Abstract
本发明涉及一种新型冠状病毒2019‑nCoV抗体谱检测试剂盒的生产方法,属于新型冠状病毒2019‑nCoV检测技术领域。利用本发明提供的生产方法得到的检测试剂盒包括桶装试纸条、瓶装样本稀释液、瓶装洗脱液、瓶装检测抗体、瓶装底物A、瓶装底物B、孵育槽、镊子、说明书、自封袋。本发明提供的生产方法中运用重组免疫印迹法制备检测试纸条,不同于传统的免疫印迹法,无需进行蛋白凝胶电泳和转膜,直接在硝酸纤维素膜上进行划线处理,大大简化了检测试剂盒的生产方法,缩短了制备时间,可在短时间内大批量生产检测试剂盒。

技术领域
本发明涉及一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产方法,属于新型冠状病毒2019-nCoV检测技术领域。
背景技术
免疫印迹法(Western blotting)是一种将高分辨率凝胶电泳和免疫化学分析技术相结合的杂交技术。免疫印迹法具有分析容量大、敏感度高、特异性强等优点,是检测蛋白质特性、表达与分布的一种最常用的方法,用于组织抗原的定性定量检测、多肽分子的质量测定及病毒的抗体或抗原检测等。
传统免疫印迹法分三个阶段进行。第一阶段为SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE):抗原等蛋白样品经SDS处理后带阴电荷,在聚丙烯酰胺凝胶中从阴极向原理阳极泳动,分子量越小,泳动速度就越快。此阶段分离效果肉眼不可见(只有在染色后才显出电泳区带)。第二阶段为电转移:将在凝胶中已经分离的条带转移至硝酸纤维素膜上,选用低电压和大电流,通电45min转移即可完成。此阶段分离的蛋白质条带肉眼仍不可见。第三阶段为酶免疫定位:将印有蛋白质条带的硝酸纤维素膜(相当于包被了抗原的固相载体)依次与特异性抗体和酶标第二抗体作用后,加入能形成不溶性显色物的酶反应底物,使区带染色。常用的HRP底物为3,3'-二氨基联苯胺(呈棕色)和4-氯-1-萘酚(呈蓝紫色)。阳性反应的条带清晰可辨,并可根据SDS-PAGE时加入的分子量标准,确定各组分的分子量。传统免疫印迹法综合了SDS-PAGE的高分辨力和ELISA法的高特异性和敏感性,是一个有效的分析手段,不仅广泛应用于分析抗原组分及其免疫活性,并可用于疾病的诊断。但是该方法操作繁琐、步骤多,生产工艺复杂,不易形成商品化的产品。
目前,血清抗体检测主要是免疫层析法,该方法操作简单,对设备要求低,甚至无需检测设备;检测时间短;对于检测环境要求不高,一般15分钟左右即可出结果。该方法的缺点是侧向层析方法使用1种最多2种蛋白进行检测,没有清洗过程,由于个别患者血液中含有类风湿因子、异嗜性抗体、自身抗体,以及药物和肿瘤细胞等,易造成试验的交叉反应,出现假阳性的结果。也有少部分的免疫印迹法,但是试纸条制备需要通过电泳和转膜操作,仪器和操作要求高。
因此,急需一种生产和使用方便、适合大规模的人群筛查、能避免假阴性和假阳性问题、灵敏度和特异性高、可以用于病程监控和疫苗效果评价的新型冠状病毒检测试剂盒。
发明内容
针对现有技术存在的上述问题,本发明提供了一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产方法。本发明提供的生产方法中运用重组免疫印迹法制备检测试纸条,不同于传统的免疫印迹法,无需进行蛋白凝胶电泳和转膜,直接在硝酸纤维素膜上进行划线处理,大大简化了检测试剂盒的生产方法,缩短了制备时间,同时,通过完成对各液体组分的配制、分装,包装等工艺流程完成检测试剂盒的生产。
本发明提供了一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产方法,技术方案如下:
一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产方法,所述生产方法是分别制备检测试纸条、样本稀释液、洗脱液、检测抗体和底物,将制备完成的检测试纸条、样本稀释液、洗脱液、检测抗体和底物,通过分装、组合、包装后形成检测试剂盒;其中,所述检测试纸条的制备方法包括以下步骤:
