本发明属于微生物技术领域,具体涉及一株运动芽孢杆菌及其应用。
背景技术
随着人们对禽畜源食品安全的重视程度越来越高,我国不断加大饲料生产和禽畜类动物绿色规范饲养监管力度,实施畜禽养殖业的饲料安全工程。自2020年7月起,饲料和动保企业都寻找和开发替抗产品,其中,饲用益生菌是替抗产品中应用最广泛、极具发展潜力的添加剂之一。益生菌被定义为一类具有调节肠道菌群平衡,增强免疫,促进营养吸收功能的活性微生物制剂。乳杆菌、双歧杆菌以及酵母类是目前研究最多的益生菌种类,但因其存在耐热性差、耐酸性差、存活率低、抑菌性差、不耐加工等缺陷,大大降低了在禽畜生产中的应用效果。
发明专利CN114591855A公开了一株运动芽孢杆菌Bacillus mobilis在增强油菜耐盐促生中的应用;CN114317314A公开了一种当归根腐病生物防治剂、制备方法及应用;CN112760270A公开了一种运动芽孢杆菌及其应用,解离煤矸石来制备微生物肥料的方法。
因此,寻找开发抗逆性好、益生性强的新菌种成为近年来益生菌替抗应用研究的热点。目前,研究人员已经从池塘底泥、土壤等自然环境和健康家禽肠道内分离出大量具有益生性的菌株,经鉴定这些菌株大多数为乳酸菌和芽孢杆菌等。鲜有以海鲈鱼为载体进行益生菌分离并应用的研究报道。

