Description
(1)材料選択:新鮮動物の腱又は靭帯を収集する。
(2)事前処理:ブロードスペクトラム、高効率、低毒の滅菌剤で予備的滅菌処理を行ってから、余分の夾雑物及び要らない部分を切り外す。
(3)脂肪除去:有機溶剤で基材中の脂肪を抽出する。
(4)抗原除去:活性試料を使って、基材蛋白質中の特異性活性グループ、例えば−OH、−NH2、−SHなどを閉鎖し、水素との結合力の強い試料を利用して基材蛋白質分子の螺旋チェーン中の特殊水素結合を置換して、特異性構造を変える。
(5)固定:固定剤を使って基材中の蛋白質分子を架橋固定する。
一つの優れた方法として、基材の表面にカップリング剤を通じて増殖因子が附着できる特定のポリペプチド又はGAG活性成分のカップリングによって活性表面層を形成させる。

を使っても良い。こちらでのR=CnH2n+1-で、n=0−10である。また、低重合エポキシ化物質、例えばポリプロピレンオキシドも使うことができる。
(1)材料選択:専門の担当者により規範化管理が行われている屠殺場から新鮮な動物の腱又は靭帯を収集し、なるべく汚染物に接触しないようにする。
(2)事前処理:ベンザルコニウムブロマイド、アジ化ナトリウム、クロルヘキシジンなどのブロードスペクトラムの高効率消毒剤を使って、予備的滅菌処理を行ってから、余分の夾雑物及び要らない部分を切って除去する。
(3)脂肪除去:有機溶剤、例えばクロロフォルム、アセトン、酢酸エステル、エーテル、無水アルコール又はこれらの混合物で基材1中の脂肪を抽出する。
(4)抗原除去:小分子の有機酸無水物、塩化アシル、酸アミド、シングルエポキシ化物質などの活性試料を使って、基材蛋白質中の特異性活性グループ、例えば−OH、−NH2、−SHなどを閉鎖し、水素との結合力の強い試料を利用して基材蛋白質分子の螺旋チェーン中の特殊水素結合を置換する。
(5)固定:ダブルエポキシ化物質
を固定剤として、基材1中の蛋白質分子を架橋固定するが、その中のn=0-10である。
(6)表面修飾:基材1の表面において、アジピン酸ジニトリル、無水二酸、又はダブルエポキシ化物質、若しくはその他−NH2、−OH、−COOHなどと縮合反応が発生できる二官能基試料などのカップリング剤を通じて、特定のポリペプチド又はGAG類活性成分をベース1の表面にカップリングすることによって、活性表面層2を形成する。
Claims (4)
- I 材料選択ステップ:新鮮動物の腱又は靭帯を収集して基材(1)とする、
II 事前処理ステップ:ブロードスペクトラム、高効率、低毒の滅菌剤を使って、基材(1)に対して予備的滅菌処理を行ってから、余分の夾雑物及び要らない部分を切り外す、
III 脂肪除去ステップ:有機溶剤で基材(1)中の脂肪を抽出する、
IV 抗原除去ステップ:活性試料を使って、基材(1)中の蛋白質中の−OH、−NH2、−SHの特定の活性基を閉鎖し、水素との結合力の強い試料を利用して基材(1)中の蛋白質分子の螺旋チェーン中の特異構造の原因となる水素結合を置換する、前記活性試料は、小分子の有機酸無水物、塩化アシル、酸アミド、シングルエポキシ化物質で、水素との結合力の強い試料はグアニジン類化合物である、
V 固定ステップ:固定剤を使って基材(1)中の蛋白質分子を架橋反応により固定する、前記固定剤は、蛋白質分子と架橋反応が発生し易いエポキシ化物質、アジピン酸ジニトリル、ヘキサメチレンジイソシアネート、又は、カルボジイミドの試料である、
VI.表面修飾ステップ:
前記基材(1)の表面にはカップリング剤を通じて、増殖因子が附着できる特定のポリペプチド又はグリコサミノグリカン類活性成分をカップリング剤によりカップリングすることによって、活性表面層(2)を形成する、前記特定のポリペプチド中の一種は、16のリジン(K16)、グリシン(G)、アルギニン(R)、アスパラギン酸(D)、セリン(S)、プロリン(P)及びシステン(C)のみから成るポリペプチドであり、前記グリコサミノグリカン類活性成分は、ヒアルロン酸や、コンドロイシン硫酸塩、デルマタン硫酸塩、ヘパリン、へパリン/へパラン硫酸(HS)、又はケラチン硫酸塩であり、前記カップリング剤は、アジピン酸ジニトリル、無水二酸、又はダブルエポキシ化物質、若しくは、−NH2、−OH、−COOHと縮合反応が発生できる二官能基試料である、
を含む生物型人工靭帯の調製方法。 - 前記II 事前処理ステップで使用されるブロードスペクトラム滅菌剤は、ベンザルコニウムブロマイド、アジ化ナトリウム、クロルヘキシジンを選択できることを特徴とする請求項1に記載されている生物型人工靭帯の調製方法。
- 前記III 脂肪除去ステップで使用される有機溶剤は、クロロフォルム、アセトン、酢酸エステル、エーテル、無水アルコール又はこれらの混合物を使用できることを特徴とする請求項1に記載されている生物型人工靭帯の調製方法。
