Claims (19)
1. Hybridom med ATCC-depositionsnummer HB 8746.
2. Receptor, kännetecknad av att den (a) im-munoreagerar med apoprotein B-100, och (b) utsöndras av hybridom med ATCC-depositionsnummer HB 8746.
3. Receptor enligt patentkrav 2, kännetecknad av att den är en intakt antikropp.
4. Hybridom med ATCC-depositionsnummer HB 8742.
5. Receptor, kännetecknad av att den a) immu-noreagerar med apoprotein B-100, och (b) utsöndras av hybridom med ATCC-depositionsnummer HB 8742.
6. Receptor enligt patentkrav 5, kännetecknad av att den är en intakt antikropp.
7. Förfarande för att bestämma mängden av apoprotein B-100 i ett kroppsvätskeprov, kännetecknat av att för-farandet omfattar följande steg: (a) det kroppsvätskeprov som skall bestämmas tas; (b) receptormolekyler produceras i biologiskt aktiv form, vilka (i) immunoreagerar med apoprotein B-100, och som (ii) utsöndras av hybridom med ATCC-depositionsnummer HB 8746 eller hybridom med ATCC-depositionsnummer HB 8742; (c) en känd mängd av nämnda kroppsvästkeprov blandas med en pä förhand bestämd mängd av nämnda receptormolekyl för att bilda en immunoreaktionsblandning; (d) nämnda blandning hälls i biologiska bestämningsförhäl-·: landen under en pä förhand bestämd tid, som med avseende pä receptormolekylerna i blandningen är tillräcklig för att immunologiskt binda apoprotein B-100 som finns i provet, varvid immunoreaktant bildas; och (e) mängden immunoreaktant som bildas i nämnda blandning bestäms. 9271 4
8. Förfarande enligt patentkrav 7, kännetecknat av att nämnda kroppsvätskeprov är antingen plasma eller se-rum.
9. Förfarande enligt patentkrav 7, kännetecknat av att nämnda receptor är en intakt antikropp.
10. Förfarande enligt patentkrav 7, kännetecknat av att kroppsvätskeprovet dessutom prepareras för bestämnin-gen genom att inkludera följande ytterligare steg: (f) biologiskt aktiva andra receptormolekyler produceras, vilka immunoreagerar med apoprotein B-100 som finns i nämnda kroppsvätskeprov, och vilka är rester av vardera receptormo-lekylsatsen, som utsöndras av hybridom med ATCC-depositions-nummer HB 8746 eller hybridom med ATCC-depositionsnummer HB 8742; och (g) en pä förhand bestämd mängd av nämnda andra receptormolekyler blandas med nämnda kroppsvätskeprov, varvid en im-munoreaktionsblandning bildas; (h) nämnda immunoreaktionsblandning hälls i biologiska be-stämningsförhällanden under en pä förhand bestämd tid, som med avseende pä receptormolekylerna i blandningen är till-räcklig för att immunologiskt binda apoprotein B-100 som finns i provet, varvid det bildas lagrad immunoreaktant.
11. Förfarande enligt patentkrav 10, kännetecknat av att nämnda receptor är en intakt antikropp.
12. Förfarande enligt patentkrav 10, kännetecknat av att den förstnämnda receptorn fästes vid en fast matris för att bilda en fast bärare; nämnda andra receptor '· innehäller en stämpel med vars hjälp närvaro av nämnda andra receptor ingäende i immunoreaktanten kan uttryckas; och den i steg (e) bestämda immunoreaktanten är en lagrad immunoreaktant i fast fas, som innehäller nämnda stämpel.
13. Diagnostiskt system för att bestämma mängden av apo-B- 100. ett kroppsprov, kännetecknat av att det omfattar: II 9271 4 (a) ett första biologiskt aktivt, specifikt bundet ämne om-fattande receptormolekyler, vilka (i) immunoreagerar med apoprotein B-100, och (ii) är antingen receptormolekyler utsöndrade av hydridom ATCC HB 8746 eller receptormolekyler utsöndrade av ATCC HB 8742? och (b) ett biologiskt aktivt, stämplat, andra specifikt bundet ämne för att uttrycka den immunoreaktion i vilken nämnda första bundna ämne deltar med apo B-100.
14. Bestämningssystem enligt patentkrav 13, k ä n n e -tecknat av att det stämplade, andra specifikt bundna ämnet omfattar andra receptormolekyler antingen av recepto-rer som utsöndras av hybridom ATCC HB 8746 eller receptorer som utsöndras av hybridom ATCC HB 8742.
