Abstract
La présente invention concerne une protéine épithéliale purifiée et isolée, un peptide issu de cette protéine et certaines de ses variantes, la présence accrue de ces éléments dans une cellule épithéliale étant l'indication d'un précancer. En l'occurrence, une protéine épithéliale qui constitue une détection précoce marquée pour le cancer du poumon a été purifiée à partir de deux lignées cellulaires de cancer du poumon humain, NCI-H720 et NCI-H157. Grâce à une procédure en six opérations, on a purifié la protéine épithéliale au moyen d'un système de détection par tampons Western sous des conditions non réductrices ainsi que sous des conditions réductrices. Les opérations de purification incluaient une chromatographie sur colonne d'échange anionique, une chromatographie préparative à focalisation isoélectrique, une chromatographie liquide haute performance du radical C18 sur base polymère et une chromatographie liquide haute performance analytique du radical C4. Après une purification d'approximativement 25 000 fois, la protéine de coloration immunologique était pure à plus de 90 % d'après le test de coloration au bleu de Coomassie après réduction de la PAGE SDS. La protéine épithéliale primaire partage une certaine homologie de séquence avec la ribonucléoprotéine A2 nucléaire hétérogène (hnRNP). Une protéine épithéliale mineure de co-purification partage une certaine homologie de séquence avec la variante d'épissage hnRNP-B1. L'analyse moléculaire des cultures de cellules normales de l'épithélium bronchique a mis en évidence un bas niveau d'expression de protéine épithéliale, en concordance avec la coloration immuno-histochimique des échantillons cliniques pour un niveau accru de l'expression dans la plupart des cellules du cancer du poumon. Cette protéine épithéliale est un marqueur de la transformation épithéliale du poumon, du sein, de l'os, de l'ovaire, de la prostate, du rein, des mélanomes et myélomes, et est susceptible d'être en liaison avec le processus de carcinogenèse. L'invention concerne également des procédés de surveillance de l'expression de la protéine épithéliale, du peptide issu de cette protéine et certaines de ses variantes par utilisation de techniques moléculaires et immunologiques pour la recherche du cancer ou du précancer chez les mammifères. L'invention concerne enfin un procédé de diagnostic informatisé du cancer ou du précancer par détection des niveaux d'ARN messager du hnRNP.
