Description
本発明のニコチニル化合物(I、II、III、IV、V、VI、VII、VIII、およびIX)を用いる本方法は、塩の形態をとってもよい。用語「塩」は、本発明の方法のニコチニル化合物(I、II、III、IV、V、VI、VII、VIII、およびIX)である遊離酸または遊離塩基の付加塩を含む。用語「薬学的に許容可能な塩」は、医薬用途において有用性を与える範囲内の毒性プロフィールを有する塩を指す。
本明細書中に記載される方法は、高用量の1つまたは複数のニコチニル化合物(I、II、III、IV、V、VI、VII、VIII、および/またはIX)もしくはそれらの塩を単独で、あるいは1つまたは複数のビタミン(X、XI、XII、および/またはXIII)と組み合わせて、例えばピルの形態で、対象に、毎日、または隔日、または週一回、投与することを含んでよい。高用量の1つまたは複数のニコチニル化合物(I、II、III、IV、V、VI、VII、VIII、および/またはIX)もしくはそれらの塩が単独で、あるいは1つまたは複数のビタミン(X、XI、XII、および/またはXIII)と組み合わされて、対象に毎日投与される実施形態において、1つまたは複数のニコチニル化合物(I、II、III、IV、V、VI、VII、VIII、および/またはIX)もしくはそれらの塩が単独で、あるいは1つまたは複数のビタミン(X、XI、XII、および/またはXIII)と組み合わされて、1日1回投与されてよい。他の実施形態では、1日2回、または3回投与される。
ニコチンアミドリボシド(NR、I)はまた、乳中で自然に見られる。図2は、ニコチンアミドリボシド(NR、I)が、店で購入した(牛)乳中に存在することを示している。図2Bおよび図2Cは、ニコチンアミドリボシド(NR、I)を既知の量で乳サンプルに加えた後のニコチンアミドリボシド(NR、I)の検出を示すコントロールクロマトグラムである。これらのコントロールクロマトグラムは、ニコチンアミドリボシド(NR、I)が乳に加えられてから、定量的に回収され得、重大な分解も、市販の乳とのニコチンアミドリボシド(NR、I)の不和合性の証拠もないことを実証している。1%ニコチンアミドリボシド(NR、I)の回収を算出すると、100%近くであった。当該結果を得るのに用いた実験方法は、以下の通りであった:乳をアセトニトリルで1:1に希釈した。その後、遠心分離を実行して、あらゆる沈殿物を除去して、上清を、標準的な方法を用いるHILIC/HPLC/UVを用いて分析した。
ヒト乳児のモデルとしての子ブタ腸における脆弱な神経発達の保護に果たすニコチンアミドリボシド(NR、I)の役割
ヒト乳児は、発生的に未発達で生まれてくる。これはとりわけ、3分の1を超える脳の成長が、出生後最初の6ヵ月で起こる神経組織に当てはまる。脳の成長は、栄養に関して広範囲に及ぶ要求を突き付けることが知られており、ヒト乳は、全基質を提供して、脳の発達を組み立てて活気付け(fuel)なければならない。現在の研究では、脳の最適な成長および発達を支持するのに必須栄養素が十分でないことが示されている。脳の成長が、発達中の他の組織の普通でない要求により損なわれる虞がある。
子ブタは、乳児腸管の発達および疾病についての選択のモデルとなっている。健康な若いブタにおける原理証明研究により、腸の健康のバイオマーカーと考えられている微生物(ビフィドバクテリウム科(Bifidobacteriaceae)および乳酸菌科(Lactobacillaceae))を富化する一方、疾患と関連する微生物を減らすことによって、ヒト乳によく似ているリゾチームリッチ乳の消費が、糞便の微生物叢組成を有益に調節することが実証された。E.A.Maga et al.,2012参照。微生物叢のシフトは、腸管の消化機能および免疫保護機能の双方の向上を示す、腸構造および遺伝子発現の双方の変化を伴った。挙げた当該変化は、腸管表面積を増大させ(絨毛はより長くなり、かつ粘膜固有層はより薄くなった)、これは、吸収機能の増大、抗炎症遺伝子(TGF-β)の発現の増大、および代謝物質の循環の好ましい変化を暗示している。C.A.Cooper et al.,2011;D.R.Brundige et al.,2010参照。ラクトフェリンリッチ乳は、微生物集団に及ぼす効果がより適度であった(データ未発表)が、腸管表面積の増大および炎症の減少の促進に及ぼす効果はより大きかった。C.A.Cooper et al.,Consumption of transgenic cows’ milk containing human lactoferrin results in beneficial changes in the gastrointestinal tract and systemic health of young pigs,22 TRANSGENIC RESEARCH 571(2012)(その全体が参照によって本明細書に組み込まれる)参照。
