Claims (16)
1. Способ индукции формирования эндокринных кластеров, включающий культивирование панкреатических эндокринных клеток с агонистом рецептора сфингозина-1.
2. Способ по п.1, в котором для обработки панкреатических эндокринных клеток в качестве агониста рецептора сфингозина-1 используют сфингозин-1-фосфат (S1P).
3. Способ по п.1, в котором панкреатические эндокринные клетки получают путем поэтапной дифференцировки плюрипотентных стволовых клеток.
4. Способ по п.3, в котором плюрипотентные стволовые клетки представляют собой плюрипотентные стволовые клетки человека.
5. Способ по п.1, где способ дополнительно содержит:
дифференцировку плюрипотентных стволовых клеток в конечные клетки энтодермы;
дифференцировку конечных клеток энтодермы в клетки первичной кишечной трубки;
дифференцировку клеток первичной кишечной трубки в клетки передней кишки;
дифференцировку клеток передней кишки в клетки-предшественники панкреатических клеток;
дифференцировку клеток-предшественников панкреатических клеток в клетки панкреатической энтодермы; и
дифференцировку клеток панкреатической энтодермы в панкреатические эндокринные клетки.
6. Способ по п.5, где плюрипотентные стволовые клетки представляют собой плюрипотентные стволовые клетки человека.
7. Способ по п.5, где способ дополнительно включает обработку клеток панкреатической энтодермы эфриновым лигандом.
8. Способ по п.7, где эфриновый лиганд представляет собой эфрин А3 или эфрин А4.
9. Способ по п.5, где способ дополнительно включает обработку клеток панкреатической энтодермы активином А или активином С.
10. Способ по п.5, где способ дополнительно включает обработку клеток панкреатической энтодермы семафорином 3А или эпигеном.