(1)将新型冠状病毒抗原和羊抗鼠IgG分别在硝酸纤维素膜上进行划线;所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组E蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白、重组E蛋白;
(2)35-39℃烘干处理1-3小时;
(3)将步骤(2)中烘干后的硝酸纤维素膜完全浸入封闭液中,15-25℃条件下,浸泡0.5-2小时;
(4)35-39℃烘干处理1-3小时;
(5)将步骤(4)中烘干后的硝酸纤维素膜固定在PVC胶板上,再在PVC胶板短边的一端贴上印有流水号的标签纸,切成3.5-4.0mm宽的试纸条,,即为新型冠状病毒2019-nCOV抗体谱检测试纸条。
进一步地,步骤(2)和步骤(4)所述烘干在湿度≤30%环境中进行。
进一步地,所述检测试纸条、样本稀释液、洗脱液、检测抗体和底物的制备过程均在10万级环境下进行。
进一步地,所述样本稀释液为pH为7-8的磷酸盐缓冲液,配制完在10万级环境下通过蠕动泵分装到样本稀释液瓶中。
进一步地,所述洗脱液为添加了吐温-20的pH为7-8的磷酸盐缓冲液,配制完在10万级环境下通过蠕动泵分装到洗脱液瓶中。
进一步地,所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗体或IgA抗体或IgE抗体,配制完在10万级环境下通过蠕动泵分装到检测抗体瓶中。
进一步地,步骤(1)划线前,利用蛋白稀释液对新型冠状病毒抗原和羊抗鼠IgG进行稀释,所述蛋白稀释液的制备方法是称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL,常温放置备用。
进一步地,步骤(3)所述封闭液的制备方法是称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;称取牛血清白蛋白10g,加纯水900mL溶解,加入NaOH溶液调整pH至7.4。吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
进一步地,所述样本稀释液的制备方法是称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL,常温放置备用。
进一步地,所述洗脱液的制备方法是称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水997.5mL溶解,分别吸取1mL proClin300和1.5mL吐温-20至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
进一步地,所述底物中含三羟甲基氨基甲烷、H2O2和3,3-二氨基联苯胺。
进一步地,所述底物的制备方法是将底物A和底物B等体积混合;所述底物A的配制方法是称取三羟甲基氨基甲烷1.21g,加纯水900mL溶解,加HCl调整pH至pH7.5。定容至1000mL,常温放置备用;所述底物B的配制方法是量取30%H2O20.1mL,称取3,3-二氨基联苯胺0.01g,定容至100mL。2-8℃条件放置备用。
进一步地,所述检测抗体的制备方法是将辣根过氧化物酶(HRP)标记的鼠抗人IgG抗体使用抗体稀释液进行稀释,使其终浓度为0.5μg/mL,混匀,2-8℃条件下放置备用;所述抗体稀释液的制备方法是称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;称取牛血清白蛋白20g,加纯水900mL溶解,加NaOH溶液调整pH至pH7.5。吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
进一步地,所述检测试纸条的宽度为3.5-4.0mm。
进一步地,所述检测试纸条中,PVC胶板长度为5-6cm,印有流水号的标签纸长度为1.1-1.2cm,硝酸纤维素膜长度为4-5cm,交互重叠部分为0.1-0.2cm,重叠部分印有流水号的标签纸在上,硝酸纤维素膜在下,质控线与相邻的检测线间隔0.9-1.1cm,各检测线间隔0.4-0.6cm。

