Claims (45)
- (a) 단일 작용성 V 세그먼트 또는 V 세그먼트의 소부류에 상보성인 서열을 각각 포함하는 다수의 V-세그먼트 프라이머 및
(b) J 세그먼트에 상보성인 서열을 각각 포함하는 다수의 J-세그먼트 프라이머를 포함하는 조성물로서,
상기 V 세그먼트 및 J-세그먼트 프라이머는 다중 폴리머라제 연쇄 반응(PCR)에 의해 TCR 또는 IG CDR3 영역을 증폭시켜 TCR 또는 IG 유전자의 다양성을 정량화하기에 충분한 다수의 증폭된 DNA 분자를 생성하는, 조성물. - 제1항에 있어서, 각각의 V-세그먼트 프라이머가 단일 Vγ 세그먼트 또는 유사한 Vγ 세그먼트 부류에 상보성인 서열을 포함하고, 각각의 J 세그먼트 프라이머가 Jγ 세그먼트에 상보성인 서열을 포함하며, V 세그먼트 및 J-세그먼트 프라이머는 TCRγ CDR3 영역을 증폭시키는 조성물.
- 제1항에 있어서, 각각의 V-세그먼트 프라이머가 단일 Vδ 세그먼트 또는 유사한 Vδ 세그먼트 부류에 상보성인 서열을 포함하고, 각각의 J 세그먼트 프라이머가 Jδ 세그먼트에 상보성인 서열을 포함하며, V 세그먼트 및 J-세그먼트 프라이머는 TCRδ CDR3 영역을 증폭시키는 조성물.
- 제1항에 있어서, 각각의 V-세그먼트 프라이머가 단일 Vα 세그먼트 또는 유사한 Vα 세그먼트 부류에 상보성인 서열을 포함하고, 각각의 J 세그먼트 프라이머가 Jα 세그먼트에 상보성인 서열을 포함하며, V 세그먼트 및 J-세그먼트 프라이머는 TCRα CDR3 영역을 증폭시키는 조성물.
- 제1항에 있어서, 각각의 V-세그먼트 프라이머가 단일 Vβ 세그먼트 또는 유사한 Vβ 세그먼트 부류에 상보성인 서열을 포함하고, 각각의 J 세그먼트 프라이머가 Jβ 세그먼트에 상보성인 서열을 포함하며, V 세그먼트 및 J-세그먼트 프라이머는 TCRβ CDR3 영역을 증폭시키는 조성물.
- 제1항에 있어서, V 세그먼트가 유사한 어닐링 강도를 갖는 조성물.
- 제1항에 있어서, 모든 J 세그먼트 프라이머가 동일한 보존된 프레임워크 영역 모티프에 어닐링되는 조성물.
- 제1항에 있어서, 상기 증폭된 DNA 분자가 상기 보존된 모티프로부터 출발하고 J 세그먼트를 진단학적으로 동정하며 V 세그먼트로의 결합을 포함하는 조성물.
- 제1항에 있어서, 증폭된 DNA 분자 내의 영역에 하이브리드화하는 서열분석 올리고뉴클레오타이드 세트를 추가로 포함하는 조성물.
- 제1항에 있어서, 상기 증폭된 DNA가 V-D-J 결합을 확장하는 조성물.
- 제1항에 있어서, 상기 V-세그먼트 또는 J-세그먼트가, 밀접하게 관련된 서열의 병합에 의해 서열 에러-보정을 함유하도록 선택되는 조성물.
- 제1항에 있어서, mRNA로부터 cDNA를 생성하기 위한 범용 C 세그먼트 프라이머를 추가로 포함하는 조성물.
- 제5항에 있어서, 상기 V 세그먼트 프라이머가 재조합 시그날 서열(RSS)에 대해 Vβ 내의 위치 -43에 고정되어 있는 조성물.
- 제5항에 있어서, 상기 다수의 V 세그먼트 프라이머가 14개의 상이한 Vβ 유전자에 특이적인 적어도 14개 프라이머로 이루어진 조성물.
