Abstract

Description
本發明係關於用於材料表面上控制微生物的組合物及方法,更具體而言使用最低風險農藥用於材料表面上控制微生物的組合物及方法。
材料表面上的微生物控制可藉由將抗微生物劑併入基礎材料來達成。當前實踐嚴重依賴於使用含金屬之活性物質,例如銀、鋅及/或銅。產品中最終可在使用期間或處置經處理產品後滲入環境中之「重金屬」的存在已成為環境政策及監管機構以及其他試圖影響消費者市場之機構關注的問題。
替代地,可利用有機分子(例如酚醛樹脂及唑)之使用以藉助併入處理材料表面。該等化學物質可係溫度受限的,對UV降解敏感且在欲處理整個物體時使用昂貴。此外,許多具有危險的毒性概況,且可造成可影響所處理物件美觀之不期望副作用。
因此,業內需要成本有效且具有良好效能但亦安全之用於材料表面上控制微生物的替代化學物質。
本發明係關於使用最低風險農藥用於材料表面上控制微生物的組合物及方法。
在本發明之實施例中,提供用於材料表面上控制微生物的組合物。組合物展現抗微生物性質。組合物包含聚合材料及具有抗微生物或防腐性質之公認安全(Generally-Recognized-as-Safe, GRAS)添加劑或組分。GRAS添加劑之實例尤其包括(但不限於)最低風險農藥、食品防腐劑。
在本發明之實施例中,GRAS抗微生物/防腐組分係有機酸。
在本發明之實施例中,有機酸係選自由以下組成之群:苯甲酸、山梨酸、檸檬酸及其組合。

在本發明之實施例中,提供用於賦予材料表面上之微生物控制之「公認安全」(GRAS)之組合物。縮寫字「GRAS」係由聯邦管理機構使用,以指根據優良藥品製造規範在使用時認為係「公認安全」之物質。其通常用作食品添加劑。舉例而言,該等法規包括21 CFR 182、21 CFR 184及21 CFR 186。

