It is known to add fluorine compounds and sulfonate cleaning agents to dentifrices. Man also already has condensation products of reactive derivatives of higher molecular weight fatty acids, such as Coconut fatty acids, with aminocarboxylic acids, such as aminoacetic acid and methylaminoacetic acid, in a mixture used with capillary-active compounds as a tooth cleaning agent (cf. German Patent 675 837). These were larger additions of 6 to 77% for the purpose of producing a rich Foam and removal of food particles from teeth. It is also already known to be practically pure To add acid amides from fatty acids and aminocarboxylic acids in amounts of up to about 5% dental care products and avoid surface-active additives such as soap.
Es wurde nun gefunden, daß man ganz vorzügliche Zahnpflegemittel mit kombinierter enzymhemmender und konservierender Wirkung erhält, wenn man ein Gemisch aus einer dissoziierbaren Fluorverbindung mit dem Säureamid einer Aminocarbonsäure der FormelIt has now been found that you can get very excellent dentifrices with combined enzyme-inhibiting and preservative effect obtained when a mixture of a dissociable fluorine compound is obtained with the acid amide of an aminocarboxylic acid of the formula

Der Gehalt an freien höheren Fettsäuren soll weniger als 15°/o, auf das Gewicht an Säureamid bezogen, betragen.preferably used in amounts of about 0.05 to 5%, in which R - CO is a saturated fatty acid residue having 12 to 16 carbon atoms, R 'is a hydrogen atom or a lower alkyl radical and X is a hydrogen atom or is a water-soluble salt-forming cation. It is useful as an acid amide N-lauroyl, N-myristoyl or N-palmitoyl sarcosine used. An alkali or tin fluoride or a mixture of both is preferably used as the fluoride. The dentifrice can contain calcium pyrophosphate and / or aluminum oxide as a polishing agent. The content of free higher fatty acids should be less than 15% based on the weight of acid amide, be.
According to a particular embodiment of the invention, the dentifrice contains insoluble polishing agents Alkaline earth polyphosphates or an insoluble alkaline earth metaphosphate, e.g. B. calcium metaphosphate or alkaline earth metal metaphosphate.
„^^^^vThe acid amides used preferably contain a dodecanoyl, tetradecanoyl, hexadecanoyl radical; it is expedient to use N-lauroyl, N-myristoyl or N-palmitoyl sarcosine. They are obtained by condensation of a compound suitable for introducing the acyl radical of a higher fatty acid with the aminocarboxylic acid in question, the latter expediently being a monocarboxylic acid having 2 to 6 carbon atoms. The condensation products can be in the form of their free acids or their salts, e.g. B. the alkali, ammonium, amine or Alkylolaniinsalze, such as mono-, di- or triethanolamine salts, can be used. For example, the sodium salt of N-lauroyl sarcosine, the potassium salt of N-lauroyl sarcosine and the sodium salt of N-myristoyl sarcosine are particularly suitable. the sodium salt of N-palmitoyl-sarcosine, the ethanolamine salt of N-lauroyl-sarcosine, the N-lauroylsarcosine, the sodium salt of N-lauroyl-alanine and the sodium salt of N-lauroyl-glycide. If in the following from the aminocarboxylic acid condensation products or sarcosine, glycine or the like. The content of soaps or free fatty acids should be below 15% and preferably are below 10%.
As the fluorine component, all fluorine compounds can be used, which in aqueous or other, z. B. alcoholic solution allow the cleavage of free fluorine ions, the degree of solubility of the fluorine compound is of minor importance. Preferably, the inorganic fluoride salts, such as alkali metal fluoride, the fluoride salts of alkaline earth metals or of heavy metals are used, in particular sodium fluoride, potassium fluoride, Ammoniutnfluorid, lead fluoride, Cuprofluorid, zinc fluoride, tin fluoride such as stannous or Stannifluorid, Kaliumstannofluorid (SnF 2 · KF) or Stannochloridfluorid, barium or the fluosilicates of sodium, ammonium and aluminum. Of these, the alkali fluorides, such as sodium fluoride and stannous fluoride, and mixtures of these salts, are particularly suitable.
Through the simultaneous use of the acid amides with the fluorine compound, a surprising and unusual combination effect is achieved, which manifests itself in the fact that acid formation and bacterial activity in the oral cavity are effectively suppressed, the loosening of tooth enamel leading to caries formation and damage to the dentin and the holding apparatus of the teeth are avoided, i.e. both the inorganic and the organic substance of the teeth, the jaw and the gums are effectively protected and preserved, so that the dental care products according to the invention meet all the requirements of an ideal dental care product. Such a result cannot be achieved with fluorides alone or only with the condensation products or even with sulfonated cleaning agents. The purity of the condensation products and the proportions are essential for achieving optimal results.
The amount of fluoride depends on the type of fluoride used, its solubility and the type of the dentifrice. The fluoride addition must of course be below the toxic dose. One 2% aqueous sodium fluoride solution, corresponding to about 1% of dissociable fluorine, has proven to be useful proven. In general, the 20% suspension of toothpaste or toothpowder should be used contain so much fluoride that there is a maximum of about 0.1% fluorine ions in solution, and it is sufficient even if the aqueous solution contains about 0.0001% fluorine in ionizable form. In order to the properties of the mixture according to the invention are not adversely affected if the fluoride is to be used are preferably used in amounts up to 2%, especially from 0.05 to about 1%.
