Abstract
Description
本発明はこれらの抗体またはそれらの機能的断片を、癌の予防的処置および/または治療的処置のための医薬品として利用することを目的とする。また、本発明は、たとえばその他の抗体および/もしくは抗癌剤と結合しているこのような抗体、または毒素と複合しているこのような抗体を含んでいる、製品および/または組成物と、ある種の癌の予防および/または治療を目的としたそれらの利用方法とを含んでいる。
1/タンパク質分解酵素(メタロプロテイナーゼなど)による、ラミニン、コラーゲン、またはフィブロネクチンのような構造タンパク質で構成される基底膜および細胞外マトリクスの分解を必要とする、原発腫瘍細胞による支持組織への浸潤、
2/組織を通過して血流に入る、腫瘍細胞の移動、
3/血管壁への接着と器官内での定着、
4/血管からの脱出(浸潤の新たな過程)と新しい環境への適応(増殖および血管新生)である。
−配列がそれぞれ配列番号7、8、および9である、CDR−H1、CDR−H2、およびCDR−H3という3つの重鎖のCDRと、
−配列がそれぞれ配列番号11、12、および13である、CDR−L1、CDR−L2、およびCDR−L3という3つの軽鎖のCDR
を含むことで定義される。
−配列がそれぞれ配列番号15、16、および17である、CDR−H1、CDR−H2、およびCDR−H3という3つの重鎖のCDRと、
−配列がそれぞれ配列番号19、20、および21である、CDR−L1、CDR−L2、およびCDR−L3という3つの軽鎖のCDR
を含むことで定義される。
TS151r抗体の生成
TS151r抗体を生成するため、107個のHeLa細胞を用い、BALB/cマウスを腹腔内経路によって免疫した。3回の免疫と最後の追加免疫注射の後、マウスの脾臓細胞を、Kohler and Milsteinによって説明されている従来的な技術によってP3X63AG8骨髄腫細胞と融合させた(5×107個の脾臓細胞/3×107個の骨髄腫細胞)。融合により生じたハイブリドーマの上澄みを、フローサイトメトリーによって、HeLa細胞を認識する能力についてスクリーニングし、次に、洗浄剤Brij97の存在下で調製したHeLa細胞の溶解物からCD151を免疫沈降させる能力と、CD9を共免疫沈降させる能力とに関してスクリーニングした。TS151r抗体はこれらのさまざまな特性を有することが明らかとなった。
TS151抗体を生成するために、107個のJurkat細胞と、107個のHEL細胞を用い、腹腔内経路でBALB/cマウスを免疫した(2回の免疫)。Jurkat細胞とHEL細胞の溶解物から得たADAM10タンパク質を含んだタンパク質複合体を用いた最後の追加免疫注射の後、脾臓細胞を、Kohler and Milsteinによって説明されている従来的な技術によってP3X63AG8骨髄腫細胞と融合させた(5×107個の脾臓細胞/3×107個の骨髄腫細胞)。融合によって生じたハイブリドーマの上澄みを、まず、フローサイトメトリーによって、Jurkat細胞とHEL細胞を認識する能力についてスクリーニングした。次に、洗浄剤Brij97の存在下で調製した細胞溶解物からCD151を免疫沈降させる能力と、その他のテトラスパニンを共免疫沈降させる能力に関してTS151抗体を選別した。
材料と方法
CD151タンパク質の発現を確認した後(データは示していない)、ATCCに由来するA549細胞を、F12K培地、10mMのグルタミン、10%FCSにおいて通常通りに培養した。これらの細胞を移植の2日前に分裂させることで、該細胞が成長の指数増殖期となるようにした。移植に関しては、7週齢の免疫不全マウスに麻酔をかけた後、1×106個のA549細胞を胸膜内経路で投与した。原発腫瘍は急速に成長し、4日で、縦隔、肺、および隔壁を含む、注射部位に隣接する構造に侵入した。病気をより良く模倣するために、治療の開始は細胞移植の7日後にようやく初めて、腹腔内経路によって開始した。500μg/マウスの攻撃投与量を注射した後、精製したTS151抗体を週に2回、5週間にわたって、250μg/マウスの投与量で投与した。先に行った実験によって、IgG1アイソタイプの対照を投与しても動物の生存率にいかなる影響もないことが示されているため、PBSを投与したマウスの群を対照として入れた。
