Abstract
Description
a)前記膜構成物質を水中に分散させる工程、
b)その分散液を混合し、均質化する工程、
c)その混合物をコーティングし、乾燥させる工程、
d)膜のスプールを切断し、包装する工程
を利用することを特徴とする
Kollicoat IR(登録商標)を70%の量の精製水に攪拌下で加えた。Kollicoat IR(登録商標)が溶解するまで攪拌を続けた。精製水に数多くの気泡が生じるため、真空下で溶解してもよいし、その溶液を静置してもよい(その粘度は非常に低いため、それは自然に脱気する)。その溶液にアジテーション下、攪拌下でTween 80を組み込み、その後、香味料(甘草濃縮抽出物およびハッカ精油)および甘味料(アセスルファムカリウム)を続けた。これらの粉末が完全に溶解するまで攪拌を続けた。ドンペリドンを、混合物中にそれが分散するまで攪拌下で加えた後、残りの水(30%)を加えた。凝集体が形成しないように、攪拌下でそのドンペリドン懸濁液にGelcarin 379(登録商標)を組み込んだ。次いで、その混合物のアリコートをポリエステル担体にコーティングし、Lab Dryer Coater Mathisタイプの装置を用いて乾燥させた。コーティングした表面を、手動プレスを用いて6cm2のユニットに切断した後、密閉小袋により手作業で包装した。
パイロットサイズのバッチを作り出すために、成分をタンクに次の順序:半分より多い量の水、Kollicoat IR(登録商標)、甘味料および香味料(甘草濃縮抽出物およびハッカ精油)、Tween 80およびドンペリドン、で加えた。このようにして得られた前混合物を、ダイスに連続通過させることによって均質化した後、メインタンクに再び加えた。その前混合物に残りの成分(Gelcarin 379(登録商標)および残りの量の水)を加え、全体を攪拌した。最終混合物を、ダイスに3回連続通過させることによって均質化した後、コーティング工程を進める前に、低窒素圧下で静置して、良好に脱気させた。その混合物を、コーティングステーション、連続する3つの乾燥領域およびラミネーションステーションからなるパイロットコーティングラインにポンプを用いて移した。コーティング速度および幅は、それぞれ、0.85m/分および17cmであった。第一に、熱の影響下での活性物質の分解を避けるために、そして第二に、膜中の水の残量をその機械的特徴に適合させるために(乾燥しすぎた膜はあまりに脆性でありすぎ、取り扱いには適さない)、温度勾配を設けた。熟成時間の後に、膜のスプールを切断し、専用装置を用いて包装した。
実施例1の方法に従って、Kollicoat IR(登録商標)とジェランガムまたはキサンタンガムの均質水性ポリマー混合物から他の速崩壊性膜を調製した。これらの結果を表3にまとめている。
本発明の膜には様々な活性物質を組み込むことができた。上記実施例1の操作モードに従って得られた膜の組成を表4に示している。
上記方法に従って、PVA−PEGコポリマー(Kollicoat IR(登録商標))とΙ−カラギーナン(Gelcarine 379(登録商標))の均質水性ポリマー混合物から特に満足のいく結果を与えた別の速崩壊性膜を調製した。
Claims (16)
- 少なくとも1種の活性物質を含有する水溶性担体を含んでなる、活性物質の経口投与のための速崩壊性単層膜であって、ポリビニルアルコールとポリエチレングリコールのコポリマー(PVA−PEG)からなる親水性膜形成剤、活性物質および親水性ゲル化剤のポリマー混合物を含んでなる、膜。
- 前記親水性ゲル化剤が、カラギーナン(好ましくはΙ−カラギーナン)、ジェランガムおよびキサンタンガムからなる群から選択されるものである、請求項1に記載の膜。
- 前記ポリマー混合物が追加の可塑剤を含まないものである、請求項1または2に記載の膜。
- 親水性ゲル化剤がカラギーナンファミリーに属する場合に、親水性ゲル化剤に対する膜形成剤の重量比率が1〜10、好ましくは3〜8である、請求項2に記載の膜。
- ポリマー混合物中の膜形成剤の重量が、総重量の10%〜30%、好ましくは13%〜20%であり、ポリマー混合物中の親水性ゲル化剤の重量が、総重量の1%〜8%、好ましくは2%〜6%である、請求項4に記載の膜。
- 前記ゲル化剤がキサンタンガムである場合に、親水性ゲル化剤に対する膜形成剤の重量比率が、10〜200、好ましくは25〜100である、請求項2に記載の膜。
- ポリマー混合物中のキサンタンガムの重量が、総重量の0.1%〜0.5%であり、好ましくは総重量の0.2%〜0.45%である、請求項6に記載の膜。
- 前記ゲル化剤がジェランガムである場合に、親水性ゲル化剤に対する膜形成剤の重量比率が、100〜400、好ましくは150〜300である、請求項2に記載の膜。
- ポリマー混合物中の親水性ゲル化剤の重量が、総重量の0.01%〜0.5%であり、好ましくは総重量の0.02%〜0.2%である、請求項8に記載の膜。
- 前記活性物質が、ドンペリドン、メキタジン、コデイン、塩酸ロペラミド、グルコン酸クロルヘキシジンとテトラカインとの組合せ、ニコチン、オキシブチニンおよびセチリジンからなる群から選択されるものであり、好ましくはドンペリドンである、請求項1〜9のいずれか一項に記載の膜。
- 1種以上の増量剤および/または1種以上の界面活性剤をさらに含む、請求項1〜10のいずれか一項に記載の膜。
- 界面活性剤としてポリソルベート80を含む、請求項1〜11のいずれか一項に記載の膜。
- 請求項1〜12のいずれか一項に記載の膜を製造するための方法であって、次の連続的工程:
a)膜構成物質を水中に分散させる工程、
b)その分散液を混合し、均質化する工程、
c)その混合物をコーティングし、乾燥させる工程
を行うことを特徴とする、方法。 - 膜構成物質の水中への分散が、半分より多い量の水に、次の順序:膜形成剤、界面活性剤、活性物質、残りの水、その後に親水性ゲル化剤、で加えることによって得られる、請求項13に記載の方法。
- 請求項12または13に記載の方法であって、均質水性ポリマー混合物を得るために、水の存在下で、周囲温度において粘度が1〜250mPa.sのポリビニルアルコールとポリエチレングリコールのコポリマー(PVA−PEG)からなる親水性膜形成剤、活性物質および親水性ゲル化剤を混合することによって、外部熱を加えることなく前記膜が得られ、従来のコーティング・乾燥技術を用いた前記膜の形成前に、該均質水性ポリマー混合物に、周囲温度での粘度250mPa.s〜15000mPa.s、好ましくは1000mPa.s〜10000mPa.s、さらに好ましくは1500mPa.s〜9000mPa.sを与えるように膜形成剤とゲル化剤との比率が決定されることを特徴とする、方法。
- 医薬品としての、請求項1〜12のいずれか一項に記載の膜。
