Description

可噴霧式假牙清潔劑已描述於1985年4月16日核發給Shah的美國專利3,808,686、3,822,212和4,511,486,其揭示一種含有表面活性劑、濕潤劑、水和大量乙醇或異丙醇的泡沫式假牙清潔劑。該乙醇係用作為各種賦形劑的助溶劑,其可使經清潔的假牙產生一種清新感及抗菌效果。
這些方法均無法使特別是假牙配戴者感受到較長時間的呼吸清新感。假牙配戴者抱怨其假牙發出一股令人不悅的味道,其通常稱為”假牙氣味(denture breath)”。目前市面上的產品如利用浸泡法需要較長的清潔時間,但無法提供例如清新氣味和清新口氣的優點。假牙配戴者在許多情況下必需依賴一般的牙膏刷洗其假牙以維持假牙/清新口氣因而由於磨擦而造成假牙表面的刮損。此外,由於這些方法均無法長期維持清新的口氣而使配戴者通常需每天依賴假牙的多次清洗而感到極不方便和厭煩。

“假牙清潔劑”一詞指一種用於口腔外部清潔牙齒器具的配製物。
“發泡性配製物”一詞指能利用推噴劑或無推噴劑之泡沫型噴霧器而被從噴霧器壓出的亞穩態液體乳劑。

除了卓越的清潔效果之外此配製物的其他優點為至少可使假牙配戴者維持5小時以上之口氣的改善和清新的口腔氣味。因此,本發明係描述一種泡沫式假牙清潔劑,其可清潔假牙、殺滅細菌,產生高達5小時之清新氣味和口氣的效果。該牙齒器具清潔劑配製物係利用亞穩態油/水乳劑,其中大部分的油相係由包括植物油及/或礦物油之風味油和不可混合油所構成,以及其水相主要係由清潔劑、乳化劑、去污劑、制菌劑、香料、甜味劑和緩衝劑所構成。該乳劑之形成為暫時性並且在使用前藉由輕輕地搖晃容器而獲得均質的產品。在容器內的配製物係呈液態其可利用推噴劑或無推噴劑而經由泡沫噴頭形成泡沫。該配製物可被直接塗抹於牙齒器具上並且建議利用牙刷或用於牙齒器具的義齒刷至少刷洗約30至60秒的一段有效時間。在一具體實施例中,該牙齒器具的清潔有效時間為至少約60秒鐘。清潔之後,以清水清除掉牙齒器具上的配製物然後將其安裝於口腔內。
泡沫內的表面活性劑和去污劑特別是在刷洗下可清潔和穿透該牙齒器具。在至少約30至60秒的刷拭及經過洗滌之後,該油相由於油滴的聚結而變得不穩定。在不侷泥於特定的理論之下,該配製物可能沈積於牙齒器具的表面而使其帶有該芳香氣味。由於該氣味無法緊密地貼著於牙齒器具的表面,因此較佳為能被吸收入該牙齒器具的結構內。此現象可產生一股引人注意的愉悅口腔氣味並且即使在洗滌之後仍可維持長期的清新口氣。
當該清潔配製物接觸牙齒器具至常約30至60秒鐘之後可維持長期的清新口氣。在一具體實施例中,該牙齒器具與清潔配製物接觸至少約60秒鐘以維持長期清新的口氣。
本發明的一項特色為該牙齒器具清潔乳劑配製物含有約2至50%重量/重量(w/w)的油相,並且可產生充氣泡沫。事實上,熟習本技術之人士無法想像該充氣泡沫之亞穩態乳劑能夠具有如此高的含油量。
適合用於本發明之不可混合油類包括,但不侷限於,芝麻油、大豆油、菜籽油、椰子油、分餾椰子油、魚油或礦物油。本發明的一具體實施例為含有芝麻油。配製物內該不可混合油的含量為約0.5~20%重量/重量。在本發明的一具體實施例中,該不可混合油的含量為約1~8%重量/重量。在本發明的另一具體實施例中,其芝麻油的含量為約3~6%重量/重量。
該牙齒清潔配製物除了不可混合油之外可含有約0.5~15%重量/重量的風味油。適合用於本發明的風味油包括,但不侷限於,精油如綠薄荷(spearmint)、冬清(wintergreen)、檸檬烯(limonine)、薄荷醇、胡椒薄荷、沉香醇(linolol)、柑橘油、茴香腦(anethole)、甜茴香(fennel sweet)、桉葉醇(eucalyptol)、洋甘菊油(camomile)、羅勒油(basil)、薑、迷迭香油、左旋薄荷醇、水楊酸甲酯、丁香油、龍蒿(tarragon)、茶樹油、瑞香酚(thymol)、小豆蔻油(cardamom)、檸檬醛(citral)、牛至(Origanum Damascone F)、癸醇、壬醇、癸醛、α-紫羅蘭酮(ionone)、香草、大茴香子(aniseed)、桂皮醛、石灰或其混合物。在本發明的一具體實施例中含有約2~6%重量/重量的風味油。
