Abstract
Description
このうち、皮膚の黒化やしみ、そばかすを防ぎ、本来の白い肌を保つための美白化粧料に応用されているものとしては、コウジ酸、アルブチン、ハイドロキノンモノベンジルエーテル、過酸化水素等が知られている。しかしながら、これらの美白剤を配合すると、若干、色黒の肌を淡色化する効果はあるものの、美白化粧料として十分満足すべき効果が得られておらず、さらに紫外線による炎症抑制効果はなく、安全性の面でも問題を残すものが多かった。また、紫外線による炎症を抑制する美白剤としては、ビタミンC及びその誘導体が知られている。
従って、本発明の課題は、良好な美白作用に加えて安定性、外用剤基剤への溶解性に優れた化合物を探索し、当該化合物を含有する皮膚外用剤を提供することにある。

4-(3-アルコキシブチル)フェノール化合物の合成はMichaelらの方法に従って行った(Journal of the American Chemical Society、pp.3659-3661、1972)。4-(4-ヒドロキシフェニル)-2-ブタノン(10mmol)及びトリエチルシラン(11mmol)を各種アルコール(125mmol)に溶解し、氷冷下でトリフルオロ酢酸(375mmol)を滴下した後、室温に戻して24時間攪拌した。反応液に氷水(100mL)を加え、ヘキサン(100mL)で2回抽出した。得られたヘキサン層を飽和食塩水及び飽和炭酸水素ナトリウム水で数回洗浄した後、無水硫酸マグネシウムで乾燥、ろ過した。ろ液を減圧下濃縮した後、シリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル溶媒系)で精製し、以下に示す各化合物を得た。これらの化合物の13C-NMRスペクトルを図1~図5に示す。

各化合物0.1gに、下記表1に示した各種溶媒1mLを加え、40℃で10分間加温後、室温に戻した。この時の溶媒への溶解性を目視にて評価した。評価は、溶解している場合を「○」、不溶物が残っている場合を「×」と判定した。
上記製造例1で得られた4-(3-アルコキシブチル)フェノール化合物について下記に示すメラニン生成抑制試験を行った。尚、比較対象として、メラニン生成抑制作用が公知であるアルブチン、アスコルビン酸、コウジ酸、リノール酸を用いた。
B16メラノーマ細胞を、10vol%牛胎児血清含有MEM培地で、12穴培養プレートに1×104個/wellとなるように播種し、常法にて24時間前培養した。前培養後、各評価試料を各種濃度で添加した試験培地と培地交換し、72時間培養を行った。試験培地としては、上記の前培養用培地にテオフィリンを2mmol/Lとなるように添加したものを使用した。培養後、細胞を回収し、10vol%ジメチルスルホキシドを含有する1mol/L水酸化ナトリウム水溶液に溶解させ、溶解液のOD475値を測定してメラニン量の指標とした。また同時に、Coomasie Plus Protein Assay Kit(PIERCE社製)を用いて、溶解液の総タンパク質量を定量し、タンパク質量当たりのメラニン量を算出した。
算出したタンパク質量当たりのメラニン量に基き、下式により各種添加濃度毎にメラニン生成抑制率を求めた。
メラニン生成抑制率(%)=(A-B)/A×100
(但し、A:試料無添加時のタンパク質量当たりのメラニン量、B:試料添加時のタンパク質量当たりのメラニン量)
次いで、横軸に試料添加濃度、縦軸にメラニン生成抑制率をプロットしたグラフを作成し、このグラフから各評価試料がメラニン生成を50%抑制する濃度(IC50)を決定した。
上記製造例1で得られた4-(3-アルコキシブチル)フェノールについて下記に示すメラニン生成抑制試験を行った。尚、比較対象として、アルキル基を除去した4-(3-ヒドロキシブチル)フェノール、同じ炭素数を有するエステル化合物である4-(3-アセトキシブチル)フェノール、4-(3-プロパノイルオキシブチル)フェノールを用いた。
メラニン細胞含有培養皮膚モデル(MEL-300、Asian Doner、倉敷紡績株式会社製)の皮膚モデルカップに被験物質溶液0.2mLを入れ、皮膚モデルを製品添付のLLMM培地を用いて14日間培養した。培養期間中、培地は3日おきに交換した。培養14日後、皮膚モデルを洗浄し、AlamarBlue(Molecular Probe社製)試薬を用いた蛍光測定(Excitation:560nm、Emmition:590nm)により生存率を測定した後、各皮膚モデル中のメラニン合成量を測定した。メラニン合成量は、各皮膚モデルをPBSに浸漬して細胞層を分離後、PBS及び含水エタノールで洗浄し、2NNaOH水溶液0.2mLを加えて100℃、3時間の条件でメラニン抽出した溶液の比色測定(405nm)により求めた。
結果を表3に示す。
上記製造例1で得られた4-(3-エトキシブチル)フェノール化合物を配合したスキンクリーム(実施例1)を用いて、下記に示す美白実用試験を行った。尚、比較対象として、美白作用が公知であるアルブチンを配合したスキンクリーム(比較例1)を用いた。
被験者20名の前腕屈側部皮膚に、夏季の太陽光を3時間(1日1.5時間で連続2日間)曝露した。曝露後より被験者の左前腕屈側部皮膚には、下記表4の実施例1のスキンクリームを1日朝夕1回ずつ13週間連続で塗布した。また被験者の右前腕屈側部皮膚には、下記表3の比較例1のスキンクリームを同様の条件で塗布した。
最終塗布終了時に、左右の前腕屈側部皮膚の連用前後における美白の程度に関し、専門判定員により評価した。尚、評価は美白効果が確認された被験者を「美白効果あり」とし、その人数を示した。
結果を下記表5に示す。この結果から、本発明の製造例1の4-(3-エトキシブチル)フェノール化合物を配合したスキンクリームは、比較例1のアルブチンを配合したスキンクリームと比較して、美白化粧料として非常に優れていることが分かる。なお、試験期間中、実施例1のスキンクリームを塗布した部位に、皮膚刺激反応及び皮膚感作反応が認められた被験者はおらず、本発明品が製剤の形態においても安全であることが確認できた。

- 一般式(1)中のRが、メチル基、エチル基、n-プロピル基又はイソプロピル基である請求項1記載の皮膚外用剤。
- 一般式(1)中のRが、エチル基である請求項1記載の皮膚外用剤。
- 一般式(1)中のRが、炭素数1~4のアルキル基である請求項5記載の美白化粧料。
- 一般式(1)中のRが、メチル基、エチル基、n-プロピル基又はイソプロピル基である請求項5記載の美白化粧料。
- 一般式(1)中のRが、エチル基である請求項5記載の美白化粧料。
- 一般式(1)中のRが、炭素数1~4のアルキル基である請求項9記載のメラニン生成抑制剤。
- 一般式(1)中のRが、メチル基、エチル基、n-プロピル基又はイソプロピル基である請求項9記載のメラニン生成抑制剤。
- 一般式(1)中のRが、エチル基である請求項9記載のメラニン生成抑制剤。
