Description

외용 소염 진통제
본 발명은 유효 성분으로서 2-(2-플루오로-4-비페닐릴)프로피온산을 함유한 외용(外用) 소염 진통제에 관한 것이다.
외용 소염 진통제로서는 살리실산 메틸 등의 살리실산 에스테르가 주성분으로 널리 사용되어 왔다.
또한, 2-(2-플루오로-4-비페닐릴)프로피온산이 산성 비스테로이드성 소염진통제로서 우수한 약제 효과를 나타내었고, 또 외용 소염 진통제로서 사용되어 왔다. 그러나, 살리실산 에스테르 화합물은 소염 진통 효력은 있으나, 소망의 약제 효과를 얻는데 다량의 용량을 요하는 결점이 있다. 또한, 2-(2-플루오로-4-비페닐릴)프로피온산은 강력한 소염 진통 효력을 나타내지만, 한편으로는 위험한 위장장애를 수반하며 그 안전 범위가 좁다는 결함들에 직면하게 된다.
외용 소염 진통제로서의 2-(2-플루오로-4-비페닐릴)프로피온산의 부작용을 감소하기 위한 다년간의 광범위한 연구 결과, 본 발명을 완성하게 되었다.
본 발명의 목적은 2-(2-플루오로-4-비페닐릴)프로피온산을 경피 흡수시켜서 국소제 농도를 강화하고, 위장장애와 기타 마찬가지의 장애들과 같은 일반적인 부작용들을 완화하며, 소염 진통 효과가 효율적으로 나타나는 2-(2-플루오로-4-비페닐릴)프로피온산 제제를 제공하는데 있다.
즉, 본 발명은 유효 성분으로서 일반식
로 표시되는 2-(2-플루오로-4-비페닐릴)프로피온산과 기제(基劑)로 된 외용 소염 진통제를 목적으로 하고 있다.
본 발명의 외용 소염 진통제는 염증 피부에 발랐을 때에 종래의 경구 소염 진통제에서와 같은 위장장애와 기타 마찬가지 장애들과 같은 일반적인 부작용들이 없이 염증 조직내로 유효 성분이 직접적으로 침투하고, 염증 조직내에서 고농도가 장시간 유지되어 지속적인 소염 진통 효과를 효율적으로 나타내는 극히 유용한 소염 진통제인 것이다.
소염 진통제 중의 유효 성분 용량은 1회 사용에 2~200mg, 바람직하게는 10~80mg이다.
본 발명에서 사용하는 기제(基劑)로서는 친유성 기제와 친수성 기제들이 있다. 친유성 기제로서는 바셀린, 유동 파라핀, 플라스티베이스(plastibase), 실리콘, 식물성 기름, 천연 고무, 합성 고무, 수지, 폴리부텐 등이 있다. 유효 성분을 상기한 유성(油性) 기제들과 직접 혼합할 수 있다. 그러나, 테르펜계 화합물, 고급 지방산 에스테르 등의 유기 화합물과 함께 본 발명의 유효 성분을 사용함으로써 피부로부터 흡수된 유효 성분의 특성이 유기 화합물의 작용으로 경피 흡수 촉진을 향상시키는 것이 유리하다.
경피 흡수 촉진제로서는, 예컨대 박하유, 유우칼리유 등의 테르펜계 화합물, 미리스트산 이소프로필 등의 고급 지방산 수지, 탄화 수소 등이 있다.
또한, 외용 소염 진통제는 피부에 사용될 수 있는 모든 의약 제제로서 사용할 수가 있다. 즉, 연고, 분말, 액제 등의 제형으로 사용함으로써 본 발명의 소염 진통제는 제제로부터 계속적으로 방출되어 피부로부터 효율적으로 흡수되며, 또한 염증 부위에 선택적으로 도달하여 목적하는 소염 진통 효과를 나타낼 수 있는 것이다.
이하, 실시예와 관련하여 본 발명을 보다 상세하게 설명하지만, 본 발명은 이에 국한되는 것은 아니다.
실시예 1 (본 발명의 외용 소염 진통 조성물의 제조)
10g의 박하유에 5g의 2-(2-플루오로-4-비페닐릴)프로피온산을 가해서 된 용액을 다음의 제1표에 기재한 조성을 가진 85g의 친유성 기제와 균질이 되게 혼합하여 본 발명의 외용 진통 조성물을 제조하였다.

실시예 2
10g의 유우칼리유에 5g의 2-(2-플루오로-4-비페닐릴)프로피온산을 가해서 된 용액을 85g의 플라스티베이스와 균질이 되게 혼합하여 본 발명의 외용 소염 진통제를 제조하였다.
실시예 3 (쥐의 등으로 부터의 경피 흡수)
본 발명의 외용 소염 진통 조성물을 쥐의 등에 발라서 혈액 농도중의 유효 성분의 시간에 따른 변화를 측정하였다. 그 결과는 다음의 제2표에 나와 있다.

본 발명의 유효 성분의 혈액 농도는 바른 후 2시간에 최고를 나타내었고, 이어서 점차로 감소되는 경향이 있었다.
실시예 4 (쥐의 등에서의 카라지이닌 유도 부종 억제 활성)
쥐 8마리를 일군으로 사용하여 쥐의 등의 털을 실험 하루전에 깎은 다음, 본 발명의 외용 소염 진통 조성물을 털을 깎은 등에 발랐다. 2시간 후에 소염 진통 조성물을 제거하고 1% 카라지아닌(carrageenin)용액 0.05㎖을 쥐의 등의 중심선의 한쪽에, 그리고 다른쪽에는 생리 식염수 0.05㎖를 각각 경피 주사하였다. 주사 직후에 시험할 조성물을 발랐다. 카라지이닌 투여후 3시간만에 출혈시켜 죽게 하고 그 피부를 절제하였다. 부종 부위와 대조 부위를 가죽 펀칭 머시인(15mm의 구멍 직경)으로 펀칭하여 중량을 달았다. 생리 식염수를 주사한 대조군의 중량을 부종 부위의 중량에서 뺀 차이를 부종의 무게로 하였다. 부종 억제율을 다음 식으로 구하였다.

그 결과는 제3표에 나와 있다.

위에 나온 실험 결과들로부터 입증되고 있는 것은 본 발명의 외용 소염 진통 조성물이 현저하게 카라지이닌 유도 부종을 억제한다는 점인데, 이것으로부터 경피 흡수에 의한 본 발명의 유효 성분의 소염 진통 효과를 알 수 있다.

Claims (2)

  1. 아래의 구조식으로 나타내어지는 2-(2-플루오로-4-비페닐릴)프로피온산을 유효성분으로하고, 테르펜계 화합물을 경피 흡수 촉진제로 하여, 이들과 친유성 기제로 된 외용 소염 진통제로서, 외용 소염 진통제 중의 유효성분의 양은 적용 1회당 2mg 내지 200mg이고, 첩부제의 형태인 것을 특징으로 하는 외용 소염 진통제.

  2. 제1항에 있어서, 친유성 기제가 바셀린, 유동 파라핀, 플라스티베이스, 실리콘, 식물성 기름, 천연 고무, 합성 고무, 수지 또는 폴리부텐인 외용 소염 진통제.