配製物內之該亞穩態乳劑的水相為約50至99%重量/重量的含量。在本發明的一具體實施例中,該水相為約50至80%重量/重量的含量。在本發明的第二具體實施例中,該水相為約75~90%重量/重量的含量。
為獲得大量的泡沫,牙齒清潔配製物內加入表面活性劑作為成泡劑,並且有助於配製物的清潔效力。
適合用於本發明的表面活性劑包括,但不侷限於,陰離子表面活性劑、兩性離子表面活性劑和非離子表面活性劑,或其組合。適合的陰離子表面活性劑包括,但不侷限於,月桂基硫酸鈉和其他烷基硫酸鹽、烷基醚硫酸鹽、月桂基磺基乙酸鈉、二烷基磺基琥珀酸鹽、烷基苯磺酸鹽、α-烯羥磺酸鹽、醯基N-牛磺酸甲酯,以及月桂醯基肌胺酸鈉。在本發明的一具體實施例中月桂基硫酸鈉的含量為0.5~4%重量/重量。
適合的非離子表面活性劑包括,但不侷限於,聚氧氫化蓖麻油、乙氧基化山梨糖醇烷酸鹽、脂肪酸乙氧基酯、脂肪醇乙氧基酯、脂胺乙氧基酯、聚氧化乙烯/聚氧化丙烯嵌段聚合物(Pluronics)和蔗糖酯。在本發明的一具體實施例中聚氧40氫化蓖麻油的含量為0.5~6%重量/重量。
適合的兩性離子表面活性劑包括,但不侷限於,椰油醯胺丙基甜菜鹼(betaine)和烷基醯胺丙基甜菜鹼。在本發明的一具體實施例中椰油醯胺丙基甜菜鹼的含量為0.5~4%重量/重量。在本發明的一具體實施例中含有月桂基硫酸鈉0.5~4%重量/重量、聚氧40氫化蓖麻油0.5~6%重量/重量和椰油醯胺丙基甜菜鹼0.5~4%重量/重量之混合物的表面活性劑。
用於本發明之含表面活性劑混合物的適合表面活性劑具有大於約10的HLB。在一具體實施例中,該表面活性劑或其混合物的HLB係大於約16。
濕潤劑的存在有助於保濕的作用。適合的水溶性醫藥上可接受濕潤劑包括,但不侷限於,甘油、山梨糖醇(通常使用其70%溶液)、木糖醇、甘露糖醇和半乳糖醇。在本發明的一具體實施例中濕潤劑的含量為約1~20%重量/重量。在本發明另一具體實施例中濕潤劑的含量為約5~15%重量/重量。
本假牙清潔配製物亦可加入賦形劑,舉例性的賦形劑為保存劑如苯甲酸鈉、苯甲酸、對羥基苯甲酸甲酯(methyl paraben)、對羥基苯甲酸丙酯等;螯合劑如依地酸二鈉(edentate disodium)、檸檬酸鹽、酒石酸鹽、偏矽酸鈉、焦磷酸鈉、焦磷酸鉀、三聚焦磷酸鹽、三聚焦磷酸鈉;抗氧化劑如丁基化羥基甲苯(BHT)、丁基化羥基苯甲醚(BHA)和第三丁基氫醌(TBHQ);緩衝劑;去污劑如聚乙二醇;著色劑;黏度調節劑;及其混合物。
可藉由任何方便的順序混合適當相對量之成分而製造根據本發明的組成物(請看下列實施例1)。
可藉由技術中習知的泡沫產生裝置輕易地製造該牙齒器具清潔泡沫。無推噴劑的噴霧器或利用推噴劑的噴霧器均適合用於本發明的配製物。實施例為Airspray公司(佛州Pompano Beach市)所製造的F2指泵製泡機或M3微指泵製泡機,或Emsar公司(荷蘭Alkmaar市)所製造的37MS手提式防塞泵製泡機。
該牙齒器具清潔劑配製物係被置於牙齒器具上一段足夠清潔該牙齒器具的時間。形成泡沫之後建議配戴者使用牙刷或假牙刷刷拭該牙齒器具,但非必需。在至少刷拭60秒鐘之後,以水洗滌該假牙然後再配戴於口腔內。
此配製物的另一態樣為其不需特殊的抗菌劑即可有效地殺死細菌和其他微生物及伴隨的假牙氣味。利用微生物學殺菌試驗測定原型液體成泡假牙清潔劑在接觸細菌90秒之後的抗菌效果。
該試驗著重述於此處之假牙清潔配製物對抗細菌如核梭桿菌(Fusobacteriumnucleatum
)、肺炎桿菌(Klebsiella pneumoniae
)、血鏈球菌(Streptococcus sasguis
)、粘性放線菌(Actinomyces viscosus
)和韋榮氏球菌(Veillonella spp.
