Claims (20)
1. Нуклеиновая кислота, которая кодирует быстросозревающий хромо- или флуоресцирующий мутант хромопротеина или флуоресцирующего белка книдарии (Cnidaria) или его мутанта.
2. Нуклеиновая кислота по п. 1, где указанная нуклеиновая кислота кодирует хромопротеин или флуоресцирующий белок из небиолюминесцирующих видов книдарии.
3. Нуклеиновая кислота по п. 2, где указанные небиолюминесцирующие виды книдарии относятся к классу Anthozoa.
4. Нуклеиновая кислота по п. 3, где указанный класс Anthozoa относится к Discosoma.
5. Нуклеиновая кислота по п. 4, где указанная нуклеиновая кислота кодирует мутант [белка] DsRed.
6. Нуклеиновая кислота по п. 5, где указанная нуклеиновая кислота кодирует продукт, имеющий точковую мутацию по меньшей мере в одном из положений - положении 2, положении 5, положении 6, положении 21, положении 41, положении 42, положении 44 и положении 117 по сравнению с [белком] DsRed дикого типа.
7. Нуклеиновая кислота по п. 6, где указанный продукт представляет собой продукт, имеющий точковую мутацию по меньшей мере в одном из положений - положении 145 и положении 217.
8. Нуклеиновая кислота по п. 7, где указанная нуклеиновая кислота содержит по меньшей мере 10 остатков последовательностей SEQ ID NO: 01 или 02.
9. Фрагмент нуклеиновой кислоты, выбранный в соответствии с пп. 1-7.
10. Конструкция, содержащая вектор и нуклеиновую кислоту по пп. 1-9.
11. Экспрессирующая кассета, содержащая
(a) область инициации транскрипции, функциональную в экспрессирующем хозяине;
(b) нуклеиновую кислоту по пп. 1-9; и
(c) область терминации транскрипции, функциональную в указанном экспрессирующем хозяине.
12. Клетка или ее предшественник, содержащая экспрессирующую кассету по п. 11 как часть экстрахромосомного элемента или интегрированную в геном хозяйской клетки в результате введения указанной экспрессирующей кассеты в указанную хозяйскую клетку.
13. Способ продуцирования хромопротеина и/или флуоресцирующего белка, где указанный способ включает в себя выращивание клетки по п. 12, в результате чего экспрессируется указанный белок или протеин; и выделение указанного белка или протеина, существенно свободного от других белков или протеинов.
14. Белок или протеин или фрагмент белка или протеина, кодируемый нуклеиновой кислотой по пп. 1-9.
15. Антитело, специфически связывающееся с белком или протеином по п. 14.
16. Трансгенная клетка или ее предшественник, содержащая трансген, который представляет собой нуклеиновую кислоту по пп. 1-9.
17. Трансгенный организм, содержащий трансген, который представляет собой нуклеиновую кислоту по пп. 1-9.
18. Усовершенствование применения, в котором используется хромопротеин или флуоресцирующий белок, где указанное усовершенствование включает в себя применение белка или протеина по п. 14.
19. Усовершенствование применения, в котором используется нуклеиновая кислота, кодирующая хромопротеин или флуоресцирующий белок, где указанное усовершенствование включает в себя применение нуклеиновой кислоты по пп. 1-9.
20. Набор, содержащий нуклеиновую кислоту по пп. 1-9.