15. Bestämningssystem enligt patentkrav 14, k ä n n e -tecknat av att stämpeln är ett enzym.
16. Bestämningssystem enligt patentkrav 13, k ä n n e -tecknat av att nämnda första specifikt bundna ämne är fästat vid en fast matris, varvid en fast bärare bildas.
17. Steril affinitetssorbent, som bildas av biologiskt ak-tiva receptormolekyler som immunoreagerar med apoprotein B-100 och väljs i gruppen bestäende av: . ' (a) receptormolekyler utsöndrade av hybridom HB 8746 (b) receptormolekyler utsöndrade av hybridom HB 8742 och (c) en blandning av receptormolekyler under punkt (a) och (b) ; kännetecknad av att nämnda receptormolekyler är fästade vid en fast matris, varvid en fast bärare bildas. « ♦ •
18. Förfarande för att bestämma apoprotein B-100 som ingär i ett kroppsvätskeprov, kännetecknat av att det omf attar följande steg: (a) det kroppsvätskeprov som skall bestämmas tas? (b) en fast bärare produceras, som bildats av en fast mat-• ris, vid vilken dessutom är fästad i biologiskt aktiv form 9271 4 en första receptor, som reagerar immunologiskt med apoprotein B-100, och som utsöndras av hybridom med ATCC-depositionsnummer HB 8746; (c) en andra biologiskt aktiv receptor produceras, som im-munoreagerar med apoprotein B-100, och som utsondras av hybridom med ATCC-depositionsnummer HB 8742, medan nämnda andra receptor är associerad med ett stämpelenzym med vars hjälp närvaro av nämnda andra receptor kan uttryckas i immunoreak-tanten; (d) väsentligt samtidigt blandas: (i) nämnda kroppsprov; och (ii) nämnda fasta bärare; och (iii) nämnda stämplade andra receptor för att bilda en im-munoreaktionsblandning i fast/västkefas; (e) nämnda blandning hälls i biologiska bestämningsförhäl-landen under en pä förhand bestämd period, som med avseende pä nämnda första receptor och nämnda andra stämplade receptor är tillräcklig för att immunologiskt binda apoprotein B-100 som finns i provet, varvid en lagrad immunoreaktant i fast fas och vätskefas bildas; (f) nämnda lagrade immunoreaktant i fast fas skiljs frän nämnda vätskefas; och (g) mängden stämplad andra receptor bestäms, som är bunden vid den lagrade immunoreaktanten i fast fas.
19. Kompetitivt förfarande för att bestämma apoprotein B-100 i ett kroppsvätskeprov, kännetecknat av att det omfattar följande steg: (a) det kroppsvätskeprov som skall bestämmas tas; (b) en fast bärare produceras, som omfattar en fast matris, vid vilken dessutom är fästad en pä förhand bestämd mängd reagens apoprotein B-100; (c) väsentligt samtidigt blandas: (i) nämnda kroppsprov; (ii) nämnda fasta bärare, (iii) en pä förhand bestämd mängd receptormolekyler som im-munoreagerar med apoprotein B-100, utsöndrade antingen ur * hybridom med ATCC-depositionsnummer HB 8746 eller ur hybri- li 9271 4 dom med ATCC-depositionsnuinmer HB 8742, varvid en blandning i fast/vätskefas bildas; (d) nämnda blandning hälls i biologiskt bestämda bestäm-ningsförhällanden under en period som med avseende pa recep-tormolekyler är tillräcklig för att immunologiskt bindas vid apoprotein B-100-molekyler i en fast bärare och apoprotein B-100-molekyler i kroppsvätskeprovet och att bilda en immu-noreaktant i fast fas och en immunoreaktant i vätskefas; och (e) nämnda immunoreaktant i fast fas separeras frän nämnda vätskefas; (f) genom att tillsätta i nämnda immunoreaktant i fast fas en biologiskt aktiv andra receptor utförs blandning, vilken andra receptor reagerar med nämnda första receptor, varvid en andra immunoreaktionsblandning i fast/vätskefas bildas, i vilken nämnda andra receptor kopplad tili enzymstämpeln för-mar uttryeka närvaro av nämnda andra receptor i immunoreaktant en; (g) nämnda andra blandning uppehälls en tid som med avseende pä den stämplade andra reeeptorn är tillräcklig för att immunologiskt binda hela den första reeeptorn, som är närva-rande som immunoreaktant i fast form, varvid en lagrad immunoreaktant i fast fas bildas; och (h) den lagrade immunoreaktanten av nämnda fasta fas separeras frän nämnda vätskefas; och (i) mängden stämplad andra receptor bestäms, som bundits vid nämnda lagrade immunoreaktant i fast fas.
