Abstract
The present invention discloses compositions containing naturally secreted human extracellular matricies, the methods of making those matricies, and the method of using those matricies. The extracellular matrix is obtained by growing cells into a three-dimensional support structure so that the cells secrete an extracellular matrix. Next, the cells are killed and removed from the support structure leaving only the extracellular matrix on the support structure. The extracellular matrix can then be removed from the support, combined with a suitable pharmacologically acceptable carrier and injected into a tissue defect area in the skin of a patient.
Claims
1. A method for the production of a naturally secreted extracellular matrix coated three-dimensional framework comprising the steps of:
(a) culturing cells on a three-dimensional framework under conditions favorable for cellular growth for a pre-determined time period such that an extracellular matrix is secreted onto the inoculated framework creating a coated framework; (b) killing the cells and
(c) removing the cells and cellular debris.
2. A method according to claim 1 wherein the three-dimensional framework is a material selected from the group consisting of polyamide, polyester, polystyrene, polypropylene, polyacrylates, polyvinyl compounds,
polycarbonate, polytetrafluorethylene, thermanox,
nitrocellulose, cotton, polyglycolie acid, catgut suture material, cellulose, gelatin, chitosan, hyaluronic acid and dextran,
3. A method according to claim 1 or 2, wherein the three-dimensional framework has pore spaces of about 150μm to about 220μm.
4. The method according to any one of claims 1 to 3, wherein the extracellular matrix is secreted by tissue specific cells selected from the group consisting of
fibroblasts, osteoblasts, odontoblasts, chondrocytes, epithelial cells, smoothst-muscle cells, retinal cells, endothelial cells, stromal cells or combinations thereof.
5. A method for the production of a naturally secreted extracellular matrix comprising carrying out steps (a), (b) and (c) according to any one of claims 1 to 4 and then (d) collecting the extracellular matrix deposited on the coated framework.
6. A composition comprising a three-dimensional framework coated with a naturally secreted extracellular matrix composed of human proteins.
7. A composition according to claim 6, wherein the framework is made from a material selected from the group consisting of polyamide, polyester, polystyrene,
polypropylene, polyacrylates, polyvinyl compounds,
polycarbonate, polytetrafluorethylene, thermanox,
nitrocellulose, cotton, polyglycolic acid, catgut sutures, cellulose, gelatin, chitosan, hyaluronic acid and dextran.
8. A composition according to claim 6 or 7, wherein the three-dimensional framework has pore spaces of about 150μm to about 220μm.
9. A composition according to any one of claims 6 to 8, wherein the naturally secreted extracellular matrix is secreted by tissue specific ceils selected from the group consisting of fibroblasts, oεteoblasts, odontoblasts, chondrocytes, epithelial cells, smooth muscle cells, retinal cells, endothelial cells, stromal cells or combinations thereof.
10. An injectable naturally secreted extracellular matrix composition for the treatment of tissue defects comprising a human naturally secreted extracellular matrix and a pharmaceutically acceptable carrier.
11. A composition according to claim 10, wherein the naturally secreted extracellular matrix is produced by a method according to claim 5.
12. A composition according to claim 11 wherein naturally secreted extracellular matrices secreted by
different tissue or cell types are mixed between said step (d) and step (e), consisting in processing the collected extracellular matrix with a pharmaceutically acceptable carrier, such that ratios of collagen types I-V, respective to each other, are adjusted.
13. A method for the repair of tissue defects comprising injecting at a site of a tissue defect a
composition according to any one of claims 10-12.
14. A method according to claim 13 wherein the naturally secreted extracellular matrix is produced by a method according to claim 5.
15. The use of a composition according to any one of claims 10-12 for the repair of tissue defects.