进一步地,所述重组RBD蛋白的制备方法为:通过合成编码新型冠状病毒2019-nCOV Spike蛋白的第319位Arg到541位Phe的基因,并利用PCR的方法将合成的基因构建到小鼠IgG FC的表达载体pCMV6-c.mFC上,得到重组质粒;将重组质粒转染Hek293细胞分泌表达融合蛋白,并通过ProteinA进行亲和纯化而获取。理论Mw:52kD。
进一步地,所述重组N蛋白的制备方法为:通过合成编码新型冠状病毒2019-nCOV核衣壳蛋白完整的基因,并利用PCR的方法将合成的基因构建到pET28A-N-His的表达载体上得到重组质粒;将重组质粒转大肠杆菌,表达重组蛋白,并通过镍柱进行亲和纯化而获取。理论Mw:46.5kD。
进一步地,所述重组E蛋白的制备方法为:通过合成编码新型冠状病毒2019-nCOV包膜蛋白完整的基因,并利用PCR的方法将合成的基因构建到HIS-C-pET28A-N-GST的表达载体上。测序正确的质粒制备后转大肠杆菌,表达重组蛋白,并通过镍柱进行亲和纯化而获取。理论Mw:36.8kD。
进一步地,所述重组抗原S1蛋白、重组抗原RBD蛋白、重组抗原N蛋白、重组抗原E蛋白的划线浓度分别为0.2-0.5mg/mL(1μL/cm)、0.2-0.5mg/mL(1μL/cm)、0.1-0.2mg/mL(1μL/cm)、0.1-0.2mg/mL(1μL/cm)。
进一步地,所述羊抗鼠IgG的划线浓度为0.2-1mg/mL(1μL/cm)。
本发明有益的技术效果在于:
本发明提供的生产方法中运用重组免疫印迹法制备检测试纸条,不同于传统的免疫印迹法,无需进行蛋白凝胶电泳和转膜,直接在硝酸纤维素膜上进行划线处理,大大简化了检测试剂盒的生产方法,缩短了制备时间,可在短时间内大批量生产检测试剂盒。利用本发明提供的生产方法生产的检测试剂盒通过多种新型冠状病毒特异性重组抗原联合检测血清或血浆样本中的抗体,可用于体外定性检测人血清或血浆样本中的新型冠状病毒抗体,有效增加灵敏度和特异性,及时发现早期感染者,同时可以用于康复患者体内抗体的评价诊断进行病程监控,未来更可广泛用于疫苗人体效果的评估。本发明提供的试剂盒组分少,生产过程简单,生产成本降低,易于放大生产;检测过程便捷,易于实现全自动化。

具体实施方式
检测试纸条蛋白划线浓度选择:显色深度以“+”表示;“+++”代表最强。
检测方法:蛋白划线制成检测试纸条后,加入胶体金平台假阳样本和阳性样本、检测抗体和底物,观察条带显色强度。如表1-5所示。

室温下,样本孵育时间设置为30min、40min、1h;检测抗体孵育时间设置为30min、40min、1h;
根据不同孵育时间,检测试纸条对阳性样本的检测显色强度,确定检测孵育的适宜时间为:样本:40min、检测抗体:40min。
温度设置为4℃、22℃、37℃,孵育时间设置为样本:40min、检测抗体:40min;
根据不同温度下,检测试纸条对阳性样本的检测显色强度,确定检测孵育的适宜温度为:室温(15-25℃)。
以48人份的试剂盒为例,试剂盒中包含的试剂、耗材如表6所示。

下面结合实施例,对本发明进行具体描述。
下述实施例中的检测试纸条宽度为3.5-4.0mm,PVC胶板长度为5-6cm,印有流水号的标签纸长度为1.1-1.2cm,硝酸纤维素膜长度为4-5cm,交互重叠部分为0.1-0.2cm,重叠部分印有流水号的标签纸在上,硝酸纤维素膜在下,质控线与相邻的检测线间隔0.9-1.1cm,各检测线间隔0.4-0.6cm。
实施例1
1.检测试纸条的制备:
1.1配制蛋白稀释液和封闭液
蛋白稀释液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
封闭液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g,牛血清白蛋白10g,加纯水900mL溶解,加入NaOH溶液调整pH至7.4。吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
1.2制备检测试纸条
(1)配制质控线溶液C:将羊抗鼠IgG抗体用蛋白稀释液稀释成0.5mg/mL;
(2)配制检测线溶液T1:将重组S1蛋白用蛋白稀释液稀释成0.2mg/mL;
(3)配制检测线溶液T2:将重组RBD蛋白用蛋白稀释液稀释成0.2mg/mL;
