이드라진과 반응시켜 상응하는 4-벤질옥시-젬-디할로부틸아민을 생성시키고, 유리 아민을 p-톨루엔설포닐 클로라이드로 보호하여 일반식(5)의 상응하는 N-(4-벤질옥시-젬-디할로부틸)-p-톨루엔 설폰아마이드 유도체를 생성시키며; (d) 상기 p-톨루엔설폰아마이드 유도체를 3-브로모프로필프탈아마이드로 알킬화하여 일반식(6)의 상응하는 1-프탈이마이도-4-p-톨루엔설포닐-젬-디할로-8-벤질옥시-4-아자-옥탄 유도체를 생성시키며; (e) 상기 벤질옥시 그룹을 분리시켜 제1알코올을 생성시키고, 상기 제1알코올을 메탄설포닐 클로라이드와 반응시킨 다음, 그와 같이 수득된 1-프탈이마이도-4-p-톨루엔설포닐-젬-디할로-8-메탄설포닐옥시-4-아자-옥탄을 칼륨 프탈이마이드와 반응시켜 일반식(7)의 상응하는 1,8-디프탈이마이도-4-p-톨루엔설포닐-젬-디할로-4-아자-옥탄 유도체를 수득하고; (f) 상기 1,8-디프탈이마이도-4-p-톨루엔설포닐-젬-디할로-4-아자-옥탄을 에탄올 중 하이드라진과 함께 가열하여 프탈로일 보호 그룹을 제거한 다음, 수성 HBr과 함께 가열하여 토실 보호그룹을 제거하여 일반식(1-a)의 목적하는 6,6-또는, 7,7-디할로-1,8-디아미노-4-아자-옥탄 유도체를 수득함을 특징으로 하여, 하기 일반식(1)의 6,6-디할로 또는 7,7-디할로-1,8-디아미노-4-아자-옥탄 유도체를 제조하는 방법.상기식에서, X 및 Y는 수소 또는 할로겐이고, 단, 두개의 할로겐은 어느 특정시간에 단 하나의 탄소원자상에 존재하며, X' 및 Y'은 염소 또는 불소이며, Bz는 벤질 그룹이며, Pht는 프탈로일 그룹이며, Tos는 p-톨루엔설포닐 그룹이다.
- (a) 2,2-디할로-1,4-부탄디올을 메탄설포닐 클로라이드 2당량과 반응시켜 상응하는 1,4-비스-메탄설포닐옥시-2,2-디할로부틸을 생성시킨 다음, 디아자비사이클로온데센과 함께 가열하여 일반식(9)의 상응하는 1-메탄설포닐옥시-2,2-디할로-3-부텐을 제조하고; (b) 상기 1-메탄설포닐옥시 유도체를 칼륨 프탈이마이드와 반응시킨 다음, 생성된 프탈로일 잔기를 분리시켜 일반식(10)의 상응하는 2,2-디할로-3-부테닐아민을 수득하고; (c) 상기 아민을 p-톨루엔설포닐 클로라이드로 보호한 다음, 사익 보호된 유도체를 4-브로모부틸 프탈이마이드로 알킬화하여 일반식(11)의 상응하는 N-(4-프탈이마이도부틸)-N-(2,2-디할로-3-부테닐)-p-톨루엔 설폰아마이드를 수득하고; (d)상기 N-(4-프탈이마이도부틸)유도체의 2중결합을 산화시킨 다음, 보란-메틸설파이드 착화합물로 환원시켜 일반식(12)의 상응하는 8-프탈이마이도-4-p-톨루엔설포닐-2,2-디할로-4-아자-옥탄올을 생성시키고; (e) 상기 2,2-디할로-4-아자-옥탄올을 유도체 메탄설포닐 클로라이드와 반응시켜 상응하는 1-메탄설포닐옥시-8-프탈이마이도-4-p-톨루엔설포닐-4-아자-옥탄을 생성시킨 다음, 칼륨 프탈이마이드와 반응시켜 일반식(13)의 상응하는 2,2-디할로-1,8-디프탈이마이도-4-p-톨루엔설포닐-4-아자-옥탄을 수득하고; (f) 에탄올중 하이드라진과 함께 가열하여 프탈로일 보호 그룹을 제거한 다음, 수성 HBr과 함께 가열하여 토실 보호그룹을 제거하여 일반식(1-b)의 목적하는 2,2-디할로-1,8-디아미노-4-아자-옥탄-유도체를 수득함을 특징으로 하여, 하기 일반식(1)의 2,2-디할로-1,8-디아미노-4-아자-옥탄 유도체를 제조하는 방법.상기식에서, X 및 Y는 수소 또는 할로겐이고 단, 두개의 할로겐은 어느 특정시간에 단 하나의 탄소원자상에 존재하며, Mes는 메탄설포닐 그룹이며, Pht는 프탈로일 그룹이며, Tos는 p-톨루엔설포닐 그룹이다.
- 치료학적 유효량의 제1항의 화합물을 약제학적으로 허용되는 담체 또는 희석제와의 배합물 또는 혼합물 형태로 함유하는 증식억제성 또는 항암성 조성물.
- 제10항에 있어서, 치료학적 유효량의 2-디플루오로메틸-2,5-디아미노펜타노산 또는 [2R, 5R]-6-헵틴-2,5-디아민을 추가로 함유하는 증식 억제성 또는 항암성 조성물.
