Abstract

Description
本發明係關於用於治療及/或預防肥胖症及肥胖症相關病症之化合物。特定而言,本發明提供用作ATF3誘導劑且用於治療及/或預防肥胖症及肥胖症相關病症(例如心臟病、高血壓、高脂血症及糖尿病)之𠳭烯酮衍生物。
肥胖症係指過量脂肪組織因基因、環境及心理學因素而累積於身體中之病狀,其會產生健康病症,且可引起嚴重健康問題(例如高血壓、高脂血症、心臟病、糖尿病、癌症等)。肥胖症亦顯著增加發生心血管疾病之風險。冠狀動脈供血不足、動脈粥樣硬化疾病及心機能不全位於由肥胖症誘導之心血管併發症之最前列。並無實際治療係基於發現於肥胖症及其相關病狀(例如代謝症候群或動脈粥樣硬化)中之原發性代謝異常之生物學。 活化轉錄因子3 (ATF3)係ATF/環AMP反應元件結合(ATF/CREB)家族之轉錄因子之成員。KR1556439 B1提供用於預防、治療及篩選粒線體功能障礙相關疾病(例如高血壓、肥胖症、胰島素抗性、脂肪肝、肝纖維化、代謝症候群、神經退化性病症、雙相型病症、糖尿病、缺血、心臟衰竭、癌症及心血管疾病)之醫藥組合物,且包括一或多種轉錄因子(包含ATF3)之啟動子活性或表現。研究表明,ATF3可在肥胖症中之脂肪細胞低氧介導性粒線體功能障礙中發揮一定作用(Biochem Biophys Res Commun. 2013 Feb 15;431(3):421-7)。 因此,仍需要用於治療患有肥胖症及肥胖症相關病症之個體之新化合物。
本發明係關於用作ATF3誘導劑且用於治療及/或預防肥胖症及肥胖症相關病症之𠳭烯酮衍生物。該等化合物係藉由以下條件來獲得:(1)
將ATF3 (pGL4-ATF3)之特殊DNA序列轉染至3T3-L1前脂肪細胞及H9C2心肌細胞中;(2)
使用穩定純系作為用於篩選抗肥胖症、抗動脈粥樣硬化、抗心血管疾病藥物且促進白褐色脂肪細胞轉分化之平臺;及(3)
使用用於特殊藥物篩選之平臺。本發明成功地基於上文所提及ATF3表現之誘導來篩選藥物且該等藥物包含(但不限於) ST32c、ST32da、ST32db及其衍生物。本發明另外發現,該等化合物對肥胖症及肥胖症誘導性糖尿病具有抑制效應且對白褐色脂肪細胞轉分化具有易化效應,由此增加身體能量消耗並阻抑糖尿病。另外,該等化合物亦可抑制高血壓或低氧誘導性心肌病以增加心臟功能。 因此,本發明提供具有如本文所揭示之式(I)之化合物。 本發明亦提供包括本文所揭示之化合物之醫藥組合物。 本發明亦提供治療及/或預防肥胖症、肥胖症誘導性糖尿病、抑制高血壓或低氧誘導性心肌病以由此增加心臟功能之方法,其包括向個體投與有效量之本文所揭示之化合物。 本發明亦提供選擇AT3誘導劑作為抗肥胖劑、抗動脈粥樣硬化劑或抗心血管劑之方法,其包括使一種化合物與經重組ATF3轉染之細胞接觸且檢測細胞之ATF3表現,並測定在細胞表現自AT3轉錄之蛋白質時該化合物是否係AT3誘導劑。

- 如請求項1之化合物,其係選自由以下組成之群:ST32c:2-甲基-1H-苯并[h]呋喃并[3,2-c]烯-11(2H)-酮;ST32da:(1R,2S)-1,2-二甲基-1H-苯并[h]呋喃并[3,2-c]烯-11(2H)-酮;ST32da':(1R,2R)-5-氯-1,2-二甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-酮;ST32db:(1R,2R)-1,2-二甲基-1H-苯并[h]呋喃并[3,2-c]烯-11(2H)-酮;ST32db:(1R,2R)-1,2-二甲基-1H-苯并[h]呋喃并[3,2-c]烯-11(2H)-酮];ST32BA:(1R,2S)-1,2,6-三甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-酮;6c:(1R,2S)-5-溴-1,2-二甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-酮;6c':(1R,2R)-5-溴-1,2-二甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-酮;11:(1R,2S)-1,2-二甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-硫酮;及11':(1R,2R)-1,2-二甲基-1,2-二氫-11H-苯并[h]呋喃并[3,2-c]烯-11-硫酮; 或其互變異構體、對映異構體、立體異構體或其溶劑合物、前藥或醫藥上可接受之鹽。

- 一種如請求項1至4中任一項之化合物或其互變異構體、對映異構體、立體異構體或其溶劑合物、前藥或醫藥上可接受之鹽之用途,其用於製備治療及/或預防肥胖症、肥胖症誘導性糖尿病、高血壓或低氧誘導性心肌病或由此增加心臟功能之藥劑。
- 如請求項7之用途,其中該肥胖症係高脂肪食物誘導性肥胖症。
- 如請求項7之用途,其中該化合物之投與劑量介於1mg/kg/天至2,000mg/kg/天之間。