1.1材料与仪器
日本花鲈成年大鱼珠海市斗门区河口渔业研究所赠予,低温条件下,充氧活体运输至实验室,解剖时体表无外伤、鳞片完好、肠道健康,无明显未消化食靡。

1.2实验方法
1.2.1肠道样品中细菌的分离与初筛
在超净工作台中取出花鲈鱼肠道,分离内容物于离心管中,加入10mL的无菌生理盐水充分振荡摇匀,制成原液备用。梯度稀释原液至10-1、10-2、10-3、10-4、10-5、10-6六个稀释梯度,分别涂布于添加了溴甲酚紫的MRS固体培养基中,30℃培养48h,挑取变色的单菌落,再次划线培养4次直至纯化,取单菌落保存。分离单菌落于MRS肉汤培养基中扩大培养,30℃培养24h,发酵液离心(8000r/min,30min)后弃沉淀取上清,经0.22μm微孔滤膜过滤,得到上清液,4℃保存备用。
在BHI液体培养基中分别接种大肠杆菌(ATCC25922)和金黄色葡萄球菌(ATCC25923)作为指示菌,30℃培养12h,取1mL发酵液稀释103倍至工作浓度,取100μL稀释后的菌液均匀涂布在PCA平板上,将牛津杯均匀放置在平板上并轻轻按压,保证与平板培养基接触紧密,然后分别吸取200μL分离菌株发酵液加入杯中,30℃培养18h,观察并测量抑菌圈直径,计算并挑选抑菌圈最大的分离菌株进行后续试验研究。
1.2.2分离菌株的复筛
针对上述分离菌株分别进行有机酸、过氧化氢和蛋白类抑菌物质的甄别筛选。①配置与发酵液pH值相同的乳酸和乙酸溶液分别进行抑菌实验,重复3次。观察并测量抑菌圈直径的大小。②用3mol/L NaOH和3mol/L HCl溶液调节发酵液pH值与氧化氢酶最适的pH值一致,然后添加过氧化氢酶至终质量浓度为1g/L,37℃孵育1h,再将pH值调回原始pH值进行牛津杯抑菌实验,重复3次,观察并测量抑菌圈直径的大小。③与②同样操作过程进行胰蛋白酶、胃蛋白酶、蛋白酶K的甄别筛选实验,记录实验结果。
1.2.3分离菌株的鉴定
1)生理生化鉴定
依据《伯杰氏细菌鉴定手册》与《常见细菌系统鉴定手册》结合生化鉴定管对菌株进行相应生化指标的测定。
2)16S rDNA序列分析
取对数生长期分离菌株发酵液经试剂盒操作提取基因组DNA,以基因组DNA为模板,使用细菌16S rDNA的通用引物(27F和1492R)进行PCR扩增,扩增产物验证后测序、比对、建树。
1.2.4生长曲线及pH曲线
采用比浊法测定生长曲线和pH曲线。将分离菌株以1%接种量接种到MRS液体培养基中,37℃恒温震荡培养(200r·min-1),每隔2h取样1次,测定600nm处吸光度值及pH。以OD600—培养时间绘制生长曲线;以pH—培养时间绘制pH曲线。
1.2.5抑菌率的测定
采用比浊法测定分离菌株的抑菌率,利用大肠杆菌、金黄色葡萄球菌和肠炎沙门氏菌常见菌株(详见3.1)作为指示菌。菌种活化后,1%接种量接种到100mLMRS液体培养基中,37℃培养24h,8000r·min-1离心5min,去沉淀留上清备用。调整指示菌菌体浓度约为0.5OD。按照实验组:4mL指示菌液+1mL实验菌液,对照组:4mL指示菌液+1mL生理盐水配置混合菌液,置于37℃共培养3h(200r·min-1),观察菌体的浑浊情况,同时测定所有试管菌体的OD600计算抑菌率。
抑菌率=(%)=[(A空-A空0)-(A样-A样0)]/(A空-A空0)×100%
1.2.6抗生素耐受性测定
采用纸片扩散法对筛选获得的菌株对常见抗生素的耐受性实验。菌种活化,于MRS液体培养基中接种,37℃震荡培养24h,调整菌液浓度为2×108cfu·mL-1,涂布平板。贴上药敏纸片,正置于37℃恒温培养24h,测量抑菌圈直径。
1.2.7产酸、产酶能力鉴定
取5mL菌体发酵液,8000r·min-1离心5min,去沉淀留上清,加上清液9倍体积生理盐稀释为待测液。按照操作说明书配置试剂并在规定波长下测定吸光值,带入公式计算样本浓度,测定的指标包括:乳酸、淀粉酶、蛋白酶、乳糖酶和纤维素酶。
1.2.8耐酸、耐胆盐能力测定
取5mL菌体发酵液,8000r·min-1离心10min,弃上清,留沉淀,PBS重悬洗涤后备用;取5mL重悬液加入45ml无菌人工合成胃液混匀,分装于3个培养管中,37℃培养分别培养0min、30min、120min后,取100μL分别用活菌平板倾注计数法检测活菌数量;MRS液体培养基中分别加入0.00%、0.03%、0.05%、0.1%、0.2%、0.3%的胆盐,按照10%(v/v)的接种量接入菌液,37℃静置培养24h,逐级梯度稀释,取合适浓度菌液100μL,进行平板倾注计数,计算耐胆盐能力。
1.2.9体内益生性评价
a.分离菌株菌粉的制备
将运动芽孢杆菌MH22活化,菌种制备后,进发酵罐发酵。所得发酵液用高速冷冻离心机4℃,4000r/min离心10min,弃上清得菌泥,用5%蔗糖、1%谷氨酸钠和1%抗坏血酸的水溶液搅拌均匀,再加入50%(v/v)脱脂奶粉,置于-80℃超低温冰箱冷冻,再真空干燥24h,取出粉碎,过60目筛,可得到活菌数为109CFU/g左右的菌粉。
b.分组设计
360只1日龄体重无差异的健康AA肉仔公鸡,随机分为4个处理组(空白对照组、108CFU/kg组、109CFU/kg组和抗生素组),每个处理组6个重复,每个重复15只鸡;空白对照组,饲喂基础日粮,抗生素组在基础日粮中添加5mg·kg-1黄霉素、20mg·kg-1吉他霉素和75mg·kg-1金霉素。试验共进行42日,分别为1-21日龄期和22-42日龄期。基础日粮配方参考(NY/T33-2004)《鸡饲养标准》设计,日粮配方及营养成分见表1。

