Abstract
1. Фармацевтическая композиция замедленного высвобождения для местного применения на поверхность кожи, содержащая:- фармацевтически активный агент, включающий один или несколько стероидов,-от 0,01% до 5% по массе от общей массы фармацевтической композиции эфира жирной кислоты,- воду,- С2-С6 моноспирт,- жирную кислоту, и- от 0,05% до 5% по массе гелеобразующего вещества,где отношение массы эфира жирной кислоты в композиции ко всему активному агенту в указанной композиции составляет, по меньшей мере, 4:1, эфир отношение жирной кислоты: активный агент, предпочтительно от 4:1 до 20:1.2. Композиция по п.1, которая дополнительно содержит сорастворитель, предпочтительно пропиленгликоль.3. Композиция по п.2, в которой сорастворитель присутствует в количестве от 0,01% до 7% по массе от общей массы фармацевтической композиции, предпочтительно от 3% до 7% по массе.4. Композиция по п.1, в которой эфир жирной кислоты выбирают из группы, состоящей из этилолеата, изопропилолеата, изопропилмиристата, изопропилизостеарата, изопропилпальмитата, этилоктаноата, этилдодеканоата, этиллинолеата, этилпальмитолеата, этилизостеарата и этиллинолената.5. Композиция по п.1, в которой эфир жирной кислоты представляет собой изопропилмиристат.6. Композиция по п.1, в которой эфир жирной кислоты представляет собой этилолеат.7. Композиция по п.1, в которой эфир жирной кислоты представляет собой эфир, который может образоваться при взаимодействии жирной кислоты, входящей в состав композиции, со спиртом.8. Claims (22)
1. Фармацевтическая композиция замедленного высвобождения для местного применения на поверхность кожи, содержащая:
- фармацевтически активный агент, включающий один или несколько стероидов,
-от 0,01% до 5% по массе от общей массы фармацевтической композиции эфира жирной кислоты,
- воду,
- С2-С6 моноспирт,
- жирную кислоту, и
- от 0,05% до 5% по массе гелеобразующего вещества,
где отношение массы эфира жирной кислоты в композиции ко всему активному агенту в указанной композиции составляет, по меньшей мере, 4:1, эфир отношение жирной кислоты: активный агент, предпочтительно от 4:1 до 20:1.
2. Композиция по п.1, которая дополнительно содержит сорастворитель, предпочтительно пропиленгликоль.
3. Композиция по п.2, в которой сорастворитель присутствует в количестве от 0,01% до 7% по массе от общей массы фармацевтической композиции, предпочтительно от 3% до 7% по массе.
4. Композиция по п.1, в которой эфир жирной кислоты выбирают из группы, состоящей из этилолеата, изопропилолеата, изопропилмиристата, изопропилизостеарата, изопропилпальмитата, этилоктаноата, этилдодеканоата, этиллинолеата, этилпальмитолеата, этилизостеарата и этиллинолената.
5. Композиция по п.1, в которой эфир жирной кислоты представляет собой изопропилмиристат.
6. Композиция по п.1, в которой эфир жирной кислоты представляет собой этилолеат.
7. Композиция по п.1, в которой эфир жирной кислоты представляет собой эфир, который может образоваться при взаимодействии жирной кислоты, входящей в состав композиции, со спиртом.
8. Композиция по п.1, в которой эфир жирной кислоты не является эфиром, который может образоваться при взаимодействии жирной кислоты, входящей в состав композиции, со спиртом, находящимся в композиции.
9. Композиция по п.1, в которой эфир жирной кислоты присутствует в количестве от 0,05% до 2,4% по массе, от общей массы фармацевтической композиции, предпочтительно от 0,1% до 2,2% по массе.
10. Композиция по п.1, в которой жирная кислота является С8-С22 жирной кислотой, которую предпочтительно выбирают из группы, состоящей из каприновой кислоты, лауриновой кислоты, миристиновой кислоты, пальмитиновой кислоты, стеариновой кислоты, олеиновой кислоты, изостеариновой кислоты, пальмитолеиновой кислоты, линолеиновой кислоты и линоленовой кислоты, и более предпочтительно жирная кислота представляет собой олеиновую кислоту.
11. Композиция по п.1, в которой жирная кислота присутствует в количестве от 0,01% до 5% по массе от общей массы фармацевтической композиции, предпочтительно от 0,05% до 3,5% по массе, и более предпочтительно от 1,0% до 3,0% по массе.
12. Композиция по п.1, содержащая 2% этилолеата в качестве эфира жирной кислоты, 2% олеиновой кислоты в качестве жирной кислоты, и 5% пропиленгликоля в качестве сорастворителя, все по массе от общей массы фармацевтической композиции.
13. Композиция по п.1, содержащая 0,3% этилолеата в качестве эфира жирной кислоты, 0,3% олеиновой кислот в качестве жирной кислоты, и 0,75% пропиленгликоля в качестве сорастворителя, все по массе от общей массы фармацевтической композиции.
14. Композиция по п.1, в которой фармацевтически активный агент содержит один или несколько стероидов, которые выбирают из группы, состоящей из эстрогенов, анти-эстрогенов, андрогенов, анти-андрогенов и прогестинов.
15. Композиция по п.1, в которой фармацевтически активный агент выбирают из одного или нескольких из эстрадиола и прогестерона, и эфир жирной кислоты представляет собой этилолеат.
16. Композиция по п.1, в которой фармацевтически активный агент выбирают из одного или нескольких из тестостерона и дигидротестостерона (DHT), и эфир жирной кислоты выбирают из этилолеата и изопропилмиристата.
17. Композиция по п.1, в которой активный агент присутствует в количестве от 0,01% до 5% по массе, от общей массы фармацевтической композиции.
18. Композиция по п.1, в которой С2-С6 моноспирт выбирают из группы, состоящей из этанола, н-пропанола, изопропанола, н-бутанола, изобутанола, трет-бутанола, и их смесей, предпочтительно С2-С6 моноспирт является этанолом.
19. Композиция по п.1, в которой С2-С6 моноспирт присутствует в количестве от 10% до 90% по массе, от общей массы фармацевтической композиции, предпочтительно от 20% до 80% по массе, и более предпочтительно от 45% до 75% по массе.
20. Способ обеспечения замедленного высвобождения фармацевтически активного агента через кожу субъекта, который включает поверхностное применение на кожу субъекта фармацевтической композиции, содержащей:
- фармацевтически активный агент, включающий один или несколько стероидов,
- от 0,1% до 20% по массе, от общей массы фармацевтической композиции, эфира жирной кислоты,
- воду,
- С2-С6 моноспирт,
- жирную кислоту; и
- от 0,05% до 5% по массе гелеобразующего вещества,
где отношение массы эфира жирной кислоты в композиции ко всему активному агенту в указанной композиции составляет, по меньшей мере, 4:1, отношение эфир жирной кислоты/активный агент, предпочтительно от 4:1 до 20:1.
21. Способ по п.20, в котором применяется композиция по любому из пунктов 2-19.
22. Способ по п.20 или 21, в котором замедленное высвобождение фармацевтически активного агента через кожу наблюдается, по меньшей мере, 24 часа после указанного применения, предпочтительно, по меньшей мере, 36 часа после указанного применения, и более предпочтительно по меньшей мере, 48 часа после указанного применения.
