- 【特許請求の範囲】 1. 親のテルマミル様α−アミラーゼの変異体であって、前記変異体はα−ア ミラーゼ活性を有し且つ前記親のα−アミラーゼと比較すると次の性質:基質特 異性、基質結合、基質開裂パターン、熱安定性、pH/活性プロフィール、pH /安定性プロフィール、酸化に対する安定性、Ca2+依存性および比活性のうちの 少なくとも1つの性質に変更を示し、前記変異体は配列番号2に示されるアミノ 酸配列中に下記の変異の1つまたは複数に相当する1つまたは複数の変異: ・変異:A181E,D,Q,N,V;I201W,F,Lを含む、I201(嵩張ったアミノ酸);Y203Q;Q9 K,L,E;F11R,K,E;E12Q;D100N,L;V101H,R,K,D,E,F;V102A,T;I103H,K;N104R,K,D;H1 05R,K,D,E,W,F;L196R,K,D,E,F,Y;I212R,K,D,E;L230H,K,I;A232G,H,F,S,V;V233D; K234L,E;I236R,K,N,H,D,E;L241R,K,D,E,F;A260S;W263H;Q264R,D,K,E;N265K,R,D; A269R,K,D,E;L270R,K,H,D,E;V283H,D;F284H;D285N,L;V286R,K,D,E;Y290R,E,K;V3 12R,K,D,E;F323H;D325N;N326K,H,D,L;H327Q,N,E,D,F;Q330L,F;G332D;Q333R,K,H, E,L;S334A,V,T,L,I,D;L335G,A,S,T,N;E336R+R375E;T337D,K;T338D,E;T339D;Q36 0K,R,E;D365N;またはG371D,R; ・H68,H91,H247,R305,K306,H382,K389,H405,H406,H450またはR483の位 置における置換; ・変異:H140Y;H142Y;H159Y;H140D+H142R;H140K+H142D;またはH142Y+H156Y; ・欠失:K370*+G371*+D372*またはD372*+S373*+Q374*を含む、 T369からI377までの部分配列中の3アミノ酸の欠失; ・次の配列(配列番号は左から右に増加する):I-P-T-H-S-V;I-P-T-H-G-V;I-P- Q-Y-N-Iのうちの1つによる、T369からI377までの部分アミノ酸配列の置換; ・R169I,L,F,TまたはR173I,L,F,Tを含む、R169またはR173の位置における置換; ・変異:H156D;I201F;I212F;A209L,T;またはV208I; ・N172R,H,K;A181T;N188P;N190L,F;H205C;D207Y;A209L,T,V;A210S;E211Q;Q264A, E,L,K,S,T;N265A,S,T,Y;または Q264S+N265Yを含む、N172,A181,N188,N190,H205,D207,A209,A210,E211 ,Q264またはN265の位置における置換; ・変異: H156Y+A181T+A209V; H156Y+A181T+N190F+A209V+Q264S; A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+A209V; A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+N190F+A209V;または A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+N190F+A209V+Q264S を含んで成る変異体。 2. 親のテルマミル様α−アミラーゼに比較して高められた、低pHおよび低 Ca2+濃度での安定性を示し、且つ次の変異: H156Y+A181T+A209V; H156Y+A181T+N190F+A209V+Q264S; A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+A209V; A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+N190F+A209V;または A1*+N2*+L3V+M15T+R23K+S29A+A30E+Y31H+A33S+E34D+H35I+H156Y+A1 81T+N190F+A209V+Q264S から選ばれた変異を含んで成る、請求項1に記載の変異体。 3. 前記親のテルマミル様α−アミラーゼが、 配列番号2に示される配列を有するB.リヘニフォルミスα−アミラーゼ; 配列番号4に示される配列を有するB.アミロリクファシエンスα−アミラーゼ ; 配列番号6に示される配列を有するB.