- 제5항에 있어서, 상기 V 세그먼트 프라이머가 서열번호: 1 내지 45로 이루어진 그룹으로부터 선택되는 서열을 갖는 조성물.
- 제5항에 있어서, 상기 V 세그먼트 프라이머가 서열번호: 58 내지 102로 이루어진 그룹으로부터 선택되는 서열을 갖는 조성물.
- 제5항에 있어서, 각각의 Vβ 세그먼트 또는 Vβ 세그먼트 부류에 대한 V 세그먼트 프라이머가 존재하는 조성물.
- 제5항에 있어서, 상기 프라이머가 인트론/엑손 경계를 교차하지 않는 조성물.
- 제5항에 있어서, 상기 J 세그먼트 프라이머가 Jβ 세그먼트의 보존된 요소와 하이브리화하고, 유사한 어닐링 강도를 갖는 조성물.
- 제5항에 있어서, 상기 다수의 J 세그먼트 프라이머가 5개의 상이한 Jβ 유전자에 특이적인 적어도 5개의 프라이머로 이루어진 조성물.
- 제5항에 있어서, 상기 J 세그먼트 프라이머가 서열번호: 46 내지 57 및 483으로 이루어진 그룹으로부터 선택되는 서열을 갖는 조성물.
- 제5항에 있어서, 상기 J 세그먼트 프라이머가 서열번호: 103 내지 113, 468 및 484으로 이루어진 그룹으로부터 선택되는 서열을 갖는 조성물.
- 제5항에 있어서, 각각의 Jβ 세그먼트에 대한 J 세그먼트 프라이머가 존재하는 조성물.
- 제5항에 있어서, 상기 증폭된 Jβ 유전자 세그먼트가 RSS 부위 하류의 위치 +11 내지 +14에 유니크(unique)한 4개 염기 태그를 갖는 조성물.
- 제24항에 있어서, 상기 서열분석 올리고뉴클레오티드가 RSS 부위 하류의 위치 +11 내지 +14에서 증폭된 Jβ 유전자 세그먼트 내에 4개 염기 태그에 인접하여 하이브리드화하는 조성물.
- 제24항에 있어서, 상기 서열분석 올리고뉴클레오티드가 서열번호: 470 내지 482로 이루어진 그룹으로부터 선택되는 조성물.
- (a) 단일 작용성 V 세그먼트 또는 V 세그먼트의 소부류에 상보성인 서열을 각각 포함하는 다수의 V 세그먼트 프라이머 및
(b) J 세그먼트에 상보성인 서열을 각각 포함하는 다수의 J 세그먼트 프라이머를 포함하는 조성물로서,
상기 V 세그먼트 및 J 세그먼트 프라이머는 다중 폴리머라제 연쇄 반응(PCR)에 의해 항체 중쇄(IGH) VH 영역을 증폭시켜 항체 중쇄 유전자의 다양성을 정량화하기에 충분한 다수의 증폭된 DNA 분자를 생성하는, 조성물. - (a) 단일 작용성 V 세그먼트 또는 V 세그먼트의 소부류에 상보성인 서열을 각각 포함하는 다수의 V 세그먼트 프라이머 및
(b) J 세그먼트에 상보성인 서열을 각각 포함하는 다수의 J 세그먼트 프라이머를 포함하는 조성물로서,
상기 V 세그먼트 및 J 세그먼트 프라이머는 다중 폴리머라제 연쇄 반응(PCR)에 의해 항체 경쇄(IGL) VL 영역을 증폭시켜 항체 경쇄 유전자의 다양성을 정량화하기에 충분한 다수의 증폭된 DNA 분자를 생성하는, 조성물. - (a) 단일 작용성 V 세그먼트 또는 V 세그먼트의 소부류에 상보성인 서열을 각각 포함하는 다수의 V 세그먼트 프라이머를 선택하는 단계;
(b) J 세그먼트에 상보성인 서열을 각각 포함하는 다수의 J 세그먼트 프라이머를 선택하는 단계; 및
(c) V 세그먼트 및 J 세그먼트 프라이머를 게놈 DNA 샘플과 조합하여 다중 폴리머라제 연쇄 반응(PCR)에 의해 TCR CDR3 영역을 증폭시킴으로써 TCR 유전자의 다양성을 정량화하기에 충분한 다수의 증폭된 DNA 분자를 생성하는 단계를 포함하는 방법. - 제29항에 있어서, 각각의 V 세그먼트 프라이머가 단일 Vβ 세그먼트 또는 Vβ 세그먼트 부류에 상보성인 서열을 포함하고, 각각의 J 세그먼트 프라이머가 Jβ 세그먼트에 상보성인 서열을 포함하며, V 세그먼트 및 J 세그먼트 프라이머와 게놈 DNA 샘플과의 조합이 다중 폴리머라제 연쇄 반응(PCR)에 의해 TCRB CDR3 영역을 증폭시켜 다수의 증폭된 DNA 분자를 생성하는 방법.