1.维生素和微量元素预混剂为每千克饲粮提供:维生素A10000IU,维生素D32000IU维生素E10IU维生素K32.5mg,维生素B11mg,维生素B26mg,维生素B310mg,维生素B540mg,维生素B63mg,维生素B110.3mg,维生素B120.01mg,生物素0.12mg,铜(硫酸铜)8mg,铁(硫酸亚铁)80mg,锰(硫酸锰)60mg,锌(硫酸锌)40mg,硒(亚硒酸钠)0.15mg,碘(碘化钾)0.35mg。
2.营养水平粗蛋白为实测值,其他为计算值。
c.饲养管理
本试验采用全封闭式鸡舍,养鸡前做好鸡舍消毒工作。鸡舍光照、温度、相对湿度均按照《爱拔益加AA肉鸡养殖手册》进行。免疫按常规程序进行,自由饮水、自由采食,人工光照18h,夜间定期关灯6h,以满足动物福利的要求。
d指标测定
分别于肉鸡1、21和42日龄,以重复为单位称重(停料12h、不停水),统计饲料总采食量,计算肉鸡平均日增重、平均日采食量和料重比。
在肉鸡21和42日龄时,采用日立7600全自动生化仪测定血清中总蛋白(TP)、总胆固醇(TC)、甘油三酯(TG)、尿酸(GA)、血氨(BA)、含量以及碱性磷酸酶(ALP)活性。采用原子吸收光谱法测定血清中钙(Ca)含量。采用钼酸铵-偏钒酸铵比色法测定血清中磷(P)含量。
在肉鸡21和42日龄时,每个重复选取体重接近平均体重的1只鸡,取胫骨,去肌肉和筋腱组织后风干,使用CMT4504型微机控制电子式万能试验机对左侧胫骨进行3点弯曲试验。记录胫骨断裂强度。胫骨强度以牛顿(kg)表示。
取右侧胫骨夹碎置于脂肪包中(无损失),经脱脂、风干、烘干,灰化后,称量并记录灰分重;然后测定胫骨灰分中钙、磷含量。
e试验数据统计分析
采用SPSS21.0软件中的独立样本T检验进行数据分析。以P<0.05作为差异显著的判断标准,试验结果均表示为“平均值±标准差”。
2结果与分析
2.1乳酸菌的分离与初筛
通过MRS平板(溴甲酚紫)初筛,从日本花鲈的肠道样本中分离到318株能产酸的菌株,绝大多数为芽孢菌,将初筛得到的菌株;通过牛津杯抑菌法复筛证实有11株菌对大肠杆菌(ATCC25922)和金黄色葡萄球菌(ATCC25923)都具有较强抑制作用,分别命名MH22、BC-2、BC-3、BC-4、BC-5、BC-6、BC-7、BC-8、BC-9、MH220和MH221。
2.2乳酸菌的复筛
通过同pH值的乳酸和乙酸溶液模拟干扰物抑制和过氧化氢酶处理排除有机酸、H2O2、细菌素等干扰物,复筛验证确定MH22(表1)、BC-6(表2)和BC-9(表3)菌株为代谢产物具有抑菌作用3株产酸芽孢菌菌株。
由表2、3和4可见,与MH22、BC-6和BC-9发酵液同pH值的乙酸和乳酸溶液均没有抑菌效果,由此可推断有机酸等酸性物质不是MH22、BC-6和BC-9发酵液中的抑菌活性物质;MH22菌株的发酵液经胰蛋白酶和胃蛋白酶处理后,抑菌效果未减弱;H2O2酶处理后,抑菌效果下降10%左右;经蛋白酶K处理后,失去抑菌效果;由此可以推测MH22菌株发酵液中的抑菌活性物质不是过氧化氢类物质而是蛋白类物质且不被胰蛋白酶和胃蛋白酶降解。BC-6菌株的发酵液经H2O2酶、胰蛋白酶、胃蛋白酶和蛋白酶K处理之后,抑菌圈均由明显减小,由此可以推断BC-6菌株的抑菌物质成分较复杂,存在H2O2和蛋白类物质,易受环境影响,不适合作为备选菌株。BC-9菌株的发酵液也存在同样问题,且抑菌圈的大小较MH22更小,据此,选用MH22菌株为后续研究所用菌株。

