Claims (29)
- 알부민과 융합된 인자 VII 또는 인자 VIIa 폴리펩타이드를 포함하는 알부민 융합 폴리펩타이드로서, 상기 인자 VII 또는 인자 VIIa 폴리펩타이드가 상기 융합 폴리펩타이드의 N-말단에 위치하고, 상기 융합 폴리펩타이드가 인자 VII 또는 VIIa 생물학적 활성을 가지며, 상기 인자 VII 또는 인자 VIIa 폴리펩타이드가 펩타이드 링커를 통해 상기 알부민과 분리되어 있고, 상기 링커가 25개 이상의 아미노산을 포함하고 글라이신 및 세린을 포함하는 반복 단위를 포함하는, 알부민 융합 폴리펩타이드.
- 제1항에 있어서, 상기 융합 폴리펩타이드가 각각의 비융합된 야생형 인자 VII 또는 인자 VIIa와 비교하여 25% 이상의 몰(molar) 특이적 인자 VII 또는 VIIa의 생물학적 활성을 갖는 알부민 융합 폴리펩타이드.
- 제1항에 있어서, 상기 융합 폴리펩타이드가 비융합된 인자 VII 또는 인자 VIIa와 비교하여 증가된 생체내 기능성 혈장 반감기를 갖는, 인자 VII 또는 인자 VIIa 폴리펩타이드를 포함하는 알부민 융합 폴리펩타이드.
- 제3항에 있어서, 상기 융합 폴리펩타이드가 비융합된 인자 VII 또는 인자 VIIa와 비교하여 100% 이상까지 증가된 생체내 기능성 반감기를 갖는, 인자 VII 또는 인자 VIIa 폴리펩타이드를 포함하는 알부민 융합 폴리펩타이드.
- 제1항에 있어서, 상기 융합 폴리펩타이드가 링커 부재인 인자 VII 또는 FVIIa의 융합 단백질과 비교하여 25% 이상 증가된 몰 특이적 활성을 갖는, 인자 VII 또는 인자 VIIa 폴리펩타이드를 포함하는 알부민 융합 폴리펩타이드.
- 제1항에 있어서, 상기 링커가 프로테아제 절단 부위를 포함하는, 알부민 융합 폴리펩타이드.
- 제6항에 있어서, 상기 절단 부위가 인자 IIa, 인자 IXa, 인자 Xa, 인자 XIa, 인자 XIIa, 활성화된 단백질 C, 엘라스타제 또는 칼리크레인으로 이루어진 그룹으로부터 선택되는 응고 프로테아제에 의해 절단될 수 있는 알부민 융합 폴리펩타이드.
- 제1항에 있어서, 상기 링커가 해독후 변형을 위한 부위의 삽입에 의해 변형되고, 상기 해독후 변형이 구조 Asn-X-Ser/Thr(여기서, X는 프롤린을 제외한 임의의 아미노산을 나타낸다)의 하나 이상의 N-당화 부위를 포함하는, 알부민 융합 폴리펩타이드.

- 제1항에 따른 알부민 융합 폴리펩타이드를 암호화하는 폴리뉴클레오타이드 서열을 포함하는 핵산 분자로서, 이때, 상기 폴리뉴클레오타이드 서열이 상기 알부민 융합 폴리펩타이드의 감마 카복실화를 매개하는 프로펩타이드를 암호화하는 뉴클레오타이드 서열의 3' 말단에 위치하는, 핵산 분자.
- 제13항에 따른 핵산 분자를 포함하는 플라스미드 또는 벡터.
- 제14항에 있어서, 발현 벡터인 플라스미드 또는 벡터.
- 제15항에 있어서, 사람 유전자 치료에 사용하기 위한 전달 벡터인 벡터.
- 제14항 내지 제16항 중 어느 한 항에 따른 플라스미드 또는 벡터를 포함하는 숙주 세포.
- a. 숙주 세포에서 발현할 수 있는 알부민 융합 폴리펩타이드를 암호화하는 뉴클레오타이드 서열을 포함하는 핵산을 제공하는 단계;b. 상기 숙주 세포에서 핵산 분자를 발현하여 알부민 융합 폴리펩타이드를 생성시키는 단계 및c. 상기 알부민 융합 폴리펩타이드를 정제하는 단계를 포함하는, 제1항 내지 제8항, 제11항 및 제12항 중 어느 한 항에 따른 알부민 융합 폴리펩타이드를 제조하는 방법.
- 제1항 내지 제8항, 제11항 및 제12항 중 어느 한 항에 따른 알부민 융합 폴리펩타이드, 제13항에 따른 핵산 분자, 또는 제14항 내지 제16항 중 어느 한 항에 따른 플라스미드 또는 벡터를 포함하는, 혈액 응고 장애를 치료하거나 예방하기 위한 약제학적 조성물.
- 제1항 내지 제8항, 제11항 및 제12항 중 어느 한 항에 따른 알부민 융합 폴리펩타이드를 포함하는, A형 혈우병, B형 혈우병, 수술 관련 출혈성 에피소드, 선천성 또는 후천성 혈우병을 갖는 환자에서 응고 인자 VIII 또는 IX에 대한 억제제를 사용한 출혈 에피소드 및 수술, 항혈소판 약물 또는 항-응고 약물로부터 선택되는 약물 치료 결과로서 발병되는 역전된 지혈 결손, 2차 지혈의 개선, 감염 또는 비타민 K 결핍 또는 심각한 간 질환으로부터 선택되는 병에 걸린동안에 발병되는 지혈 결손, 간 절제, 뱀에 물린 결과로서 발병되는 지혈 결손, 위장 출혈, 외상, 대량 수혈 결과(희석 혈액응고병증), FVIII 및 FIX이외의 응고 인자 결핍, 본 빌레브란트 질환, FI 결핍, FV 결핍, FVII 결핍, FX 결핍, FXIII 결핍, 용혈성 요독증성 증후군, 선천성 또는 후천성 혈소판 질환, 혈소판 감소증, 특발성 혈소판 감소성 자반증, 혈전성 혈소판 감소성 자반증, 용혈 상승 간 효소 낮은 혈소판 증후군, 버나드-소울리어(Bernard-Soulier) 증후군, 글랜즈만(Glanzmann) 혈소판무력증, 헤파린 유도 혈소판 감소증, 체디악-히가히(Chediak-Higahi) 증후군, 헤르만스키-푸들락(Hermansky-Pudlak)-증후군, 렌두-오슬러(Rendu-Osler) 증후군, "헤노흐-숀라인(Henoch-Schonlein) 자반증 및 상처 치유중 하나 이상의 증상을 치료하기 위한 약제학적 조성물.