)所造成之口臭的效果。表1顯示懸浮液內與稀釋產品接觸90秒後之細菌死亡百分比的結果,該產品係屬於本發明範圍內的配製物。以1:1比例稀釋之產品在滅菌自來水、滅菌人工涶液和其他干擾物質如牛血清白蛋白的懸浮液內作為其殺菌動力模型進行測定。此模型類似其在假牙上混合涶液、水和其他物質的稀釋系數。此模型代表該產品在假牙上可能遇到的實際情況。
試驗計劃
:在胰蛋白豆培養液內培養兼性細菌(例如,肺炎桿菌和血鏈球菌)及以含2% oxyrase之硫乙醇酸鹽培養基培養厭氧菌(例如,核梭桿菌和韋榮氏球菌)。該兼性細菌係在30~35℃下最少培養48小時及該厭氧菌係最少培養72小時。建議使用T3至T5的培養基。
利用含2.4毫升滅菌自來水(SICW)、2.2毫升人工唾液(AS)和0.2毫升牛血清白蛋白(BSA)的試管培養0.2毫升的受測細菌作為對照。各受測細菌的測定係分別重覆此步驟。從各對照試管取出1毫升的等量樣本並以Dey/Engley中性培養液進行10倍系列稀釋。從該系列稀釋中取從稀釋4至6之2x1毫升的等量樣本。將20~25毫升之胰蛋白豆瓊脂加入兼性細菌的稀釋等量樣本內。將20~25毫升含10%之oxyrase的Schaedler瓊脂加入厭氧菌的稀釋等量樣本內。在瓊脂固化後,將5毫升Schaedler瓊脂覆層加至含厭氧菌的平板內。
以0.2毫升的受測細菌接種第二組試管;該第二組試管含有1.1毫升之SICW、1.0毫升之AS和0.2毫升之BSA。將述於實施例2中之2.5毫升本發明配製物加至第二組各試管(受測溶液)內。在90和120秒(±5秒於各時間點)之後,從各受測溶液試管取出1.0毫升之等量的受測溶液,然後以Dey/Engley中性肉汁進行10倍的系列稀釋。在採樣點之間藉由混合進行輕微的攪動。取出10-4
稀釋係數的系列稀釋液。將取自各稀釋液的1.0毫升重複等量樣本置入150毫米的培養皿內。在30~35℃下將兼性細菌培養最少48小時,及厭氧菌培養最少72小時。在培養之後,測定其菌落形成單位(CFU),取其平均數及將該平均值乘以稀釋係數。取該值的對數10(log10
)(小數點以下兩位數)。全部細菌均進行重複的測定。
依如下方法計算在90秒時間點的降低百分比:利用介於10~100 CFU間之CFU計數的稀釋液測定各細菌在90秒時間點的對數值。列於下表1的結果為藉由下列公式(a-b)/a x 100所計算的降低百分比,其中變數“a”為接種對照溶液內之細菌數目的反對數;以及變數“b”為受測溶液內存活細菌數目的反對數。變數“a”和“b”將為取自全部重複測試在90秒時間點之各細菌的平均對數值。
表1中之“RT”指室溫,以及“2 mo @40/75”指在40℃和75%相對濕度下預測儲存2個月後的穩定性加速試驗。
此牙齒器具清潔配製物的另一獨特態樣為其具有低的界面表面張力以及特別指其形成於丙烯酸樹脂假牙上之不尋常的接觸角。液體/表面接觸角和表面張力為配製物能擴散、潮濕和最後以較低接觸角穿透表面的指標,以及表面張力為是否能潮濕/擴散和穿透的指標。具有較低接觸角和表面張力的配製物較能滲入孔隙內而有利其從孔隙構造的內部移除污穢物。目前的液體牙齒器具清潔劑具有大於約25。的丙烯酸接觸角(acrylic contact angle)。此處所述之配製物具有小於24°的丙烯酸接觸角,其大部分為介於5°和18°之間。此極低的丙烯酸接觸角歸功於該表面活性劑系統、苯甲酸、酸鹼度和油類存在的組合因素。
再一次不侷泥於特定理論之下,這些配方的酸鹼度配合有機酸(例如,苯甲酸、山梨酸、蘋果酸、檸檬酸、水楊酸和丙酸(亦稱為正丙酸))的存在下可在約2.5至5.5 pH間進一步降低在丙烯酸樹脂假牙上的接觸角。該牙齒器具清潔配製物適合的pH為在約4.0至5.5之間。本發明一具體實施例之配製物具有4.8的pH。較低之pH亦被認為可最佳化保存劑的活性、改善抗菌活性的效力以及有助於氣味的維持(藉由降低該配方的接觸角)而因此加強清新的口氣。