2.3菌株的鉴定
2.3.1形态学鉴定
菌株MH22的菌落大小在3.0~4.0mm左右(图1A),菌落呈乳白色、扁平状,光滑,凸起,表面光亮、干燥,颜色一致;共聚焦显微镜下在菌体内或端侧能观察出形如豌豆样半透明的芽孢(图1B);扫描电镜下菌体外观平整,形如短棒,长度8~15μm(图1C)。
2.3.2生理生化鉴定
采用乳酸菌专用生化鉴定试剂参照说明书操作对MH22菌株进行生化鉴定,详细结果见表5。

根据表5生理生化鉴定结果可知,菌株MH22符合芽孢杆菌属细菌特征;能够水解利用七叶苷;初步判定该菌株为肠球菌;且在7% NaCl、55℃条件下能生长。上述结果与《伯杰细菌鉴定手册》和《常见细菌鉴定手册》中关于运动芽孢杆菌的分析描述相符合,初步鉴定为运动芽孢杆菌。
2.3.3 16S rDNA序列分子鉴定
对MH22菌株进行16S rDNA序列扩增,其16S rDNA核苷酸序列如SEQ ID NO:1所示:

将SEQ ID NO:1所示的核苷酸序列与GenBank中的16S rDNA序列进行比对,并用MEGA11.0软件构建系统进化发育树(图2),显示该菌株与运动芽孢杆菌(Bacillustoyonensis)在同一分支上,结合生理生化特性,将菌株MH22进一步鉴定为运动芽孢杆菌(Bacillus toyonensis)MH22,将该菌株在2023年5月5保藏于广东省微生物菌种保藏中心,保藏号为GDMCC No:63422,保藏地址:广州市先烈中路100号大院59号楼5楼、广东省科学院微生物研究所。
2.4生物学特性
2.4.1生长曲线的测定
测定菌株MH22发酵液在24小时内的生长曲线(图3A)和pH曲线(图3B),在一个生长周期内,菌株在0~6h内生长缓慢,发酵液pH值缓慢下降,在6~16h时间内处于生长对数期,发酵液pH值极剧下降,16~24h处于生长平台期,发酵液pH值趋于平稳,24h菌体生长量达到最高,pH值达到最低值4.3左右。
2.4.2菌株MH22药敏性实验
根据抑菌圈直径测量评判标准判定,药敏性试验结果(表6)表明,MH22对新霉素耐药,对红霉素处于中间敏感程度,对其余20种常见抗生素均高度敏感。

2.4.3菌株MH22的抑菌实验
菌株MH22发酵液对大肠杆菌、金黄色葡萄球菌和肠炎沙门氏菌常见菌株的抑菌率均达到90%以上(表7),说明MH22发酵液中的抑菌物质对此3种9株致病菌有抑制作用。

2.4.4菌株MH22的产乳酸能力
菌株MH22具有一定的产乳酸能力,同等条件下,与质控标准菌株植物乳杆菌Lactobacillus plantarum ATCC14917相比,产乳酸量约为植物乳杆菌产乳算量的45%(表8)。

2.4.5菌株MH22的产酶能力
菌株MH22产淀粉酶、蛋白酶和乳糖酶,不产纤维素酶;同等条件下,蛋白酶产酶量高于植物乳杆菌ATCC14917(P<0.05),淀粉酶和乳糖酶产酶量无差异(表9)。

2.4.6菌株MH22的模拟胃液实验
由表10所示,菌株MH22在pH 2.0的模拟胃液中能够保存120min,在60min、90min、120min相对存活率分别为69.57%、59.10%和57.65%(表10)。通常食物在动物胃内停留时间少于120min,该菌株能够以有效活菌数量状态经胃进入肠道继续发挥作用。

2.4.7菌株MH22的胆盐耐受实验
由表11所示,菌株MH22在0.03%~0.3%胆盐浓度下均能存活,在0.3%胆盐浓度下存活率为33.15%,有效菌体数量能维持在108数量级,陈晓春等从猪场土样中分离到的运动芽孢杆菌C11对猪胆盐的最高耐受为0.25%,说明菌株MH22耐受度优于C11,能够在肠道内定植并发挥作用。