本研究は、母乳から離乳させた場合に食事のシフトが急な子ブタ、および食事のシフトがより緩やかなヒトの双方において起こることが知られている典型的な腸内バランス異常のバックグラウンドでの、乳児期中の腸のレベルでのエネルギー代謝の支持におけるニコチンアミドリボシド(NR、I)の役割を理解する必要に取り組む。S.A.Frese et al.,Diet shapes the gut microbiome of pigs during nursing and weaning,3 MICROBIOME 28(2015);J.E.Koenig et al.,Succession of microbial consortia in the developing infant gut microbiome,108 PROCEEDINGS NAT’L ACAD.SCI.4578(2011)(これらの文献はそれぞれ、その全体が参照によって本明細書に組み込まれる)参照。当該動物モデルを用いて、ヒト乳中のオリゴサッカライドと乳児微生物叢との関係を明らかにした。ビフィドバクテリウム・インファンティス(B.infantis)が占める微生物叢を乳児期中に確立できないと、慢性炎症性の状態が生じることが示された。ニコチンアミドリボシド(NR、I)の消費が、微生物発酵を通してのエネルギーの生成を支持して、結腸細胞用のエネルギーソースを提供し、腸管プロセスの適切な活気付けを増強し、かつ神経発生を維持すると予想された。これらは全て、最終的に、寿命の全体を通じて、健康を促進し、かつ疾患リスクを低くする。具体的な目的は:(1)成長および発達、例えば体重増加、成長、飼料効率比、糞便の稠度、および活性レベルの重要なパラメータに及ぼす、ニコチンアミドリボシド(NR、I)補給の効果を特徴付けること;(2)中間体、および腸内のエネルギー代謝の機能を、例えば、腸管内の代謝の測定として血液および糞便中の代謝物質を分析することによって、そしてニコチンアミドリボシド(NR、I)補給に起因する差異を評価することによって特徴付けることであった。
動物
16頭(n=16)のヨークシャー/ハンプシャー品種間交雑種の子ブタを、University of California,Davis Swine Teaching and Research Centerから得た。授乳14日目(「LD」)に受け取った。到着前、供給者が、1~3日齢の間、University of California,Davis Swine Facilityにて慣例の鉄および抗生物質(ブタ用Excede)の投与によって、子ブタを処理した。鉄および抗生物質の第2の用量を与えることは、必須にならない限り、University of California,Davis Swine Facilityの慣例でない。
離乳(ベースライン)時に、そしてニコチンアミドリボシド(NR、I)補充の1週および2週後に、全ての動物を秤量した。糞便スコアおよび活性スコアを、以下で一覧にするスケールを用いて、毎日記録した。体重および糞便スコアを、二因子反復測定ANOVA(混合モデルANOVA)を用いて、p値<0.05を有意と考えて分析した。用いた糞便の稠度スケールは、以下の通りである:4=標準(固体);3=軟便(半固体);2=軽度の下痢(半液体);1=重度の下痢(液体)。用いた活性レベルスケールは、以下の通りである:4=警戒、注意深い(移動、摂食、飲水、澄んだ眼);3=警戒、あまり活発でない(存在に応じて移動するが、遠くには行かない、摂食および飲水、澄んだ眼);2=やや無気力、疲れている(騒ぐが起立しない、食物および水に対する若干の関心、震えの発作、若干どんよりした、腫れた眼);1=非常に無気力(起立する気がない、食物および水に無関心、持続的な震え、どんよりした、腫れた眼)。
