Abstract

【課題】１又は複数種類の病原微生物を検出・定量する場合など、試料中の微量なＲＮＡからを簡便かつ迅速に標的ＲＮＡを検出・定量することができる方法やキットを提供する。 【解決手段】１）標的ＲＮＡを含む試料から、プロモーター配列を有する液相プライマーと逆転写酵素とを用いてｃＤＮＡを合成し、ｃＤＮＡ－ＲＮＡ複合体を得て、２）該複合体のＲＮＡを分解し、３）前記２）で得たｃＤＮＡと前記固相プライマーとを介して、二本鎖ＤＮＡを合成し、４）該二本鎖ＤＮＡからＲＮＡを合成し、５）前記４）で得たＲＮＡと前記固相プライマーとを介してｃＤＮＡを合成し、ｃＤＮＡ－ＲＮＡ複合体を得て、６）前記５）で得た複合体のＲＮＡを分解し、７）前記６）で得たｃＤＮＡと前記液相プライマーとを介して、二本鎖ＤＮＡを合成し、８）前記３）及び７）で得た二本鎖ＤＮＡを定量する。これらは、一の反応液中で行うことができる。
