Claims (10)
- 試料支持部(120)の下方に取り付けられて使用時に前記試料支持部(120)で光の線または平面を提供する反転照明対物レンズ(110)と、前記支持部(120)の上方に取り付けられた第1の画像収集対物レンズ(130)及び第2の画像収集対物レンズ(140)と、を含み、前記反転照明対物レンズ(110)は、照明対物レンズ光軸(111)を有し、前記第1の画像収集対物レンズ(130)及び前記第2の画像収集対物レンズ(140)のそれぞれは、画像収集対物レンズ光軸(131)及び画像像収集対物レンズ光軸(141)を有する、選択的平面照明を提供する光学配置(100)であって、切り替え可能な照明光(132)及び照明光(142)は、前記照明対物レンズ光軸(111)に対して互に平行であると共に前記照明対物レンズ光軸(111)に対してそれぞれ横方向にオフセットされて前記照明対物レンズ(110)に向かって伝搬するように構成され、それにより前記照明対物レンズ(110)から出る前記照明光(132)及び前記照明光(142)は、前記照明対物レンズ光軸(111)に対して斜めの角度で前記試料支持部(120)に向かって伝搬し、前記画像収集対物レンズ光軸(131)及び前記画像像収集対物レンズ光軸(141)はそれぞれ、前記照明対物レンズ光軸(111)に対して鈍角である角度αを成しており、前記試料支持部(120)で伝搬する光にほぼ垂直である、光学配置(100)。
- 前記角度αが、100〜170度である、請求項1に記載の光学配置(100)。
- 前記照明光(132)及び前記照明光(142)が、前記第1または第2の画像収集対物レンズ(130,140)に交互に画像光を提供するように、それぞれの第1および第2の経路に沿って前記照明対物レンズ光軸(111)に対して互いに反対側に交互に伝搬するように構成される、請求項1または2に記載の光学配置(100)。
- 前記照明光(132)及び前記照明光(142)が、コヒーレント光であり、前記光学配置(100)がさらに、光線に集光した、またはコリメートされた光の平面またはシートの形態の前記照明光(132)及び前記照明光(142)を提供する光学マニピュレータを含む、請求項1、2または3に記載の光学配置(100)。
- 試料を照明してSPIMを提供する方法であって、
a)請求項3または4に記載の光学配置(100)を提供するステップと、
b)コヒーレントな照明光(132)及び照明光(142)を照明の方向に伝搬させるステップと、
c)前記第1および第2の画像収集対物レンズ(130,140)によって収集された1つまたは複数の画像を捕捉するステップと、
を含む、方法。 - 請求項1〜4のいずれか1項に記載の光学配置(100)を含む、SPIM機器(10)。
- SIM、TIRFおよびPK顕微鏡法技術の任意の1つまたは複数をさらに実行するように適合可能な、請求項6に記載のSPIM機器(10)。
- 前記照明対物レンズ(110)と前記試料支持部(120)との間に空間があり、前記空間が、液体によって架橋される、請求項6または7に記載のSPIM機器(10)。
- 前記試料支持部(120)が、好ましくは連続動作で前記照明対物レンズ光軸(111)に垂直に移動可能である、請求項6、7または8に記載のSPIM機器(10)。
- 照明対物レンズ光軸(111)を有する照明対物レンズ(110)と、前記照明対物レンズ光軸(111)に垂直な方向に前記光軸(111)に対して、好ましくは連続動作で移動可能な試料支持部(120)とを含む、SPIM機器(10)。
