Abstract

"MéTODO DE PREPARAçãO DE PLASMìDEO DNA DE GRAU FARMACêUTICO". A invenção proporciona um processo para lise alcalina contínua de uma suspensão bacterial de modo a recolher pDNA. Ela também proporciona etapas de purificação adicional opcional, incluindo filtração de lisato, uma cromatografia de troca de ânion, cromatografia de afinidade triplex, e cromatografia de interação hidrofóbica. Estas etapas de purificação adicionais podem ser combinadas com a lise contínua de modo a produzir um produto de pDNA altamente purificado substancialmente livre de gDNA, RNA, proteína, endotoxina, e outros contaminantes.