血液を、1、8、および14日目に、頸静脈穿刺により各ブタから採取した。各例において、血液を、ニコチンアミドリボシド(NR、I)の投薬前に採取した。1日目に、雌ブタを、サンプル採取前におおよそ3時間、子ブタから取り出した。8および14日目に、サンプル採取の12時間前に、動物の檻から餌を取り去った。血液を採取して、CBC分析用にパープルトップバキュテーナに、そして血液化学分析用にレッドトップバキュテーナに入れた。CBC分析を、Sysmex XT-iV Vet Hematology Autoanalyzer(Sysmex America Inc.,Lincolnshire,Illinois)を用いて、West Sacramento,CaliforniaのIDEXX Laboratoriesにて実行した。レッドトップチューブをスピンして、血清および凍結血清を収集した。凍結したアリコート(500μL)を、Cobas 6000 C501 Clinical Chemistry Analyzer(Roche Diagnostics,Indianapolis,IN)を用いるUniversity of California,Davis Veterinary medicine Teaching Hospital Clinical Diagnostic Laboratory for Blood Chemistry Analysisに提出した。残りの凍結血清のアリコートは、血清代謝物質分析に用いることとする。CBC化学パラメータおよび血液化学パラメータを、反復測定(混合モデルANOVA)を考慮する2因子ANOVA(処置および時間)を用いて分析した。p値<0.05を有意と考えた。6週齢のブタ(Cooper et al.,5 J.ANIM.SCI.BIOTECHNOL.5(2014)(その全体が参照によって本明細書に組み込まれる))および一般のブタ(University of California,Davis Veterinary Testing Laboratoryによって供給される)の双方についての基準範囲を、表2に示す。
フレッシュな糞便サンプルを、各ブタから1日目(ベースライン)、8日目(1週目)、そして14日目(2週目)に採取して、凍結した。フレッシュな排泄サンプルを得ることができなかった場合は、直腸をスワブで拭った。各ブタ由来の合計100mgの糞便を、酢酸、プロピオン酸、イソ酪酸、酪酸、イソ吉草酸、および吉草酸について知られている標準と共に、ガスクロマトグラフィを用いて、SCFA分析に用いた。サンプルを、25%メタリン酸で抽出して、各抽出物を、PeakSimple Chromatography Data Systemを備えたGCで3通りランさせた。一元配置ANoVAを、または値がガウス分布しなければノンパラメトリックKruskal-Wallis検定を用いて、データを分析した。データの保存分析を以下で表3に示し(糞便サンプルを得るためにスワブで拭わなければならなかった動物)、そしてスワブサンプルを含むデータの分析を表8に示す。傾向は類似するが、p値は類似しない。
Claims (3)
- 高レベルの少なくとも1つのニコチニルリボシド化合物を哺乳類に送達し、乳児用粉ミルク組成物の1週間の投与後に、糞便の短鎖脂肪酸レベルを統計的に有意に増大させるための乳児用粉ミルク組成物であって、前記組成物が:
(i)ニコチンアミドリボシド(NR、I)、ニコチン酸リボシド(NAR、II)、還元ニコチンアミドリボシド(NRH、IV)、還元ニコチン酸リボシド(NARH、V)、ニコチンアミドリボシドトリアセテート(NRTA、VI)、ニコチン酸リボシドトリアセテート(NARTA、VII)、還元ニコチンアミドリボシドトリアセテート(NRH-TA、VIII)、および還元ニコチン酸リボシドトリアセテート(NARH-TA、IX)からなる群から選択される少なくとも1つのニコチニルリボシド化合物であって、前記少なくとも1つのニコチニルリボシド化合物の量は、前記乳児用粉ミルク組成物の100キロカロリー当たり約1μgから約10,000μgである少なくとも1つのニコチニルリボシド化合物;および
(ii)不活性希釈剤、同化可能な食用のキャリア、または賦形剤
を含むことを特徴とする乳児用粉ミルク組成物。 - 請求項1に記載の乳児用粉ミルク組成物において、前記乳児用粉ミルク組成物は、ビタミンB1(チアミン、XI)、ビタミンB2(リボフラビン、XII)、ビタミンB3(ナイアシン、X)、およびビタミンB6(ピリドキシン、XIII)からなる群から選択される少なくとも1つのビタミンをさらに含むことを特徴とする乳児用粉ミルク組成物。
- 請求項1に記載の乳児用粉ミルク組成物において、前記乳児用粉ミルク組成物は、ホエーおよびカゼインからなる群から選択される少なくとも1つのタンパク質をさらに含むことを特徴とする乳児用粉ミルク組成物。
