Abstract

Изобретението се отнася до подобрен метод за изолиране на клавуланова киселина и нейните фармацевтично приемливи соли като калиев клавуланат от воденразтвор на ферментационна хранителна среда на Streptomyces sp. P 6621 FERM P 2804. От ферментационната хранителна среда на мицела главната част на протеините и други суспендирани твърди частици се отделят чрез микрофилтруване при рН на средата 5,8-6,2 и температура от 20 до 400С. Пречистената хранителна среда /водната фаза/ по желание допълнителносе пречиства чрез ултрафилтруване, след което се концентрира чрез обратна осмоза и директно се екстрахира в противоток в центрофужни екстрактори с водоразтворим органичен разтворител при рН на средата 1-3.Останалите протеини едновременно се отделят от получената органична фаза на водоразтворимите нежелателни примеси, отделени чрез промиване на органичната фазас вода. След това органичната фаза сеизсушава под вакуум в ректификационна колона до водно съдържание под 0,1% об., концентрира се чрез изпаряване и се обезцветява чрез обработване с активен въглен. Клавулановата киселина, присъстваща всухата органична фаза, за предпочитане в етилацетатната, се изолира и пречиства чрез взаимодействие с N,N'-диизопропилетилендиамин при стайна температура до междинното съединение N,N'-диизопропилетилендиамониев диклавуланат. Полученото съединение се изолира, след което реагира с калиев 2-етил хексаноат в изопропанол до получаването на калиева солна клавуланова кисели