本發明組合物可包含一或多種添加劑。舉例而言,精油可與苯甲酸、山梨酸或其組合來組合添加以具有較強對抗真菌物種之活性。一些精油(例如,肉桂油)幾乎沒有抗細菌活性,但對真菌物種具有獨特影響。
精油之實例包括(但不限於)雪松木油、茶油(互葉白千層(Melaleuca alternifolia))、桉樹油(藍桉樹(Eucalyptus globulus)或狹葉桉(E. radiata))、丁香油(丁香(Eugenia caryophyllata))、牛至油(牛至(Origanum vulgare))、麝香草油(麝香草(Thymus vulgaris))、香葉油(香葉天竺葵(Pelargonium graveolens))、肉桂皮油(肉桂(Cinnamomum zeylanicum))、薄荷油(西洋薄荷(Mentha piperita))、檸檬香茅油(蜿蜒香茅(Cymbopogon flexuosus))、羅勒油(羅勒(Ocimum basilicum))、薰衣草油(狹葉薰衣草(Lavandula angustifolia))、檸檬油(檸檬(Citrus limon))、迷迭香油(迷迭香(Salvia rosmarinus))、香檸檬油(香檸檬(Citrus bergamia))、紫蘇油(紫蘇(Perilla frutescens))、芫荽油(芫荽(Coriandrum sativum))、香櫞油(枸櫞(Citrus medica))及其組合。較佳精油包括(但不限於)肉桂皮油(肉桂)、檸檬香茅油(蜿蜒香茅)、麝香草油(麝香草)、雪松木油、香茅油、薄荷油、迷迭香油、丁香油及其組合。
另外,可能期望控制非微生物相關氣味。有機酸(或其組合)可提供藉由其酸性性質中和基於鹼之氣味(例如氨)之能力。具有芳基之酸(例如苯甲酸)具有螯合環基臭味物質(例如吡啶)之潛力。為補充該等效應,可將額外氣味減輕化合物添加至有機酸及/或精油之組合中,以去除存在的氣味。氣味減輕化合物之添加產生可提供抗微生物及氣味控制益處之獨特摻合物。氣味減輕化合物之實例包括(但不限於)金屬氧化物、活性碳、碳酸氫鈉、碳酸鈣及蓖麻醇酸鋅。
金屬氧化物(例如,ZnO)至任何化學物質之添加均產生可提供抗微生物及氣味控制益處之獨特摻合物。
在本發明之實施例中,較佳呈聚合濃縮物形式之本發明組合物(例如,用於塗層之母料或聚合黏合劑)包含苯甲酸、山梨酸或其組合。在聚合濃縮物中,需要以組合物之重量計1%至60%或更佳5%至40%之活性濃度。在用於耐久塗層之聚合黏合劑中,基於溶液%固體,需要在0.1%至40%範圍內之濃度。若組合使用一種以上酸,則活性酸之總重量介於最終組合物重量之5%至40%之間。
本發明之組合物可視情況包含以總組合物之重量計1%至50%或更佳5%至20%之濃度的精油,且可視情況包含以總組合物之重量計1%至60%、或更佳5%至40%之濃度的氣味減輕化合物。
母料之載劑較佳係與母料欲併入之基礎材料相容之低熔點聚合物或共聚物。期望使熱敏酸活性物質之任何高熱歷史最小化。載劑將在高溫加工期間提供保護,以併入摻入較高溫度之聚合物(例如聚丙烯(PP))中。基礎聚合材料之實例包括(但不限於) PP、EMA、聚烯烴、SAN、TPU、ABS、PS及PC及PVC。舉例而言,聚丙烯(PP)係常用於製作許多消費品之聚烯烴聚合物。然後將此濃縮物與基礎原料(例如,PP)球粒-球粒摻和,並注射模製以形成具有0.05%至10%活性濃度(以最終物件之重量計)之部分。精油之最終濃度較佳為0.05%至5.0%,且氣味減輕化合物之類似濃度範圍與酸類似,例如0.05%至10%。使用甲基丙烯酸乙二酯(EMA)、丙烯-乙烯共聚物(例如Vistamaxx)、SEBS、PE及PP作為母料載劑係以下概念之實例:使用低熔點聚合物以囊封原料、熱敏性添加劑(例如,該等GRAS酸)而不會破壞其或使其揮發,且然後使用濃縮物以處理必須在高於原本可能之溫度下加工之聚合物。
直接併入該等酸用於抗微生物保護而非使用典型塗佈製程係重要改良,此乃因其消除昂貴的塗佈步驟且進一步改良產品品質;併入之抗微生物劑更耐久且不會被磨損或磨掉。在聚合物之情形中,當有機酸與基礎樹脂不相容時,可將穩定劑或增容劑添加至母料以增加產品之儲存壽命穩定性。
在期望塗層之實施例中,聚合黏合劑可由以下組成或基本上由以下組成:丙烯酸、丙烯酸酯、聚胺基甲酸酯、異氰酸酯、聚二氯亞乙烯(PVDC)、聚二氟亞乙烯(PVDF)、六氟丙烯、羧甲基纖維素(CMC)、聚乙烯醇(PVA)、聚乙酸乙烯酯(PVAc)、聚醯胺及聚醯亞胺。該等可以與任何適宜寡聚物、均聚物、共聚物或彈性體之任何組合添加。在一些實施例中,黏合劑可為水性、非水性的,或含有痕量共溶劑。聚合黏合劑之類型並無具體限制且可包括其組成可適於塗佈之任何聚合物。添加GRAS以形成以最終塗層之重量計具有0.05%至10%濃度之塗層。非限制性實例可為具有0.5%苯甲酸活性濃度之PVDC聚合黏合劑水性塗層。一旦併入,GRAS酸便結合至基材。
應用包括(但不限於)處理聚合物材料以在該等材料之表面上生成抗微生物及/或氣味益處。
與被分類為可能的毒素之現有技術不同,本發明之組合物係無害的,適用於大多數形式之聚合物製造,對聚合物加工剪切及熱穩定,且不會顯著影響最終產品性質。下表A顯示當將苯甲酸以不同含量併入不同聚合物濃縮物載劑時,PP之拉伸、撓曲及衝擊量測值變化不大。

實例1
最初工作集中於三種GRAS化學物質,即苯甲酸、山梨酸及檸檬酸之鹽。圖1顯示該等鹽之TGA圖表,其指示所有鹽對於高於300℃之溫度均係熱穩定的。
在圖1中,以10℃/min之TGA圖表顯示GRAS鹽在高達275℃之溫度下揮發。山梨酸鹽及檸檬酸鹽在高於275℃之溫度下係穩定的(未損失任何質量),而檸檬酸鹽具有大約10%水,此係為何其損失大約10%重量、然後變平穩之原因,鹽本身不會揮發。此材料穩定性及所報告之抗微生物效能使得該等化合物看起來理想地用於此應用。
每一鹽化學性質係藉由以下評估:以10濃縮物含量複合於聚烯烴載劑中,然後使用該聚合物濃縮物作為母料以各種最終用途含量注射模製PP板用於測試。將商品PP及EMA材料在雙螺桿擠出機中複合,並在35噸注射模製機中用於製作測試板。
此具有測試加工相容性以及提供樣品以測試活性效能位準之雙重目的。