Quantities of up to about 5% of the fatty acid amides mentioned can be added. With toothpaste take about 0.5 to 5% of the condensation product, the proportion of which is advantageously greater than that of fluoride.
The mixtures according to the invention can be used for the treatment of the oral cavity, in particular for dental care, be used, e.g. B. in the form of toothpastes, tooth powders, mouthwashes, lozenges, tablets, chewing gum, Dental soap and the like
B. die Calciumsalze derAll suitable, water-insoluble polishing agents can be added to the mixture according to the invention be like sec. and tert. Calcium phosphate, aluminum oxide or hydroxide, magnesium carbonate, Bentonite and mixtures of these substances. In addition, the insoluble alkaline earth polyphosphates are suitable, such as calcium pyrophosphate, which are condensed as derivatives Orthophosphoric acid are considered and the z. B. be obtained by bound water removed by heating. They are also known as molecularly dehydrated phosphates. To this include the alkaline earth salts, e.g. B. the calcium salts of
Die Zahnpasten enthalten meist etwa 20 bis 75%, Zahnpulver 70 bis 95°/o Poliermittel.ίο polyphosphoric acids, e.g. B. pyrophosphoric acid, the Metaphosphoric acid, triphosphoric acid and the like, also calcium pyrophosphate and sodium calcium pyrophosphate, Calcium metaphosphate, magnesium pyrophosphate, calcium triphosphate and the like. As mentioned, mixed cation salts such as calcium and sodium can also be used, such as Sodium calcium pyrophosphate. These polishes generally make up the bulk of the amount, up to 99% of the solids. The toothpastes usually contain about 20 to 75%, tooth powder 70 to 95% Polishes.
The dental care products according to the invention are generally produced in the customary manner the known additives by mixing the solid components. The consistency of toothpastes should be adjusted so that that the mass can be expressed from tubes. The liquid of the pastes consists of well-known Usually from water, glycerine, sorbitol, propylene glycol or the like. As well as mixtures of these Fabrics. Mixtures of water and a moistening agent or binding agent are expediently used in a known manner, such as glycerine or sorbitol, used, the total liquid content being about 20 to 75%. You can also use gelling agents, such as natural or artificial gums and in a known manner gummy additives such as Irish moss, tragacanth, sodium carboxymethyl cellulose, polyvinylpyrrolidone, Add starch and the like, usually in amounts up to about 10%, preferably from about 0.5 to 5%.
Ammoniumphosphat zugeben. The usual additives can also be incorporated into the dentifrice according to the invention, insofar as they are used they do not significantly affect the properties of the active ingredients, such as soluble sweetener, flavor and odorous substances, such as spearmint oil, peppermint oil, wintergreen oil, as well as dyes or whitening agents, such as titanium dioxide, preservatives such as sodium benzoate, emulsifiers, silicones, alcohols, Menthol, chlorophyll compounds such as sodium-copper-chlorophyllin and the like. A wide variety of substances containing ammonia, such as urea, sec. Add ammonium phosphate and mixtures of these substances, which release ammonia in the oral cavity. For example, you can add 3% urea and 5% sec. Ammonium phosphate or of 10 to 25% urea and 5% sec. Add ammonium phosphate.
The p n value of the finished mixture can fluctuate within a relatively wide range and can be adjusted to the stability of the solution. The information about p H values relate to solutions and in toothpastes and powders 2O ° / o aqueous suspensions. Under these conditions the pH value to between about 5 and 10, preferably, be between 5.5 and 9 with the optimum being achieved in practically neutral solution with a pH value of about 6 to eighth

BeiAnimal experiments on rats found that the mixture of sodium fluoride and sodium N-lauroyl sarcoside is considerably less toxic than would be expected based on the action of the individual components was. Similar results were obtained with mixtures of stannous fluoride and sodium N-lauroyl sarcoside achieved. For these experiments three groups of albino rats were given per os with sodium fluoride and sodium lauroyl sarcoside alone and a mixture of sodium fluoride and sodium lauroyl sarcoside fed in a ratio of 1:10. In this way, the toxic doses of each of these two became Components alone as well as the mixture determined. at

Finney, »Probit Analysis«, Cambridge, 1942,The inventor also carried out statistical analysis. In doing so, he has the values according to Litchfield and Wilcoxon determined graphically (see Finney, "Probit Analysis", Cambridge, 1942,
S. 200). The lethal dose values were plotted on log paper and indicated by a line connected, which was about a straight line.
After this statistical analysis, the calculated value of LD 50 for the mixture was found to be 1010 mg / kg. The mixture of the two components is 37.5% less toxic than could be expected.