図4は、計算されたT/C%が140%である、TS151抗体の抗腫瘍活性を示している。
材料と方法
実施したプロトコルは上記実施例2と同じである。
得られたデータは、TS151抗体が50−6抗体の示したものを明らかに上回る抗腫瘍活性を有することを明らかに示しており、該50−6抗体は、118のT/C%、すなわち125%という閾値よりも低い値しか示さなかった(データは示していない)。
前立腺癌または肺癌を有する患者に由来するヒト組織のサンプルにおいて、CD151タンパク質の発現を免疫組織化学によって研究した。これらの患者については、腫瘍に隣接する正常組織のスライドも利用可能であり、したがって、腫瘍組織対正常組織の発現レベルを較正するために該スライドも含めた。
前立腺の「組織アレイ」において免疫組織化学によって得られた結果を考慮して、PC3腫瘍の異種移植片に対する抗CD151抗体の評価を計画した。PC3株は、ATCCに由来する、アンドロゲンに依存しない前立腺株であり、F12K培地+10%FCS+L−グルタミンで培養した。評価のために、5×106個のPC3細胞をスイスヌードマウスの右脇腹に移植した。移植の5日後、腫瘍の容積に基づいて動物を無作為抽出し、3つの比較群に配分した。選択した移植動物群における腫瘍の容積は、治療0日目で41mm3から47mm3であった(π/6×長さ×幅×厚みという式で計算した容積)。それから、動物に、試験すべき精製した抗体またはPBSを投与した。抗体の投与量および注射の頻度は、2mgの攻撃投与量/投与の抗体で、1mgの維持投与量/投与を週に2回である。
ウェスタンブロットによってTS151抗体とTS151r抗体の特異性を評価した。ヒトとマウスの、肺、膵臓、および結腸の組織の溶解物(Biochain、10μgの全タンパク質)、ならびに、段階的に増加する量のHT−29細胞溶解物(10、20、および50μgの全タンパク質)を、4〜12%アクリルアミドゲル(BioRad)の上に置いた。電気泳動の後(非還元条件)、タンパク質をニトロセルロース膜に移した。次に、転写膜を、精製したTS151抗体とTS151r抗体と共にインキュベートし、そして、ペルオキシダーゼに結合したマウスのウサギ抗Igポリクローナル抗体(GE Healthcare)を用いてインキュベートした後に、ECLタイプの視覚化を行った。
CD151が結合することのできるインテグリンα3β1とインテグリンα6β4のリガンドであるラミニン5に対する腫瘍細胞の接着実験を、96ウェルプレートで行った。ラミニン5(Chemicon、1μg/mlで200μl)を37℃で1時間固定した後、ウェルに2mg/ml(200μl、37℃で1時間)のBSAを満たした。懸濁させたA549細胞を5−クロロメチルフルオレセインジアセテート(CMFDA、Invitrogen)で標識し、抗体の存在下(100μl)または不存在下で、ウェルあたり100000個の細胞(100μl)の割合で加えた。15分、30分、または60分にわたって37℃でインキュベートした後、接着していない細胞を除去した。ルミノメーター(Mithras、Berthold)を用いた化学発光の読み取りの後、接着した細胞のパーセンテージをCMFDAで標識した細胞のスケールを用いて判定した。抗CD151抗体TS151および抗インテグリンα3抗体P1B5、抗α6抗体NKI−Go3、および抗β4抗体ASC−3(Chemicon)を、20μg/mlの最終濃度で評価した。大腸菌の膜タンパク質に対する9G4抗体をアイソタイプの対照として用いた。

