Abstract

Se describe un polinucleótido donante universal que puede insertarse en ubicaciones reconocidas como objetivo en genomas de plantas para facilitar la integración rápida y de alto rendimiento de una molécula donante dentro de una ubicación genómica específica. Reivindicación 1: Un casete de donante polinucleótido que comprende un dominio de unión de nucleasa específica de un sitio, un dominio analítico, y un dominio de plásmido. Reivindicación 36: Un método para integración objetivada de un casete de donante polinucleótido dentro del genoma de una célula de planta, en donde el método comprende: a) expresar una nucleasa de unión de ADN específico de sitio, que comprende por lo menos un dominio de unión de ARN y por lo menos un dominio de nucleasa, en donde por lo menos un dominio de unión de ADN se une a un sitio objetivo dentro del genoma de la célula de planta; b) poner en contacto la célula de planta con un casete de donante polinucleótido; c) clivar el sitio objetivo dentro del genoma de la célula de planta con la nucleasa de unión de ADN especifico de sitio; y d) integrar el casete de donante polinucleótido dentro del sitio objetivo dentro del genoma de la célula de planta.
