Abstract
patente de invenção "processo para determinar a presença e/ou quantidade de um microorganismo e/ou seu material intracelular presente em uma amostra, estojo de teste, e, aparelho para realizar um processo". um processo é fornecido para determinar a presença e/ou quantidade de um microorganismo e/ou seu material intercelular presente em uma amostra caracterizada em que: (a) expor a amostra a um agente ligante específico que tenha sido imobilizado em um substrato sólido, o agente ligante específico sendo capaz de se ligar ao microorganismo ou seu material intracelular tal que ele se torna associado com o substrato sólido, (b) expor o substrato sólido a um agente capaz de fazer adenilato quinase associada com o microorganismo e/ou seu material intracelular acessível a soluções aplicadas ao substrato, (c) aplicar uma solução contendo difosfato de adenosina (adp) ao substrato sob condições por qual meio trifosfato de adenosina (atp) pode ser produzido por qualquer adenilato quinase presente, e (d) medir a quantidade de trifosfato de adenosina (atp) e relacionar isto à presença e/ou quantidade de microorganismo ou conteúdo intracelular. etapa (d) pode ser realizada usando um teste que inclui uma reação formadora de cor, mas é preferência realizada pelo uso de reagente luciferase/luciferina para produzir luz proporcional à quantidade de atp produzida, eisto é detectado usando um luminômetro. de preferência etapa (c) inclui presença de íons de magnésio em uma concentração molar suficiente para permitir conversão máxima de adp a atp. de maior preferência etapas (b) e (c) realizadas adicionando-se extrator, adp e íons de magnésio na amostra e incubando a mistura por predeterminado período para efetuar conversão de adp a atp.
