Abstract
Moléculas de unión biespecíficas que se unen tanto a VEGF como a Ang2, preferiblemente en forma de dominios variables individuales de inmunoglobulina tales como inmunoglobulina G de dominio variable de cadena pesada (VHH) y anticuerpos de dominio, composiciones farmacéuticas que las contienen y su uso en el tratamiento de enfermedades que están asociadas con los efectos mediados por VEGF y/o Ang2 sobre la angiogénesis (tratamiento de enfermedades cancerosas y enfermedades oftálmicas). Además, también ácidos nucleicos que codifican moléculas de unión biespecíficas, células huésped y métodos para prepararlos.Reivindicación 1: Una molécula de unión biespecífica caracterizada porque comprende al menos un componente de unión a VEGF, al menos un componente de unión a Ang2 y al menos un componente de unión de albúmina sérica, en la que dicho componente de unión a Ang2 se une con Ang2 con una potencia al menos 5.000 veces más alta que a Ang1 o a Ang4. Reivindicación 2: Una molécula de unión biespecífica de la reivindicación 1, caracterizada porque dicho componente de unión a VEGF comprende al menos un dominio variable con cuatro regiones marco y tres regiones determinantes de complementariedad CDR1, CDR2 y CDR3, respectivamente, en la que dicha CDR3 tiene la secuencia de aminoácidos Ser Arg Ala Tyr Xaa Ser Xaa Arg Leu Arg Leu Xaa Xaa Thr Tyr Xaa Tyr como se muestra en SEC ID Nº 1, en la que Xaa en la posición 5 es Gly o Ala; Xaa en la posición 7 es Ser o Gly; Xaa en la posición 12 es Gly, Ala o Pro; Xaa en la posición 13 es Asp o Gly; Xaa en la posición 16 es Asp o Glu; y en la que dicho componente de unión a VEGF es capaz de bloquear la interacción de VEGF165 recombinante humano con el VEGFR-2 recombinante humano con una tasa de inhibición de ³ 60%.