- 제30항에 있어서, 증폭된 DNA 분자를 서열분석하는 단계를 추가로 포함하는 방법.
- 제31항에 있어서, 상기 서열분석 단계가, 증폭된 DNA 분자 내의 소정 영역과 하이브리드화하는 서열분석 올리고뉴클레오티드 세트를 사용하는 방법.
- 제32항에 있어서, 증폭된 DNA 분자 중에서 TCRβ CDR3 서열의 전체 다양성을 계산하는 단계를 추가로 포함하는 방법.
- 제33항에 있어서, 상기 방법은 정상 사람 피검체의 전체 다양성이 1*106개 초과의 서열임을 나타내는 방법.
- 제33항에 있어서, 상기 방법은 정상 사람 피검체의 전체 다양성이 2×106개 초과의 서열임을 나타내는 방법.
- 제33항에 있어서, 상기 방법은 정상 사람 피검체의 전체 다양성이 3×106개 초과의 서열임을 나타내는 방법.
- 환자의 TCR CDR3 서열의 다양성을 측정하는 단계; 및
피검체의 다양성을 정상 피검체로부터 수득한 다양성과 비교하는 단계를 포함하는,
사람 환자에서 면역결핍을 진단하는 방법. - 제37항에 있어서, TCR 서열의 다양성을 측정하는 단계가
(a) 단일 작용성 V 세그먼트 또는 V 세그먼트의 소부류에 상보성인 서열을 각각 포함하는 다수의 V 세그먼트 프라이머를 선택하는 단계;
(b) J 세그먼트에 상보성인 서열을 각각 포함하는 다수의 J 세그먼트 프라이머를 선택하는 단계;
(c) V 세그먼트 및 J 세그먼트 프라이머를 게놈 DNA 샘플과 조합하여 다중 폴리머라제 연쇄 반응(PCR)에 의해 TCR CDR3 영역을 증폭시킴으로써 다수의 증폭된 DNA 분자를 생성하는 단계;
(d) 증폭된 DNA 분자를 서열분석하는 단계; 및
(e) 증폭된 DNA 분자 중에서 TCR CDR3 서열의 전체 다양성을 계산하는 단계를 포함하는 방법. - 제38항에 있어서, 게놈 DNA의 적어도 2개 샘플의 다양성이 비교되는 방법.
- 제40항에 있어서, 게놈 DNA의 하나의 샘플이 환자의 것이고 다른 샘플이 정상 피검체의 것인 방법.
- 제40항에 있어서, 게놈 DNA의 하나의 샘플이 치료학적 치료 전의 환자의 것이고 다른 샘플이 치료 후의 환자의 것인 방법.
- 제40항에 있어서, 게놈 DNA의 2개 샘플이 치료 동안 상이한 시간에서의 동일한 환자의 것인 방법.
- 제40항에 있어서, 질환이 게놈 DNA의 샘플 중에서 다양성의 비교에 기초하여 진단되는 방법.
- 제40항에 있어서, 사람 환자의 면역경쟁이 비교에 의해 평가되는 방법.
