Abstract
Description
Claims (21)
- 폴리아제티딘으로의 교차결합에 의하여 생물학적 물질을 고정시키는 방법으로서, 생물학적 물질을 폴리아제티딘과 수성혼합하는 단계, 그 결과의 혼합물을 응집시키는 단계, 응집생성물을 탈수하는 단계, 탈수된 생성물을 하위 분할하는 단계 및 하위 분할된 물질을 경화하여 입자형태의 고정생성물을 얻는 단계의 연속 단계로 이루어진다.
- 제1항에 있어서, 폴리아제티딘의 양이 생물학적 물질의 건조물 중량의 약 0.1% 내지 약 10%, 바람직하게는 약 0.5% 내지 약 5%인 것을 특징으로 하는 방법.
- 제1항 또는 2항에 있어서, 글루타르알데히드가 탈수단계가 실시되기 전에 폴리아제티딘/생물학적 물질 혼합물에 혼입되는 것을 특징으로 하는 방법.
- 제3항에 있어서, 글루타르알데히드의 양이 생물학적 물질중의 건조물의 약 40% 이하, 바람직하게는 약 15% 이하인 것을 특징으로 하는 방법.
- 제3항에 있어서, 글루타르알데히드가 생물학적 물질과 수성 혼합된 후, 글루타르알데히드/생물학적 물질 혼합물이 그것과 폴리아제티딘과의 혼합전에 보유되는 것을 특징으로 하는 방법.
- 제3항에 있어서, 글루타르알데히드와 폴리아제티딘이 생물학적 물질과 수성혼합되고 그런 다음 글루타르알데히드/폴리아제티딘/생물학적 물질 혼합물이 그의 탈수전에 보유되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 탈수단계전에 바람직하게는 약 100% 건조물 이하의 폴리에틸렌이민(가장 바람직하게는 약 10% 이하), 알부멘(가장 바람직하게는 약 50% 이하), 젤란틴 (가장 바람직하게는 약 50% 이하), 또는 카르복시메틸셀룰로스(가장 바람직하게는 약 10% 이하)(모든 %는 생물학적 물질중의 건조물에 관련한 중량%이다)를 혼합하는 단계를 추가로 포함하는 것을 특징으로 하는 방법.
- 제1항에 있어서, 유효량의 응집제가 탈수단계전에 첨가되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 생물학적 물질이 수성분산액 또는 용액의 형태로 첨가되는 것을 특징으로 하는 방법.
- 제1항에 있어서, 생물학적 물질이 바람직하게는 식물 또는 미생물기원의 전체세포 또는 완전히 또는 부분적으로 파괴된 세포로부터의 세포덩어리를 포함하는 것을 특징으로 하는 방법.
- 제10항에 있어서, 고정된 물질이 바람직하게는 중금속의 흡착제거에 사용될 수 있는 것을 특징으로 하는 방법.
- 제1항에 있어서, 생물학적 물질이 효소를 포함하는 것을 특징으로 하는 방법.
- 제12항에 있어서, 효소가 글루코스이성화제인 것을 특징으로 하는 방법.
- 제13항에 있어서, 효소가 스트렙토마이세스종, 가장 바람직하게는 스트렙토마이세스뮤리누스로부터, 바실러스 코아귤란스로부터 또는 악티노 플라네스 미오리엔시스로부터 유도되는 것을 특징으로 하는 방법.
- 제12항에 있어서, 효소가 페니실린 아실라제인 것을 특징으로 하는 방법.
- 제15항에 있어서, 효소가 푸사리움종, 바람직하게는 푸사리움 안구이오이데스, 푸사리움 아르길라세움, 푸사리움 아베나세움, 푸사리움 불비게늄, 푸사리움 코에룰레움, 푸사리움 쿨로룸, 푸사리움 에퀴세티, 푸사리움 라테리티움, 푸사리움 미니뭄, 푸사리움 모노리포르메, 푸사리움 옥시스포룸, 푸사리움 삼부시눔, 푸사리움 세미섹툼, 푸사리움 솔라니 또는 푸사리움 슬푸레움으로부터 유도되는 것을 특징으로 하는 방법.
- 제12항에 있어서, 효소가 바람직하게는 로도코커스종(바람직하게는 로도코커스 에리트로폴리스), 슈도모나스종 또는 브레비박테리움종으로부터의 니트릴라제인 것을 특징으로 하는 방법.
- 제12항에 있어서, 효소가 시안화물 히드라타제인 것을 특징으로 하는 방법.
- 제1항에 있어서, 생물학적 물질이 조효소인 것을 특징으로 하는 방법.
- 제1항에 있어서, 생물학적 물질이 항체인 것을 특징으로 하는 방법.
- 제1항의 방법에 의하여 제조된 고정된 효소의 효소-촉매 과정에서의 이용.