出乎意料地,上述每一酸之單獨鹽(苯甲酸鈉、山梨酸鉀及檸檬酸三鈉)儘管如圖1所指示具有熱穩定性,但如表1所示,根據ISO 22196測試,在高達1%之任何活性濃度下均未產生顯著細菌效能。使用抑制區域測試(AATCC TM90),樣品亦未展現任何功效。另外,使用鹽處理該等板導致該等板因高使用含量而變得不透明。
其次,儘管酸部分(苯甲酸、山梨酸及檸檬酸)如圖2中所檢查似乎不具有足夠熱穩定性以經受PP加工,但仍對其進行研究。檸檬酸在200℃開始揮發。苯甲酸及山梨酸在大約100℃開始揮發且在TGA測試樣品達到200℃時基本上完全消失,此表明其不適於直接加工於PP中。與其直接複合於PP中以製成PP母料濃縮物,不如選擇具有較低加工溫度範圍之聚合物。EMA可與PP相容且具有極寬之加工溫度範圍。其可在低至120℃之溫度下複合且可承受超過250℃之加工溫度。以上三種酸均成功地以10%濃度複合於EMA中,以使母料濃縮物具有可忽略之揮發損失。
儘管具有TGA熱性質,但將三種EMA/酸母料在高於200°C之溫度下複合於PP中且令人驚訝地,分析指示在加工期間酸活性物質之損失極少。無論加工溫度如何,預先將酸囊封於EMA中防止活性成分因揮發而損失,並使PP部件之生產成為可能。已發現將酸複合於相容之低熔點載劑中以使GRAS酸在加工期間之揮發損失最小化係併入製程之關鍵部分。
測試板係藉由以0.5%及1%活性含量濃度注射模製於PP中製成,所有樣品均顯示較少酸添加劑之美觀影響。所生成之所有樣品均顯示良好抗細菌效能,惟檸檬酸(GCA)在較高含量下良好但在較低含量下有所下降(表2)。進一步實驗指示,認為低至1000 ppm之山梨酸(GSA)及2000 ppm至3500 ppm之苯甲酸(GBA)之效能對於併入PP係嚴重失望的。

表2顯示TR19031105 (即,GRAS酸於PP板中之測試)之對數減少結果。要求對數減少>1以顯示細菌群體之顯著差異。NZ=無區域,指示對於TM90而言係不合格的。GCA=檸檬酸,GBA=苯甲酸,GSA=山梨酸。酸未浸出,因此TM90不適於測定效能,但除最低GCA樣品外,所有樣品均通過ISO22196。在Xenon Arc及QUV儀器中測試板以確保良好UV穩定性,發現樣品之色彩僅輕微變化,此反應遠好於吡硫鎓鋅處理之樣品產生之深黃色色移或銀處理之樣品產生之棕色色移。
除在200℃下加工之外,樣品亦與PP一起在250℃下運行,以模擬該等GRAS酸在較聚丙烯高之溫度下加工之聚合物中之使用。樣品亦使用在注射模製機中之延長駐留時間(自正常30秒駐留時間至120秒駐留時間)運行,以模擬具有較長循環時間之製程,該製程將聚合物材料在一定溫度下保持延長時期,延長之循環係在200℃及240℃下運行。自該等運行中產生之樣品亦展示優良效能及極少美觀變化(在較高溫度下稍微變黑)。

使用定性方法針對真菌測試經苯甲酸及山梨酸處理之材料在未處理與經處理之間並未顯示任何差異。眾所周知,大多數塑膠不支持真菌生長且常見工業定性標準(例如AATCC TM30 (第3部分)或ASTM G21)不敏感,不足以證實效能。測試後,即使未經處理之PP或PE材料通常亦無可觀察到之真菌生長。然而,ASTM E2180方案中之定量真菌方法更為靈敏,且證實將GRAS有機酸(例如苯甲酸)併入材料中將為PE提供超過99% (2 log)之真菌生長減少且當併入EVA中時超過90%減少。將精油添加於苯甲酸或山梨酸將進一步增加所處理材料之抗真菌效能。
