- 【特許請求の範囲】 1.ユビキチン-S27a 遺伝子のプロモーター配列および/または他の調節配列を 含む核酸分子。 2.前記のプロモーター配列および/または他の調節配列がハムスターのユビキ チン-S27a 遺伝子に由来する、請求項1に記載の核酸分子。 3.前記のプロモーター配列および/または他の調節配列が図5の配列に含まれ る、請求項1または2のいずれか1項に記載の核酸分子。 4.前記のプロモーター配列が図5の配列の-161〜-45 位置に対応する配列に含 まれる、請求項3に記載の核酸分子。 5.図5の配列の1塩基、2塩基、3塩基またはそれ以上の塩基の置換、挿入ま たは欠失によって生じるプロモーター配列であって、それらの置換、挿入または 欠失によりプロモーター活性の顕著な低下を起こしていないプロモーター配列を 含む核酸分子。 6.前記のプロモーター配列に機能的に結合した1以上のエンハンサーを含む、 請求項1〜5のいずれか1項に記載の核酸分子。 7.前記のプロモーターの転写活性を制御し得る調節配列を含むことを特徴とす る、請求項1〜6のいずれか1項に記載の核酸分子。 8.前記のプロモーター配列および/または調節配列が遺伝子に機能的に接続さ れていることを特徴とする、請求項1〜7のいずれか1項に記載の核酸分子。 9.前記の遺伝子が、組織プラスミノーゲン活性化因子、第二世代組織プラスミ ノーゲン活性化因子、インターフェロン、腫瘍壊死因子、エリスロポイエチン、 顆粒球コロニー刺激因子、マンガンスーパーオキシドジスムターゼ、免疫グロブ リン鎖、免疫グロブリン鎖の可変領域、ヒト化免疫グロブリン鎖、ヒト化免疫グ ロブリン鎖の可変領域、一本鎖抗体および/またはCD44バリアントに対する特異 的抗体をコードする遺伝子であることを特徴とする、請求項8に記載の核酸分子 。 10.発現ベクターであることを特徴とする請求項1〜9のいずれか1項に記載の 核酸分子。 11.前記のプロモータが組換えによって真核宿主細胞、好ましくはハムスター細 胞、もっとも好ましくはCHO 細胞のゲノム中に組み込まれ得ることを特徴とする 、請求項1〜10のいずれか1項に記載の核酸分子。 12.増幅促進配列を含むことを特徴とする、請求項1〜11のいずれか1項に記 載の核酸配列。 13.請求項1〜12のいずれか1項に記載の核酸分子を導入した宿主細胞。 14.ハムスターUb/S27a 遺伝子の前記プロモーターと結合した異種遺伝子産物の 遺伝子を含む核酸分子を導入された宿主細胞。 15.請求項1〜11のいずれか1項に記載の核酸分子を真核宿主細胞に導入し、 その宿主細胞を培養して合成された遺伝子産物を単離することを特徴とする、真 核宿主細胞、好ましくはハムスター細胞、もっとも好ましくはCHO 細胞において 異種遺伝子産物を調製する方法。 16.前記の異種遺伝子産物がユビキチン-S27a 遺伝子のプロモーター配列および /または調節配列の制御下で発現していることを特徴とする、真核宿主細胞、好 ましくはハムスター細胞、もっとも好ましくはCHO 細胞において異種遺伝子産物 を調製する方法。 17.前記のプロモーター配列および/または調節配列が図5 に含まれていること を特徴とする、請求項16に記載の方法。 18.前記のプロモーター配列が図5 の位置-161〜-45 に対応する配列中に含まれ ることを特徴とする、請求項17に記載の方法。 19.前記の異種遺伝子が、組織プラスミノーゲン活性化因子、第2 世代組織プラ スミノーゲン、インターフェロン、腫瘍壊死因子、エリスロポイエチン、顆粒球 コロニー刺激因子、マンガンスーパーオキシドジスムターゼ、免疫グロブリン鎖 、免疫グロブリンの可変領域、ヒト化免疫グロブリン鎖、ヒト化免疫グロブリン 鎖の可変領域、1本鎖抗体および/またはCD44バリアントに特異的な抗体をコー ドすることを特徴とする、請求項15〜18のいずれか1項に記載の方法。 20.ハムスターの強い同種プロモーター 21.ヘルペス単純ウイルスのチミジンキナーゼプロモーターよりも強い転写活性 を有することを特徴とする、請求項20に記載のプロモーター。 22.少なくともSV40のプロモーターと同程度の強さの転写活性を有することを特 徴とする、請求項20に記載のプロモーター。 23.以下の少なくとも1つの性質を有することを特徴とする、請求項20〜22 のいずれか1項に記載のプロモーター:GC- リッチ配列領域、Sp1 結合領域、ポ リピリミジンエレメント、CAATボックスの欠如、TATAボックスの欠如。 24.Sp1 結合部位を有し、TATAボックスを有さないことを特徴とする、請求項2 0〜23のいずれか1項に記載のプロモーター。 25.図5の配列含まれることを特徴とする、請求項21〜24のいずれか1項に 記載のプロモーター。 26.図5の位置-161〜-45 に対応する配列中に含まれることを特徴とする請求項 25のプロモーター。 27.遺伝子産物が請求項21〜26のいずれか1項に記載したプロモーターの制 御下で発現されていることを特徴とする、ハムスター細胞、好ましくはCHO 細胞 中で異種遺伝子産物を発現させる方法。 28.ハムスター細胞、好ましくはCHO 細胞中で異種遺伝子産物を調製するための 、前記全ての請求項のいずれか1項に記載したプロモーターの使用。 29.ハムスターのUb/S27a 遺伝子。 30.図1の配列、または図1の配列の核酸分子とストリンジェントな条件下でハ イブリダイズする配列を有する請求項29に記載のUb/S27a 遺伝子。
