Abstract
本发明涉及一种多蛋白联检组合检测新型冠状病毒2019‑nCoV的方法,属于新型冠状病毒2019‑nCoV检测技术领域。本发明提供了一种多蛋白联检组合检测新型冠状病毒2019‑nCoV的方法,是将高纯度的、特异性的新型冠状病毒抗原直接在硝酸纤维素膜上进行划线,制备用于检测新型冠状病毒2019‑nCoV抗体的检测试纸条,能利用多种抗原同时对样本中IgG、IgM和/或IgA抗体进行检测。本发明通过3种或4种特异性抗原联检,同时对抗原的蛋白序列也进行了优化设计,有效增加了灵敏度和特异性,有效避免了假阴性和假阳性的问题。
Description
技术领域
本发明涉及一种多蛋白联检组合检测新型冠状病毒2019-nCoV的方法,属于新型冠状病 毒2019-nCoV检测技术领域。
背景技术
冠状病毒是一个大型病毒家族,已知可引起感冒以及中东呼吸综合征(MERS)和严重 急性呼吸综合征(SARS)等较严重的疾病。新型冠状病毒是以前从未在人体中发现的冠状病 毒新毒株,世界卫生组织将这种冠状病毒引起的疾病命名为2019年冠状病毒病(COVID-19), 致死性病毒严重急性呼吸综合征冠状病毒2型(SARS-CoV-2)。
冠状病毒基因组依次编码棘突蛋白(spike protein)、包膜蛋白(Envelopeprotein,E蛋 白)、膜蛋白(Membrane protein)和核衣壳蛋白(Nucleocapsid,N蛋白)。棘突蛋白(spike protein)是冠状病毒最重要的表面膜蛋白,含有两个亚基(subunit),S1和S2。其中S1主 要包含有受体结合区(receptorbinding domain,RBD蛋白),负责识别细胞的受体。S2含有 膜融合过程所需的基元件。棘突蛋白承担病毒与宿主细胞膜受体结合及膜融合功能,是宿主 中和抗体的重要作用位点以及疫苗设计的关键靶点。SARS-CoV-2(2019-nCoV)的棘突蛋白 与人ACE2互作来感染人的呼吸道上皮细胞。核衣壳蛋白是冠状病毒中含量最丰富的蛋白。 在病毒体组装过程中,N蛋白与病毒RNA结合并导致螺旋核衣壳的形成。核衣壳蛋白是一种 高度免疫原性的磷蛋白,与病毒基因组复制和调节细胞信号通路有关。
目前,血清抗体检测主要是免疫层析法,该方法操作简单,对设备要求低,甚至无需检 测设备;检测时间短;对于检测环境要求不高,一般15分钟左右即可出结果。该方法的缺点 是侧向层析方法使用1种、最多2种蛋白进行检测,没有清洗过程,由于个别患者血液中含 有类风湿因子、异嗜性抗体、自身抗体,以及药物和肿瘤细胞等,易造成试验的交叉反应, 出现假阳性的结果。
因此,急需一种对检测环境要求不高、适合大规模的人群筛查、能基本避免假阴性和假 阳性问题、灵敏度和特异性高、可以用于病程监控和疫苗效果评价的新型冠状病毒检测方法。
发明内容
针对现有技术存在的上述问题,本发明提供了一种多蛋白联检组合检测新型冠状病毒 2019-nCoV的方法。本发明通过3种或4种特异性抗原联检,同时对抗原的蛋白序列也进行 了优化设计,多种蛋白相互验证、使用免疫印迹法改变抗体结合方式(增加清洗、改变抗体 标记物)有效增加了灵敏度和特异性,能同时检测病毒中多种特异性抗原,有效避免了假阴 性和假阳性的问题。
本发明的技术方案如下:
一种多蛋白联检组合检测新型冠状病毒2019-nCoV的方法,所述方法是利用检测试纸条 对人血浆或血清样本进行检测,检测时,将检测试纸条浸入人血浆或血清样本中进行一次孵 育;清洗;再加入检测抗体进行二次孵育;再清洗;进行显色反应;所述检测试纸条联合使 用3种或4种新型冠状病毒抗原对样本中的IgG、IgM和/或IgA抗体进行定性检测;所述检 测试纸条是以硝酸纤维素膜为固相载体,在硝酸纤维素膜上划3条或4条检测线(T线)和1 条质控线(C线)制得的;所述检测线上固定有新型冠状病毒抗原;所述新型冠状病毒抗原 为重组S1蛋白、重组RBD蛋白、重组N蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、 重组RBD蛋白、重组E蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、 重组N蛋白、重组E蛋白;所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗 体或IgA抗体或IgE抗体。
进一步地,所述重组S1蛋白的氨基酸序列如SEQ ID NO.1所示。
进一步地,所述重组RBD蛋白的氨基酸序列如SEQ ID NO.2所示。
进一步地,所述重组N蛋白的氨基酸序列如SEQ ID NO.3所示。
进一步地,所述重组E蛋白的氨基酸序列如SEQ ID NO.4所示。
进一步地,所述质控线上固定有羊抗鼠IgG。
进一步地,所述方法具体包括以下步骤:
(1)润湿:在反应槽中放入检测试纸条,加入样本稀释液,15-25℃条件下,混匀1-2min; 所述样本稀释液为pH为7-8的磷酸盐缓冲液;
(2)一次孵育:加入人血清或血浆样本,15-25℃条件下进行孵育30-60min;
(3)清洗:倒掉反应槽内液体,在反应槽中加入洗脱液,15-25℃条件下进行孵育4-6min; 重复清洗一次;
(4)二次孵育:倒掉反应槽内液体,在反应槽中加入检测抗体,15-25℃条件下进行孵 育30-60min;所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗体或IgA抗体或IgE抗体;
(5)清洗:倒掉反应槽内液体,在反应槽中加入洗脱液,15-25℃条件下进行孵育4-6min;
(6)清洗:倒掉反应槽内液体,重复清洗一次;
(7)显色:倒掉反应槽内液体,将反应槽进行控干,在反应槽中加入底物,避光反应2-10min;若有两条以上的条带显色,则新型冠状病毒抗体检测结果为阳性;若无条带显色,则新型冠状病毒抗体检测结果为阴性。
进一步地,所述重组抗原S1蛋白、重组抗原RBD蛋白、重组抗原N蛋白、重组抗原E蛋白的划线浓度分别为0.2-0.5mg/mL(1μL/cm)、0.2-0.5mg/mL(1μL/cm)、0.1-0.2mg/mL (1μL/cm)、0.1-0.2mg/mL(1μL/cm)。
进一步地,所述羊抗鼠IgG的划线浓度为0.2-1mg/mL(1μL/cm)。

