Abstract
Способ рекомендован дл внедрени в практическое здравоохранение .The invention relates to medicine and can be used in the manufacture of an artificial vascular prosthesis. The purpose of the invention is to prevent complications of prosthetics by enhancing the thrombogenic properties of neointima. with endothelial cells germinated on it. The method was applied in 11 experiments; in all cases, the inner surface of the implanted prosthesis was coated with endothelial cells without admixtures of smooth muscle cells and signs of stenosis of the vascular anastomoses. The method is recommended for implementation in practical health care.
Description
The invention relates to medicine, namely to vascular transplantation, and can be used to make artificial blood vessels.
The purpose of the invention is to prevent complications of vascular prosthetics by increasing the atrombogenic properties of neointima.
The method is carried out as follows.
In the recipient, a section of the saphenous vein is excised, the endothelium is peeled off, and the endothelial cells thus obtained are germinated for at least 24 hours on a collagen film. The artery segment is resected, replaced with an artificial vascular prosthesis, after which the outer surface of the prosthesis is wrapped with a collagen film with endothelial cells previously passaged on it, which ensures that the inner surface of the prosthesis is covered with endothelium cells without the addition of smooth muscle and other cells.
MRKZExample. ZAZ days before the main experiment, the dog had an excised vein, placed it in a special solution and delivered to the culture box. Endothelial cells were shed from the inner surface of the vein and applied to a previously prepared collagen film, the dimensions of which were determined in accordance with the dimensions of the vascular prosthesis, and the thickness was 50–200 µm. Cells deposited on the film were cultured on it for 2-3 days (depending on their initial concentration). After the formation of a monolayer in the same dog, the carotid artery segment was resected, the diastasis of the artery was replaced with an artificial vessel.