(4)配制检测线溶液T3:将重组N蛋白用蛋白稀释液稀释成0.1mg/mL;
(5)配制检测线溶液T4:将重组E蛋白用蛋白稀释液稀释成0.1mg/mL;
(6)裁膜:将硝酸纤维素膜裁切成310mm±5mm长的膜;
(7)划线:用喷金划线仪进行划线包被,如图2所示,检测线和质控线包被的线浓度均为1μL/cm;
(8)干燥:对已包被好的硝酸纤维素膜进行烘干,温度37℃±2℃,时间120min(烘干过程在湿度≤30%环境中进行);
(9)封闭:对已干燥好的硝酸纤维素膜用封闭液进行浸泡30min;
(10)干燥:对已封闭好的硝酸纤维素膜进行再烘干,温度37℃±2℃,时间120min(烘干过程在湿度≤30%环境中进行);
(11)贴膜:将PVC胶板中间部分的表面贴膜撕掉,并贴上已经制备好的硝酸纤维素膜;
(12)利用裁切机将印有流水号的标签纸裁切成30cm×1.1cm;
(13)按图1所示,在PVC胶板短边的一端贴印有流水号的标签纸,组合好;
(14)利用斩切机将组合好的试纸板条竖切成宽度为3.5mm的试纸条。
2.样本稀释液的制备:
0.1M、pH7.4磷酸盐缓冲液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
3.洗脱液的制备:
称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水997.5mL溶解,分别吸取1mL proClin300和1.5mL吐温-20至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
4.检测抗体的制备:
将辣根过氧化物酶(HRP)标记的鼠抗人IgG抗体使用抗体稀释液进行稀释,使其终浓度为0.5μg/mL,混匀,2-8℃条件下放置备用。
5.底物的制备:
底物A的配制:称取三羟甲基氨基甲烷1.21g,加纯水900mL溶解,加HCl调整pH至pH7.5。定容至1000mL。常温放置备用。
底物B的配制:量取30%H2O20.1mL,称取3,3-二氨基联苯胺0.01g,定容至100mL。2-8℃条件放置备用。
底物制备:将底物A和底物B等体积混合。现配现用。
以上制备过程均在十万级环境下进行。
将检测试纸条、样本稀释液、检测抗体、洗脱液、底物A、底物B、孵育槽、自封袋分别按不小于某一检测次数的用量组合、包装,形成试剂盒,将镊子、说明书一同放入试剂盒中。其中,检测试纸条使用白色聚丙烯材质的桶装;所述样本稀释液、底物A和洗脱液使用白色聚丙烯材质的瓶装;所述检测抗体和底物B使用棕色聚丙烯材质的瓶装;所述说明书为纸质;所述镊子和孵育槽为聚丙烯材质;所述自封袋为聚乙烯材质。
实施例2
1.检测试纸条的制备:
1.1配制蛋白稀释液和封闭液

(7)干燥:对已包被好的硝酸纤维素膜进行烘干,温度37℃±2℃,时间90min(烘干过程在湿度≤30%环境中进行);
(8)封闭:对已干燥好的硝酸纤维素膜用封闭液进行浸泡40min;
(9)干燥:对已封闭好的硝酸纤维素膜进行再烘干,温度37℃±2℃,时间90min。(烘干过程在湿度≤30%环境中进行);
(10)贴膜:将PVC胶板中间部分的表面贴膜撕掉,并贴上已经制备好的硝酸纤维素膜;
(11)利用裁切机将印有流水号的标签纸裁切成30cm×1.1cm;
(12)在PVC胶板短边的一端贴印有流水号的标签纸,组合好;
(13)利用斩切机将组合好的试纸板条竖切成宽度为3.6mm的试纸条。
2.样本稀释液的制备:
0.1M、pH 7.4磷酸盐缓冲液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
3.洗脱液的制备:
称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水997.5mL溶解,分别吸取1mL proClin300和1.5mL吐温-20至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
4.检测抗体的制备:

本发明中的一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产工艺流程图如图5所示。