ステアロサーモフィラスα−アミラーゼ ; 図1および2に示される配列を有するバシラスNCIB 12512株α−アミラーゼ; 図2に示される配列を有するバシラスNCIB 12513株α−アミラーゼ;および 図2に示される配列を有するバシラス種#707α−アミラーゼから選ばれる、請求 項1または2に記載の変異体。 4. 請求項1〜3のいずれか一項に記載のα−アミラーゼ変異体をコードする DNA配列を含んで成る、DNA構成物。 5. 請求項4に記載のDNA構成物を担持する組換え発現ベクター。 6. 請求項4に記載のDNA構成物または請求項5に記載のベクターにより形 質転換されている細胞。 7. 微生物である、請求項6に記載の細胞。 8. 細菌または真菌である、請求項6に記載の細胞。 9. グラム陽性菌、例えばバシラス・サチリス(Bacillus subtilis )、バシラス・リヘニフォルミス(Bacillus licheniformis)、バシ ラス・レンタス(Bacillus lentus)、バシラス・ブレビス(Bacillus brevis )、バシラス・ステアロサーモフィラス(Bacillus stearothermophilus)、バ シラス・アルカロフィラス(Bacillus alkalophilus)、バシラス・アミロリク ファシエンス(Bacillus amyloliquefaciens)、バシラス・コアギュランス(Ba cillus coagulans )、バシラス・サーキュランス(Bacillus circulans)、バシ ラス・ロータス(Bacillus lautus)またはバシラス・スリンジエンシス(Baci llus thuringiensis )である、請求項8に記載の細胞。 10.洗浄および/または食器洗いのための、請求項1〜3のいずれか一項に記 載のα−アミラーゼ変異体の使用。 11.織物デサイジングのための、請求項1〜3のいずれか一項に記載のα−ア ミラーゼ変異体の使用。 12.デンプン液化のための、請求項1〜3のいずれか一項に記載のα−アミラ ーゼ変異体の使用。 13.場合により無塵性粉末、安定化液体または保護酵素の形で請求項1〜3の いずれか一項に記載のα−アミラーゼ変異体を含んで成る洗剤添加剤。 14.添加剤1gあたり0.02〜200mgの酵素タンパク質を含有する、請求項13に 記載の洗剤添加剤。 15.別の酵素、例えばプロテアーゼ、リパーゼ、ペルオキシダーゼ、別のデン プン分解酵素および/またはセルラーゼを更に含んで成る、請求項13または14に 記載の洗剤添加剤。 16.請求項1〜3のいずれか一項に記載のα−アミラーゼ変異体を含んで成る 洗剤組成物。 17.別の酵素、例えばプロテアーゼ、リパーゼ、ペルオキシダー ゼ、別のデンプン分解酵素および/またはセルラーゼを更に含んで成る、請求項 16に記載の洗剤組成物。 18.請求項1〜3のいずれか一項に記載のα−アミラーゼ変異体を含んで成る 手動または自動食器洗い用洗剤組成物。 19.別の酵素、例えばプロテアーゼ、リパーゼ、ペルオキシダーゼ、別のデン プン分解酵素および/またはセルラーゼを更に含んで成る、請求項18に記載の食 器洗い用洗剤組成物。 20.請求項1〜3のいずれか一項に記載のα−アミラーゼ変異体を含んで成る 手動または自動洗濯用洗剤組成物。 21.別の酵素、例えばプロテアーゼ、リパーゼ、ペルオキシダーゼ、別のデン プン分解酵素および/またはセルラーゼを更に含んで成る、請求項20に記載の洗 濯用洗剤組成物。 22.下記のもの: (i)配列番号2に示される配列を有するB.リヘニフォルミスからのα−アミ ラーゼと、配列番号6に示される配列を有するB.ステアロサーモフィラスα− アミラーゼ(親のテルマミル様α−アミラーゼとして)に由来する本発明の1ま たは複数の変異体との混合物; (ii)配列番号6に示される配列を有するB.ステアロサーモフィラスからのα −アミラーゼと、1または複数の別の親のテルマミル様α−アミラーゼに由来す る1または複数の本発明の変異体との混合物;または (iii)配列番号6に示される配列を有するB.ステアロサーモフィラスα−ア ミラーゼ(親のテルマミル様α−アミラーゼとして)に由来する1または複数の 本発明の変異体と、1または複数の別の親のテルマミル様α−アミラーゼに由来 する1または複数の本発明の変異体との混合物 を含んで成る組成物。 23.親のテルマミル様α−アミラーゼの変異体の作製方法であって、前記変異 体は親のα−アミラーゼと比較して高められた低pHおよび低カルシウム濃度で の安定性を示し、 (a) 親のテルマミル様α−アミラーゼをコードするDNA配列をランダム変異 誘発にかけ、 (b) 段階(a)で得られた変異型DNA配列を宿主細胞中で発現せしめ、 そして (c) 親のα−アミラーゼに比較して高められた低pHおよび低カルシウム濃度 での安定性を有する変異型α−アミラーゼを発現する宿主細胞についてスクリー ニングする ことを含んで成る方法。