具体实施方式
样本稀释液的配制:0.1M、pH7.4磷酸盐缓冲液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g, 氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。 常温放置备用。
洗脱液的配制:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水997.5mL 溶解,分别吸取1mL proClin300和1.5mL吐温-20至溶液中,搅拌混匀,定容至1000mL。常 温放置备用。
底物A的配制:称取三羟甲基氨基甲烷1.21g,加纯水900mL溶解,加HCl调整pH至pH7.5。定容至1000mL。常温放置备用。
底物B的配制:量取30%H2O20.1mL,称取3,3-二氨基联苯胺0.01g,定容至100mL。2-8℃ 条件放置备用。
底物制备:将底物A和底物B等体积混合。现配现用。
抗体稀释液制备:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;称取牛血清白 蛋白20g,加纯水900mL溶解,加NaOH溶液调整pH至pH7.5。吸取1mL proClin300至溶 液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
检测抗体制备方法:
将辣根过氧化物酶(HRP)标记的鼠抗人IgG抗体使用抗体稀释液进行稀释,使其终浓 度为0.5μg/mL,混匀,2-8℃条件下放置备用。
重组S1蛋白制备:通过合成编码新型冠状病毒2019-nCOVSpike蛋白的全长基因,并利 用PCR的方法将合成的基因构建到小鼠IgG FC的表达载体pCMV6-c.mFC上,得到重组质 粒;将重组质粒转染Hek293细胞分泌表达融合蛋白,并通过ProteinA进行亲和纯化而获取。 理论Mw:76.5kD。
重组RBD蛋白制备:通过合成编码新型冠状病毒2019-nCOV Spike蛋白的第319位Arg 到541位Phe的基因,并利用PCR的方法将合成的基因构建到小鼠IgG FC的表达载体pCMV6-c.mFC上,得到重组质粒;将重组质粒转染Hek293细胞分泌表达融合蛋白,并通过ProteinA进行亲和纯化而获取。理论Mw:52kD。
重组N蛋白制备:通过合成编码新型冠状病毒2019-nCOV核衣壳蛋白完整的基因,并 利用PCR的方法将合成的基因构建到pET28A-N-His的表达载体上得到重组质粒;将重组质 粒转大肠杆菌,表达重组蛋白,并通过镍柱进行亲和纯化而获取。理论Mw:46.5kD。
重组E蛋白制备:通过合成编码新型冠状病毒2019-nCOV包膜蛋白完整的基因,并利 用PCR的方法将合成的基因构建到HIS-C-pET28A-N-GST的表达载体上。测序正确的质粒制备后转大肠杆菌,表达重组蛋白,并通过镍柱进行亲和纯化而获取。理论Mw:36.8kD。
检测试纸条上蛋白划线浓度选择:显色深度以“+”表示;“+++”代表最强。
检测方法:蛋白划线制成检测试纸条后,加入胶体金平台假阳样本和阳性样本、检测抗 体和底物,观察条带显色强度。如表1-5所示。