2.5益生性评价
2.5.1饲粮菌株MH22对肉鸡生长性能的影响
由表12所示,1~21日龄时,与对照组相比,饲粮菌株MH22对肉仔鸡终末体重和平均日增重无影响(P>0.05);饲粮菌株MH22组和抗生素组肉仔鸡平均日采食量和料重比均降低(P<0.05),饲粮菌株MH22低剂量组肉仔鸡平均日采食量低于MH22高剂量组和抗生素组肉仔鸡平均日采食量(P<0.05)。22~42日龄时,与对照组相比,饲粮菌株MH22组肉仔鸡平均日增重和平均日采食量显著提高(P<0.05),料重比显著降低(P<0.05)。饲粮菌株MH22组和抗生素组之间终末体重及料重比无明显差异(P>0.05)。

2.5.2饲粮菌株MH22对肉鸡血脂、钙和磷的影响
由表13所示,1~21日龄时,与对照组相比,饲粮菌株MH22组肉仔鸡血清中总胆固醇含量显著增加(P<0.05),碱性磷酸酶的活性增强(P<0.05),血清甘油三酯的含量显著降低(P<0.05);另外,饲粮菌株MH22组肉仔鸡血清中钙和磷含量显著提高(P<0.05),各浓度组无显著差异(P>0.05)。抗生素组总胆固醇含量和甘油三酯含量均高于菌株MH22处理组(P<0.05),与对照组也具有显著差异(P<0.05),但血清中钙和磷含量与对照组无明显差异(P>0.05)。22~42日龄时,饲粮菌株MH22对肉鸡血清中总胆固醇和甘油三酯的含量无显著影响(P>0.05);与对照组相比,饲粮菌株MH22组肉仔鸡血清中钙和磷含量显著提高(P<0.05),碱性磷酸酶的活性增强(P<0.05),但总体活性值较1~21日龄时低。

2.5.3饲粮菌株MH22对肉鸡胫骨的影响
由表14所示,1~21日龄时,与对照组相比,饲粮菌株MH22组肉仔鸡胫骨强度显著增强(P<0.05),胫骨钙含量和磷含量显著增加(P<0.05),高剂量处理组效果好于低剂量处理组(P<0.05),且均显著高于抗生素组(P<0.05);但对胫骨灰分无影响(P>0.05)。22~42日龄时,与对照组相比,饲粮菌株MH22组肉仔鸡胫骨强度显著增强(P<0.05),胫骨钙含量显著增加(P<0.05),但对胫骨灰分无影响(P>0.05)。相对于对照组抗生素组,MH22高剂量组显著提高了胫骨磷含量(P>0.05)。

结果显示,添加运动芽孢杆菌MH22能提高肉仔鸡的平均日增重,降低料重比;其次,添加运动芽孢杆菌MH22能促进肉鸡的钙、磷的吸收转化,增强胫骨强度,减少软骨病的发生。

1.一株运动芽孢杆菌(Bacillus toyonensis)MH22,其特征在于,所述运动芽孢杆菌(Bacillus toyonensis)MH22的保藏号为GDMCC No:63422。
2.权利要求1所述的运动芽孢杆菌(Bacillus toyonensis)MH22在抑制大肠杆菌、金黄色葡萄球菌和肠炎沙门氏菌生长中的应用。
3.权利要求1所述的运动芽孢杆菌(Bacillus toyonensis)MH22在制备抑制大肠杆菌、金黄色葡萄球菌或肠炎沙门氏菌生长的药品中的应用。
4.一种治疗肠炎或者痢疾的药物,其特征在于,所述药物含有权利要求1所述的运动芽孢杆菌(Bacillus toyonensis)MH22。
5.权利要求1所述的运动芽孢杆菌(Bacillus toyonensis)MH22在制备饲料添加剂中的应用。
6.根据权利要求5所述的应用,其特征在于,所述饲料添加剂能降低肉鸡的平均日采食量、降低料重比;促进肉鸡对钙、磷元素的吸收。
7.一种微生态制剂,其特征在于,所述微生态制剂包含权利要求1所述的运动芽孢杆菌(Bacillus toyonensis)MH22。
8.权利要求7所述的微生态制剂在降低肉鸡的平均日采食量、降低料重比或促进肉鸡对钙、磷元素的吸收中的应用。
9.权利要求7所述的微生态制剂在制备饲料添加剂中的应用。