- 前記少なくとも一つの抗CD151抗体またはその機能的断片が、CD151タンパク質のアミノ酸40〜57(配列番号6)と113〜221(配列番号4)にそれぞれ対応する細胞外ループ1(EC1)および/または2(EC2)、好ましくはEC2に含まれるエピトープと結合する能力を有することを特徴とする、請求項1〜請求項7のいずれか一つに記載の利用方法。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片が、CD151タンパク質の194位、195位、および196位(QRD194〜196)にそれぞれあるアミノ酸であるグルタミン、アルギニン、およびアスパラギン酸を少なくとも含むEC2領域のエピトープと結合する能力を有することを特徴とする、請求項8に記載の利用方法。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片がモノクローナル抗体で構成されることを特徴とする、請求項1〜請求項9のいずれか一つに記載の利用方法。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片がTS151抗体で構成されることを特徴とする、請求項1〜請求項10のいずれか一つに記載の利用方法。
- 前記抗体が、
−配列がそれぞれ配列番号7、8、および9である、CDR−H1、CDR−H2、およびCDR−H3という3つの重鎖のCDRと、
−配列がそれぞれ配列番号11、12、および13である、CDR−L1、CDR−L2、およびCDR−L3という3つの軽鎖のCDR
を含むことを特徴とする、請求項11に記載の利用方法。 - 前記抗体が配列番号10の配列を含んだ重鎖と配列番号14の配列を含んだ軽鎖を含むことを特徴とする、請求項12に記載の利用方法。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片がTS151r抗体で構成されることを特徴とする、請求項1〜請求項10のいずれか一つに記載の利用方法。
- 前記抗体が、
−配列がそれぞれ配列番号15、16、および17である、CDR−H1、CDR−H2、およびCDR−H3という3つの重鎖のCDRと、
−配列がそれぞれ配列番号19、20、および21である、CDR−L1、CDR−L2、およびCDR−L3という3つの軽鎖のCDR
を含むことを特徴とする、請求項14に記載の利用方法。 - 前記抗体が配列番号18の配列を含んだ重鎖と配列番号22の配列を含んだ軽鎖を含むことを特徴とする、請求項15に記載の利用方法。
- 前記癌が、結腸癌、肺癌、前立腺癌、または膵臓癌で構成されることを特徴とする、請求項1〜請求項16のいずれか一つに記載の利用方法。
- 薬学的に許容可能な少なくとも一つの担体をさらに含む製薬組成物の調製を目的とする、請求項1〜請求項17のいずれか一つに記載の利用方法。
- CD151タンパク質と結合する能力を有する少なくとも一つの抗CD151抗体またはその機能的断片を活性成分として含むことを特徴とする、癌の治療を目的とした組成物。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片が、原発腫瘍の成長を阻害する能力を有する抗体であることを特徴とする、請求項19に記載の組成物。
- 前記少なくとも一つの抗CD151抗体またはその機能的断片が、TS151抗体またはTS151r抗体から選択されるモノクローナル抗体であることを特徴とする、請求項19または請求項20に記載の組成物。
- TS151抗体およびTS151r抗体またはその機能的断片の組み合わせを含むことを特徴とする、請求項19〜請求項21のいずれか一つに記載の組成物。
- 同時利用、別々の利用、または時間間隔をあけた利用を目的とした組み合わせ製品として、さらに少なくとも一つの細胞傷害剤/細胞増殖抑制剤および/または細胞毒および/または放射性元素および/またはモノクローナル抗体を含むことを特徴とする、請求項19〜請求項22のいずれか一つに記載の組成物。
- 癌の治療を目的とした医薬品を調製するための、請求項19〜請求項23のいずれか一つに記載の組成物の利用方法。
- 原発腫瘍の治療を目的とした医薬品を調製するための、請求項19〜請求項24のいずれか一つに記載の組成物の利用方法。