确定划线浓度如下:
0.2-1mg/mL(1μL/cm)的羊抗鼠IgG抗体;
0.2-0.5mg/mL(1μL/cm)的重组S1蛋白;
0.2-0.5mg/mL(1μL/cm)的重组RBD蛋白;
0.1-0.2mg/mL(1μL/cm)的重组N蛋白;
0.1-0.2mg/mL(1μL/cm)的重组E蛋白。
检测时间和温度的选择:
室温下,样本孵育时间设置为30min、40min、1h;检测抗体孵育时间设置为30min、40min、1h;
根据不同孵育时间,检测试纸条对阳性样本的检测显色强度,确定检测孵育的适宜时间 为:样本:40min、检测抗体:40min。
温度设置为4℃、22℃、37℃,孵育时间设置为样本:40min、检测抗体:40min;
根据不同温度下,检测试纸条对阳性样本的检测显色强度,确定检测孵育的适宜温度为: 室温(15-25℃)。
下面结合实施例,对本发明进行具体描述。
下述实施例中使用的检测试纸条宽度为3.5-4.0mm,PVC胶板长度为5-6cm,印有流水 号的标签纸长度为1.1-1.2cm,硝酸纤维素膜长度为4-5cm,交互重叠部分为0.1-0.2cm,重 叠部分印有流水号的标签纸在上,硝酸纤维素膜在下,质控线与相邻的检测线间隔0.9-1.1cm, 各检测线间隔0.4-0.6cm。
实施例1
检测试纸条的制备:
1、试剂制备:
1.1蛋白稀释液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
1.2封闭液的配制:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g,牛血清白蛋白 10g,加纯水900mL溶解,加入NaOH溶液调整pH至7.4。吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
2、制备步骤:
2.1硝酸纤维素膜裁成长宽为31cm×5cm大小,将4种高纯度的特异性的重组S1蛋白、 重组RBD蛋白、重组N蛋白、重组E蛋白分别在硝酸纤维素膜上进行划线,得到4条检测线(划线浓度分别为0.2mg/mL、0.2mg/mL、0.1mg/mL、0.1mg/mL,划线量均为1μL/cm), 如图1和图2所示(各检测线在硝酸纤维素膜上的分布次序对检测结果不产生显著影响);
2.2将羊抗鼠IgG在硝酸纤维素膜上进行划线(质控线,C线;划线浓度为0.5mg/mL,划线量为1μL/cm);
2.3 37℃进行烘干2小时;
2.4将烘干后的硝酸纤维素膜完全浸入封闭液中,室温(15-25℃)条件下,完全浸泡1 小时;
2.5 37℃进行烘干2小时;
2.6将第二次烘干后的硝酸纤维素膜和印有流水号的合成纸依次粘贴在PVC胶板上,切 成3.5-4.0mm宽度,即为新型冠状病毒2019-nCOV抗体谱检测试纸条。
利用上述新型冠状病毒2019-nCOV抗体谱检测试纸条检测新型冠状病毒2019-nCoV抗 体的方法:
(1)润湿:在反应槽中放入检测试纸条,加入样本稀释液1mL,室温(15-25℃)条件下,混匀1min;
(2)一次孵育:加入样本(人血浆或血清)1mL,室温(15-25℃)条件下进行孵育40min;

实施例2
检测试纸条的制备:
1、试剂制备:
1.1蛋白稀释液:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;加纯水999mL溶解,吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。常温放置备用。
1.2封闭液的配制:称取磷酸氢二钠55.8g,磷酸二氢钠5.5g,氯化钠9g;称取牛血清白 蛋白10g,加纯水900mL溶解,加入NaOH溶液调整pH至7.4。吸取1mL proClin300至溶液中,搅拌混匀,定容至1000mL。2-8℃条件下放置备用。
2、制备步骤:
2.1硝酸纤维素膜裁成长宽为31cm×5cm大小,将3种高纯度的特异性的重组S1蛋白、 重组RBD蛋白、重组N蛋白分别在硝酸纤维素膜上进行划线,得到3条检测线(划线浓度分别为0.3mg/mL、0.3mg/mL、0.2mg/mL,划线量均为1μL/cm),如图3所示(各检测线在 硝酸纤维素膜上的分布次序对检测结果不产生显著影响);
2.2将羊抗鼠IgG在硝酸纤维素膜上进行划线(质控线,C线;划线浓度为0.2mg/mL,划线量为1μL/cm);
2.3 37℃进行烘干2小时;
2.4将烘干后的硝酸纤维素膜完全浸入封闭液中,室温(15-25℃)条件下,完全浸泡1 小时;
2.5 37℃进行烘干2小时;
2.6将第二次烘干后的硝酸纤维素膜和印有流水号的合成纸依次粘贴在PVC胶板上,切 成3.5-4.0mm宽度,即为新型冠状病毒2019-nCOV抗体谱检测试纸条。
利用上述新型冠状病毒2019-nCOV抗体谱检测试纸条检测新型冠状病毒2019-nCoV抗 体的方法:
(1)润湿:在反应槽中放入检测试纸条,加入样本稀释液1mL,室温(15-25℃)条件下,混匀90s;
(2)一次孵育:加入样本1mL,室温(15-25℃)条件下进行孵育50min;
(3)清洗:倒掉反应槽内液体,在反应槽中加入1mL洗脱液,室温(15-25℃)条件下进行孵育4min;重复清洗一次;
(4)二次孵育:倒掉反应槽内液体,在反应槽中加入1mL检测抗体,室温(15-25℃)条件下进行孵育50min;

