Abstract

L'invention concerne un procédé de détection de gène, au moyen duquel un gène peut être détecté par formation d'un polymère grâce au procédé PALSAR sans capture des oligonucléotides ligaturés ni lavage des oligonucléotides en excès. En utilisant plusieurs paires de sondes constituées de n (n≥3) zones de séquence de base mutuellement complémentaires, un point d'un côté du domaine complémentaire, un point de chaque côté de celui-ci ou deux ou plusieurs points d'un gène cible de la paire de sondes précitée sont préalablement clivés, puis une réaction d'hybridation, une réaction de ligature et une réaction de dissociation sont effectuées à température contrôlée. Ainsi, les sondes clivées sont ligaturées afin de former une sonde entière. En utilisant un procédé de construction d'un polymère de sonde, un polymère bicaténaire est formé et le gène précité est détecté.
