該等組成物需要符合另外的標準,例如:
- 效力(特別是其中活性化合物全身作用)
- 目標動物安全性、使用者安全性
- 耐受良好
- 便利性
本發明係關於一種根據請求項1和另一實施態樣中根據請求項5之組成物、根據請求項14之方法、根據請求項15之用作為藥物之組成物及根據請求項16之用於治療及/或預防動物的寄生蟲感染之組成物。有利的實施態樣為附屬項之標的。除非上下文另有明確指示,否則彼等可自由組合。
因此本發明之一個實施態樣為一種包含泰格藍喏和1,2-異亞丙基甘油之組成物。該組成物較佳可包含1重量%至15重量%的含量之泰格藍喏;根據另外的較佳實施態樣,組成物可含有1重量
%至11重量%或7重量%至11重量%或1重量%至9.5重量%或7重量%至9.5重量%的含量之泰格藍喏。組成物視需要另外包含吡喹酮,其較佳濃度為1重量%至15重量%(較佳6重量%至9重量%)。在含有泰格藍喏和丙酮縮甘油但不含有吡喹酮的根據本發明之組成物中,該泰格藍喏的含量較佳為<10重量%,更佳<9.5重量%。
根據另一實施態樣,本發明包含一種包含吡喹酮、艾默德斯和溶劑組分之組成物另外包含泰格藍喏。這些物質具有上述段落中已經定義的結構。根據此實施態樣之組成物擴充吡喹酮和艾默德斯的殺內寄生蟲作用與泰格藍喏的殺外寄生蟲劑作用。特別地,可控制絛蟲、吸蟲、線蟲、棘頭蟲、蚤和蜱。

泰格藍喏顯示長期效力。本發明組成物因此可以4週或更長,較佳8週或更長,更佳10週或更長,特別是12週或更長的間隔施用於宿主動物。
實施例
下列實施例中將進一步描述本發明但不希望受彼等限制。丙酮縮甘油(solketal)為1,2-異亞丙基甘油。所有顯示之實施例包括丙酮縮甘油,其為了溶劑的一般穩定性已含有0.3%BHA。

狗矩頭壁蝨(Dermacentor variabilis)-對抗大鼠上之美國狗蜱若蟲的全
身體內活性:以10mg/kg的施用率在第2天>90%對抗蜱若蟲之效力。
貓櫛頭蚤(Ctenocephalides felis)-對抗大鼠上之跳蚤的全身體內活性:以10mg/kg的施用率在第2天>95%和在第9天>90%之效力。
C.體內研究內寄生蟲:沾點(spot-on)調配物對抗實驗感染之貓的貓蛔蟲(Toxocara cati)和犬絛蟲(Dipylidium caninum)之效力。
在治療之前,將16隻貓各自以貓蛔蟲(幼蟲卵(larvated eggs))和犬絛蟲之貓品系(使用經感染之貓櫛頭蚤跳蚤-口腔和局部感染)進行實驗性感染。
在第-1天,將14隻同時以貓蛔蟲和犬絛蟲專利感染之貓納入研究。將貓分成兩組,每組7隻貓。
沾點獸醫研究產品(IVP)為根據本發明之組成物,其以丙酮縮甘油為基礎含有在溶液中之10%泰格藍喏、7.94%吡喹酮和1.98%艾默德斯(w/v)。在第0天將IVP以3.2mg艾默德斯加12.7mg吡喹酮和16mg泰格藍喏/kg BW(BW=體重)劑量比(對應於0.16mL的沾點調配物/kg BW)投予至IVP組(組2)之貓。組1中之貓作為負對照組。在第10天,將貓進行安樂死並在驗屍期間回收胃腸蠕蟲。鑑定和計數蠕蟲。效力計算係根據相較於負對照組之IVP組中驗屍時回收的蠕蟲數。使用下列公式:

沒有發生不良事件(AE)。
D.體內研究外寄生蟲:沾點調配物對抗貓之實驗感染的蜱和跳蚤之效力。
在SD-4該研究中包括十二隻貓。在SD-1,以蓖麻豆蝨(Ixodes ricinus)蜱實驗感染貓,在SD 0,其在沒有移除下計數(用於組分配)及在SD 2被移去並計數(治療效力)。在SD 0,以沾點施用IVP一次而以每kg體重14mg泰格藍喏+3mg艾默德斯+12mg吡喹酮之劑量治療六隻貓。六隻貓作為未治療的對照組。
該IVP以丙酮縮甘油為基礎含有在溶液中之10%(m/V)泰格藍喏、8.58%(m/V)吡喹酮和2.14%(m/V)艾默德斯。
每兩週以蜱和跳蚤實驗感染貓。藉由比較治療組相對於對照組的蜱和跳蚤計數確定IVP之效力。每天觀察一般健康狀況。
*每隻貓以20雌性和20雄性蓖麻豆蝨(Ixodes ricinus)蜱感染。**每隻貓以100貓櫛頭蚤跳蚤感染。
單次局部治療後,IVP對貓的耐受性非常好。在此研究中,沒有與IVP治療相關的不良事件。
Claims (19)
- 一種組成物,其包含泰格藍喏(tigolaner)和1,2-異亞丙基甘油。
- 根據請求項1或2之組成物,另外包含吡喹酮(praziquantel)。
- 一種組成物,其包含吡喹酮、艾默德斯(emodepside)和溶劑組分,其特徵在於該組成物另外包含泰格藍喏。
- 根據請求項5之組成物,其中該溶劑組分包含1,2-異亞丙基甘油和其中該組成物的含水量為最多5重量%,較佳為最多3重量%,更佳為最多2重量%。
- 根據前述請求項中之一項之組成物,其中該溶劑組分僅包含1,2-異亞丙基甘油。
- 根據前述請求項中之一項之組成物,其另外包含抗氧化劑。
- 根據請求項9之組成物,其中該抗氧化劑為丁基羥基甲氧苯(BHA)及/或丁基羥基甲苯(BHT)。
- 根據前述請求項中之一項之組成物,其另外包含酸。
- 根據請求項11之組成物,其中該酸為乳酸。
- 一種製造根據請求項5之組成物之方法,其包含將吡喹酮、艾默德斯和泰格藍喏溶解在溶劑組分中的步驟。
- 一種用作為藥劑之根據請求項1至13中之一項之組成物。
- 一種用於治療及/或預防動物的寄生蟲感染之根據請求項1至13中之一項之組成物。
- 根據請求項16使用之組成物,其中該等動物為貓。
- 根據請求項16或17使用之組成物,其中該等寄生蟲為內寄生蟲和外寄生蟲。