实施例3
检测试纸条的制备:
1、试剂制备:

利用上述新型冠状病毒2019-nCOV抗体谱检测试纸条检测新型冠状病毒2019-nCoV抗 体的方法:

测试例
对200例阴性血清样本(包含9例胶体金试剂盒检测出来的假阳性样本;包含5个乙肝, 甲、乙流抗体阳性和2个乙肝抗原阳性样本等)和20例阳性血清样本进行检测,实施例1制 得的检测试纸条的预测灵敏度和特异性分别为100%、100%;使用全自动免疫分析仪在2h 内就能得到200例样本的检测结果。操作过程方便、快捷、检测结果准确。实施例2和实施例3制得的检测试纸条也能实现这些检测效果。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人, 在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以 权利要求书所界定的为准。

1.一种多蛋白联检组合检测新型冠状病毒2019-nCoV的方法,其特征在于,所述方法是利用检测试纸条对人血浆或血清样本进行检测,检测时,将检测试纸条浸入人血浆或血清样本中进行一次孵育;清洗;再加入检测抗体进行二次孵育;再清洗;进行显色反应;所述检测试纸条联合使用3种或4种新型冠状病毒抗原对样本中的IgG、IgM和/或IgA抗体进行定性检测;所述检测试纸条是以硝酸纤维素膜为固相载体,在硝酸纤维素膜上划3条或4条检测线和1条质控线制得的;所述检测线上固定有新型冠状病毒抗原;所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组E蛋白,或,所述新型冠状病毒抗原为重组S1蛋白、重组RBD蛋白、重组N蛋白、重组E蛋白;所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗体或IgA抗体或IgE抗体。
2.根据权利要求1所述的方法,其特征在于,所述重组S1蛋白的氨基酸序列如SEQ IDNO.1所示。
3.根据权利要求1所述的方法,其特征在于,所述重组RBD蛋白的氨基酸序列如SEQ IDNO.2所示。
4.根据权利要求1所述的方法,其特征在于,所述重组N蛋白的氨基酸序列如SEQ IDNO.3所示。
5.根据权利要求1所述的方法,其特征在于,所述重组E蛋白的氨基酸序列如SEQ IDNO.4所示。
6.根据权利要求1所述的方法,其特征在于,所述质控线上固定有羊抗鼠IgG。
7.根据权利要求1所述的方法,其特征在于,所述方法具体包括以下步骤:
(1)润湿:在反应槽中放入检测试纸条,加入样本稀释液,15-25℃条件下,混匀1-2min;所述样本稀释液为pH为7-8的磷酸盐缓冲液;
(2)一次孵育:加入人血清或血浆样本,15-25℃条件下进行孵育30-60min;
(3)清洗:倒掉反应槽内液体,在反应槽中加入洗脱液,15-25℃条件下进行孵育4-6min;重复清洗一次;
(4)二次孵育:倒掉反应槽内液体,在反应槽中加入检测抗体,15-25℃条件下进行孵育30-60min;所述检测抗体为辣根过氧化物酶标记的鼠抗人IgG抗体或IgM抗体或IgA抗体或IgE抗体;
(5)清洗:倒掉反应槽内液体,在反应槽中加入洗脱液,15-25℃条件下进行孵育4-6min;
(6)清洗:倒掉反应槽内液体,重复清洗一次;
(7)显色:倒掉反应槽内液体,将反应槽进行控干,在反应槽中加入底物,避光反应2-10min;若有两条以上的条带显色,则新型冠状病毒抗体检测结果为阳性;若无条带显色,则新型冠状病毒抗体检测结果为阴性。