使用含和不含苯甲酸/苯甲酸鹽和油相之配方的丙烯酸樹脂假牙上之接觸角數據示於下列表2。下列配製物1~4的pH被調整至介於4.8和5.0之間。配製物5為調整至pH 7.5並含有3當量氫氧化鈉溶液的配製物1(全配方)。
表2中的數據顯示pH 4.8~5.0及存在油相下之含苯甲酸的配製物可具有低丙烯酸接觸角。例如配製物1之8.5。的所需低接觸角歸因上述的綜合因素。當配製物1的pH從4.9增加至7.5(配製物5)時,其在丙烯酸樹脂上的接觸角從8.5°增高至26.4°。
配合下列非限制性實施例更明確地描述屬於本發明範圍內的牙齒器具清潔配製物。
將2,101.2克的純水USP加入配備漿式混合機的一適當容器內,然後加入210.0克之99.7%的甘油USP。以中速混合該內容物。在加入下列成分之前繼續攪拌該混合物並且必需確認各成分已完全分散/溶解:120.0克之70%的山梨糖醇溶液USP;15.00克之聚乙二醇400 NF;30.00克之苯甲酸鈉NF;15.00克之椰油醯胺丙基甜菜鹼;45.00克之月桂基硫酸鈉NF;1,500克之依地酸二鈉USP;60.00克之聚氧40氫化蓖麻油USP;90.00克之Gantrez酸S97 BF(13%溶液);12.00克之糖精鈉USP;150.0克之芝麻油NF;135.0克之香料;0.300克之丁基化羥基甲苯NF;以及15.00克之苯甲酸NF。
在最後成分被溶解之後,將混合物設定在”高速”的位置,確認該漿式混合機可產生使其均勻分散的激烈攪拌速度。繼續混合約40~50分鐘。最後經脫氣之分散體的比重為1.032。
下表列舉適合用於本發明之配製物的成分。
上述的說明可完全披露本發明較佳的具體實施例。特別揭示於此處之具體實施例的改良和改善屬於下列申請專利請求項的範圍內。相信熟習本技藝之人士可利用上述的說明在不需進一步努力之下得將本發明發揮至極致。因此任何實施例僅作為說明的用途,並且無論如何本發明並非僅侷限於該範圍內。本發明具體例所主張的排他權或專屬權界定於下述的申請專利範圍內。
Claims (8)
- 一種不含酒精之充氣泡沫形式的牙齒器具清潔配製物,其係為一種含約50~99%重量/重量(w/w)之水相、約1~50% w/w之水不可混合油相的亞穩態乳劑、約0.5~6% w/w選自陰離子表面活性劑、兩性離子表面活性劑和非離子表面活性劑或其組合的表面活性劑,及約0.0-1.0% w/w選自苯甲酸鈉、苯甲酸、對羥基苯甲酸甲酯及對羥基苯甲酸丙酯或其之組合的保存劑,其中該油相具有一或多種不可混合油和一或多種風味油的組合,其中該不可混合油選自芝麻油、大豆油、菜籽油、椰子油、分餾椰子油、魚油和礦物油,並進一步含有選自由苯甲酸、山梨酸、蘋果酸、檸檬酸和水楊酸之群組所構成的有機酸。
- 如申請專利範圍第1項之配製物,其中該風味油為一種精油,選自綠薄荷、冬清、檸檬烯、薄荷醇、胡椒薄荷、沉香醇、柑橘油、茴香腦、甜茴香、桉葉醇、洋甘菊油、羅勒油、薑、迷迭香油、左旋薄荷醇、水楊酸甲酯、丁香油、龍蒿、茶樹油、瑞香酚、小豆蔻油、檸檬醛、牛至、癸醇、壬醇、癸醛、α-紫羅蘭酮、香草、大茴香子、桂皮醛、石灰或其混合物。
- 如申請專利範圍第1項之配製物,其中該配製物的pH為介於約4.0至5.5之間。
- 如申請專利範圍第1項之配製物,其具有低於24°的丙烯酸接觸角(acrylic contact angle)。
- 如申請專利範圍第4項之配製物,其中該丙烯酸接觸角為介於5°至18°之間。
- 一種於口腔外部清潔牙齒器具的方法,其包括使該牙齒器具與如申請專利範圍第1項之牙齒器具清潔配製物接觸。
- 如申請專利範圍第6項之方法,其包括以水清洗該牙齒器具的步驟。
- 一種用於傳遞如申請專利範圍第1項之牙齒器具清潔配製物的套組,其中該套組包括一噴霧器和分開包裝的牙齒器具清潔配製物。