测试例
利用本发明提供的检测试剂盒检测新型冠状病毒2019-nCoV抗体的方法:
(1)润湿:在反应槽中放入检测试纸条,加入样本稀释液1mL,室温(15-25℃)条件下,混匀1min;
(2)一次孵育:加入样本(人血浆或血清)1mL,室温(15-25℃)条件下进行孵育40min;
(3)清洗:倒掉反应槽内液体,在反应槽中加入1mL洗脱液,室温(15-25℃)条件下进行孵育5min;重复清洗一次;
(4)二次孵育:倒掉反应槽内液体,在反应槽中加入1mL检测抗体,室温(15-25℃)条件下进行孵育40min;
(5)清洗:倒掉反应槽内液体,在反应槽中加入1mL洗脱液,室温(15-25℃)条件下进行孵育5min;
(6)清洗:倒掉反应槽内液体,重复清洗一次;
(7)显色:倒掉反应槽内液体,将反应槽进行控干,在反应槽中加入200μL底物,避光反应5min。
观察条带是否显色以及条带深浅。当两个及两个以上条带显色时,新型冠状病毒抗体检测结果为阳性,显色越深,抗体含量越高;当所有条带均不显色时,新型冠状病毒抗体检测结果为阴性。
对200例阴性血清样本(包含9例胶体金试剂盒检测出来的假阳性样本;包含5个乙肝,甲、乙流抗体阳性和2个乙肝抗原阳性样本等)和20例阳性血清样本进行检测,实施例1制得的检测试剂盒的预测灵敏度和特异性分别为100%、100%;使用全自动免疫分析仪在2h内就能得到200例样本的检测结果。操作过程方便、快捷、检测结果准确。实施例2和实施例3制得的检测试剂盒也能实现这些检测效果。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。

Claims (9)
1.一种新型冠状病毒2019-nCoV抗体谱检测试剂盒的生产方法,其特征在于,所述生产方法是分别制备检测试纸条、样本稀释液、洗脱液、检测抗体和底物,将制备完成的检测试纸条、样本稀释液、洗脱液、检测抗体和底物通过分装、组合、包装后形成检测试剂盒;其中,所述检测试纸条的制备方法包括以下步骤:
(1)将新型冠状病毒抗原和羊抗鼠IgG分别在硝酸纤维素膜上进行划线;所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组E蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白、重组E蛋白;
(2)35-39℃烘干处理1-3小时;
(3)将步骤(2)中烘干后的硝酸纤维素膜完全浸入封闭液中,15-25℃条件下,浸泡0.5-2小时;
(4)35-39℃烘干处理1-3小时;
(5)将步骤(4)中烘干后的硝酸纤维素膜固定在PVC胶板上,再在PVC胶板短边的一端贴上印有流水号的标签纸,切成3.5-4.0mm宽的试纸条,即为新型冠状病毒2019-nCOV抗体谱检测试纸条。
2.根据权利要求1所述的生产方法,其特征在于,步骤(2)和步骤(4)所述烘干在湿度≤30%环境中进行。
3.根据权利要求1所述的生产方法,其特征在于,所述检测试纸条、样本稀释液、洗脱液、检测抗体和底物的制备过程均在10万级环境下进行。
4.根据权利要求1或3所述的生产方法,其特征在于,所述样本稀释液为pH为7-8的磷酸盐缓冲液。
5.根据权利要求1或3所述的生产方法,其特征在于,所述洗脱液为添加了吐温-20的pH为7-8的磷酸盐缓冲液。
6.根据权利要求1或3所述的生产方法,其特征在于,所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗体或IgA抗体或IgE抗体。
7.根据权利要求1或3所述的生产方法,其特征在于,所述底物中含三羟甲基氨基甲烷、H2O2和3,3-二氨基联苯胺。
8.根据权利要求1或3所述的生产方法,其特征在于,所述检测试纸条的宽度为3.5-4.0mm。
9.根据权利要求1或3所述的生产方法,其特征在于,步骤(1)中,所述重组抗原S1蛋白、重组抗原RBD蛋白、重组抗原N蛋白、重组抗原E蛋白的划线浓度分别为0.2-0.5mg/mL、0.2-0.5mg/mL、0.1-0.2mg/mL、0.1-0.2mg/mL,所述羊抗鼠IgG的划线浓度为0.2-1mg/mL。
