Abstract
Un método para la producción masiva del factor de coagulación VII humano. El método comprende a) construir un vector de expresión que contiene i) un promotor de la dihidrofolato reductasa desprovista de una de más de las secuencias de repetición CCGCC de la región rica en GC del mismo y un gen de dihidrofolato reductasa (DHFR) ligado operativamente al mismo, y ii) un promotor de citomegalovirus (CMV) y un gen del factor de coagulación VII humano ligado operativamente al mismo; b) transfectar el vector de expresión del paso a) en una línea celular animal; c) cultivar la línea celular animal transfectada del paso b) en la presencia de un inhibidor dihidrofolato reductasa para seleccionar células que expresan al factor de coagulación VII humano con gran eficacia; y d) agregar butirato de sodio a las células animales del paso c), y una línea celular para la producción masiva del factor de coagulación VII humano. El uso del vector que contiene un promotor DHFR desprovisto de las secuencias de repetición ricas en GC en el método permite producir al factor de coagulación VII humano con una gran eficacia a gran escala, que contribuye en el tratamiento de la hemofilia. Una línea celular HMF 708 para producir factor de coagulación humano VII.
